Book excerpt: Inhalation of airborne particulate (PM) matter has been epidemiologically correlated to numerous cardiopulmonary morbidities and mortalities, particularly in susceptible populations such as asthmatics and children. To date, PM has been classified by size by the scientific and regulatory communities. PM10 is the designation for particles less than 10 micrometers in diameter and PM2.5 for particles less than 2.5 micrometers. Particles smaller than 100 nanometers, or nanoparticles, are of special interest. Nanoparticles have unique properties that distinguish them from lager particles, including their behavior in air and their interaction with biological organisms. Nanoparticles can form from natural sources or as the byproduct of human activity. Additionally, specific engineered nanoparticle are being manufactured for use in electronics, textiles, personal care products and medicine. The increasing use of nanoparticles has raised concerns over possible health effects. Age differences in nanoparticle deposited dose, key to associating exposure to physiologic effect, is poorly understood. Silver nanoparticles are widely used for their antimicrobial activity. Exposure to aerosolized silver nanoparticles in the workplace has been documented. While many characteristics of silver nanoparticles are known, their retention and clearance is poorly understood. It is believed that particle surface coating and size can affect the clearance of silver nanoparticles from the lung. First, we examined the spatial and temporal pattern of clearance for particles of two sizes, 20 and 110nm, and two surface coatings, citrate and polyvinylpyrrolidone (PVP), four particle types in total, by instilling the nanoparticles into the lungs of rats. We then studied two sizes of selected one of the coating types, citrate, and performed an inhalation study to study how the mode of delivery influences silver particle clearance. Finally, to examine age differences in nanoparticle deposition, we used a europium doped gadolinium oxide nanoparticle as a surrogate for ambient nanoparticles and exposed by inhalation 1, 3 and 12 week old rats. We found the instilled PVP coated particles of both sizes had significantly greater clearance by 21 days compared to the citrate coated particles. We also found the terminal bronchiolar alveolar duct junction region of the lung the location with greatest retention. In rats exposed to aerosolized citrate coated silver nanoparticles, we saw a greater silver burden in the macrophages of the 20nm exposed animals. We also saw that approximately of the 30% initial silver dose remained in the lungs at 56 days for both particles. Finally, when we examine the age difference of nanoparticle deposition in three ages of rats, the 3 week old rats received the greatest dose of particles and that 1 week old rats had significantly lower deposition compared to older animals.
