The stability and assembly of functionalized nanoparticles are still challenges for practical applications. Polymers are excellent stabilizing agents which can be used in nanoparticles synthesis. This dissertation describes polymer mediated nanoparticles which synthesized by both chemical and photo reduction techniques. The synthesis methods and assembly of polymer mediated nanoparticles were systematically studied. Two kinds of polymers were used in this research: PAMAM dendrimer and hyperbranched polyethylenimine. PAMAM dendrimer was used as the host for the synthesis of FePt nanoparticles with narrow size distribution by chemical reduction. Magnetic and structural properties of the as-made and annealed samples were studied. The as-made FePt nanoparticles have the chemically disordered fcc structure and can be transformed into the chemically ordered fct structure after annealing. But the annealing process caused both the size and size distribution to increase. To prevent size increase during annealing, dendrimer mediated alloys were encapsulated in silica microspheres. Their magnetic properties can be manipulated by synthesis conditions. Polyethylenimine is a kind of hyperbranched polymer, which was used to encapsulate nanoparticles. With polyethylenimine, Pt and Co3O4 nanoparticles were synthesized by photochemical processes. Their formation mechanisms were discussed. The chemical and electrochemical catalytic activity of polyethylenimine mediated Pt nanoparticles were studied. The results indicate that polyethylenimine is a good capping and reducing agent, and a carrier which improve samples stability and allows patterning. Ordered discrete magnetic nanoparticle arrays were also fabricated. Polyethylenimine was used as a stabilizer to synthesize magnetite nanoparticles. Polyethylenimine coating of magnetite nanoparticles can effectively prevent the aggregation of the nanoparticles. By combining the capillary filling technique with a magnetic field, polyethylenimine mediated magnetite nanoparticles form ordered assembly, in which the resolution of the pattern is doubled. The formation process of ordered structures was systematically investigated. This method is technologically feasible and scalable to make ordered magnetic nanoparticles over a large area. The result of this dissertation demonstrates that polymer stabilizers play an essential role in the synthesis of nanoparticles as well as their assembly.

Book excerpt: How can the two dimensional crystallization of colloids be used to form highly ordered colloidal monolayers on solid substrates? What application does this have in generating arrays of nanostructures? These questions are addressed in Nicolas Vogel's thesis. Vogel describes a simple preparation method for the formation of uniform colloidal crystals over large areas, which he refines to yield more complex binary and non-close-packed arrangements. These monolayers can be applied to a process termed colloidal lithography which is used to prepare high quality metallic nanostructures with tailored properties defined to suit a variety of applications. Moreover, the author describes a method used to create metallic nanodot arrays with a resolution unprecedented for colloidal lithography methods. The author also outlines methodology to embed nanoparticle arrays into the substrate, which is developed and used to design robust, re-usable biosensor platforms and nanoscale patterns of biomimetic lipid bilayer membranes. The research in this thesis has led to a large number of publications in internationally renowned journals.

Book excerpt: The development of practical strategies for the assembly of semiconductor and metal colloid nanoparticles into ordered architectures is an area of considerable current interest, since it offers an opportunity for exploiting the optical and electronic properties of these colloids for device development. Prior research has explored creating such organized nanoparticle assemblies by Langmuir-Blodgett techniques or controlled solvent evaporation on suitable substrates. These approaches suffer from several limitations, however, most notably the generation of relatively simple structures and the lack of structural tailorability, preventing full exploitation of these materials. More recently, directed assembly using chemisorption of streptavidin-biotin or thiol-derivatized gold nanoparticles onto substrates has been described. Alternative approaches to achieving two-dimensional confinement of nanoparticles that do not involve substrate-supported materials, but rather organize the nanoparticles into mesoscopically-ordered soft condensed matter, may offer the advantage of enhanced processability and may permit construction of nanocomposite structures based on functional nanoparticles embedded in a processable, polymer-based matrix. This work describes the development of an alternative strategy for constructing 2-D arrays of functional metal and semiconductor nanoparticles. The approach involves directing the organization of nanocrystals into a processable (i.e., by externally applied magnetic and electric fields) polymer-grafted lipid-based complex fluid. By altering the surface chemistry of the nanoparticles, they can be selectively placed into defined regions encapsulating matrix.

Book excerpt: This dissertation demonstrates chemically-driven self-assembly techniques to produce assemblies of closely-spaced metal nanoparticles from colloidal nanoparticle solution in order to engineer enhanced optical fields. Planar nanoparticle assemblies provide a platform for a multitude of applications and material architectures. With nanoscale inter-particle spacing, metallic nanoparticles enable increased efficiency of photovoltaic devices due to light focusing and enhancement of electromagnetic fields useful for optical sensing of molecules due to coupling of the plasmon resonance in nanoparticle gaps. For molecular sensors, development of self-assembled two-dimensional assemblies of closely-spaced nanoparticles is useful for producing surface plasmon resonance sensors and surface-enhanced Raman spectroscopy (SERS) based sensing. Using chemical self-assembly, monodisperse, colloidal gold nanoparticles were attached on self-organized polymer templates in order to pattern assemblies of nanoparticle clusters with sub-10 nanometer inter-particle spacing. First citrate-stabilized Au nanoparticles were functionalized with thioctic acid ligands in solution. Then poly(methyl methacrylate) domains in phase-separated poly(styrene-block-methyl methacrylate) (PS-b-PMMA) thin films were chemically modified with surface amine functional groups. Au nanoparticles were preferentially attached to the functionalized PMMA surface domains using cross-linking chemistry. This method allows for versatility of size, shape, and composition. In this dissertation, we demonstrated attachment of 5, 10, and 20 nm Au and 20 nm Ag nanoparticles. PS-b-PMMA thin films also exhibit versatility of domain size and morphology by varying polymer molecular weights. The nanoparticle diameter to PMMA domain size ratio influenced the cluster size. As the ratio decreased, larger clusters were observed on PMMA domains with increased frequency. SERS measurement of nanoparticle assemblies showed uniform signal enhancement across samples with enhancements as high as 109. Reusability of SERS substrates with simple analyte removal between measurements was also observed. Two different deposition methods, diffusion and electrophoresis, were compared in terms of Au nanoparticle coverage, and inter-particle spacing. We found electrophoresis provided comparable nanoparticle coverage to diffusion deposition with 83% reduced processing time, 23% increased cluster formation, and 40% reduced inter-particle spacing. As optical field strength increases with decreased inter-particle spacing, fast processing times and high coverage of closely-spaced clustered nanoparticles would benefit for efficient and economical manufacturing of chemically assembled sensors and light-trapping surfaces on solar cells.

That is the reason why, although mainly dealing with synthetic drug carriers, the first chapter is related to preparation and physicochemical properties of nanoparticles constructed from natural polymers. This is followed by two chapters involving the application of polymeric drug carriers in cancer therapy. The second chapter extensively describes in vitro and in vivo behaviour of purely synthetic nanoparticles. As ub the case of liposomes, a problem can be that some particles are preferentially taken up by the reticuloendothelial system. Magnetic drug carriers provide a partial answer to this problem since their tropism can be managed by an external magnet. The third chapter is entirely devoted to this aspect. A brief but original application of nanoparticles is given in the fourth chapter by reviewing their use in ocular therapy. Finally, two general applications of polymeric microspheres are discussed in chapters 5 and 6.
