Book excerpt: It is appro pi ate to the contents of this book to recall a few highlights in the history of plant cytology from its inception over three centuries ago. Robert Hooke in 1663 presented his observations of what he called « cells" in cork and other plant parts and beautifully illustrated and described these in his classic « Micrographia" published two years later. More detailed exploration of the cell and its contents awaited almost two centuries for Robert Brown's discovery of the nucleus in 1831. Discoveries of other cell organelles followed, particularly in the latter part of the 19th and early part of this century. As is frequently noted each of these achievements was preceeded by advances in the resolution of the microscope. Now history repeats and recent developments in electron microscopy have given the biologist the opportunity to study cell morphology in far greater detail than at any time previously. Indeed, the resolution of the electron microscope is several hundredfold better than that available in the finest light microscopes. These advances in instrumentation plus improvements in the techniques of specimen preparation have made possible the ex amination of plant cells of almost any type. It is the resulting wealth of new information now accessible to the botanical cytologist that has prompted this publication. In this book we have brought together electron micrographs represent ing a number of cell types from higher plants.

Book excerpt: Since the publication of my monograph "Die submikroskopische Struktur des Cytoplasm as" [Protoplasmatologia III A12 (1955)], science has increased our knowledge on this cell constituent in an amazing way. At that time, cytoplasm was still considered as an amorphous colloid with an amicroscopic structure capable of sol/gel transformations. Meanwhile, within the past 18 years, ultrastructure research and molecular biology have shown that it consists of structured organelles, thereby revolutionizing our concept of the cytoplasm. There is an ontogeny of these organelles which deals with their appearance, development, growth and eventual disappearance. Although the results of relevant studies are less spectacular at this time than those concerning nucleic acids, they will become more and more important in the near future, because they touch the basic question of how molecular morphogenesis is realized. Therefore, an attempt is made to portray the actual state of these problems from a biological point of view which involves a comparison of the established organelles. Available in PDF, EPUB and Kindle. Book excerpt: In the first part (Part A) of this volume on transport, there was an emphasis on the processes occurring at the membranes bounding the cells. It was convenient to distinguish active and passive processes of transport across the membranes, and to recognize that certain transport processes may be regulated by internal factors in the cells such as cytoplasmic pH, concentrations of ions, of malate or of sugar in the vacuoles, or the hydrostatic pressure. Cells in tissues and organs show the same kinds of properties as individual cells, but in addition there can be cell to cell transport related to the organization of the tissue. Firstly cells within a tissue are separated from the external solutions by a diffusion path comprising parts of the cell walls and intercellular spaces; more generally this extra-cytoplasmic part of the tissue has been called the apoplasm. A similar term is "free space". Secondly, the anatomy of cells in tissues seems to allow some facilitated, local transport between cells in a symplasm. Entry into the symplast and subsequent transport in a symplasmic continuum seems to be privileged, in that ions may not have to mix with the bulk of the cytoplasm and can pass from cell to cell in particular cytoplasmic structures, plasmodesmata. In Chara plants, this kind of transport is found operating across the multi-cellular nodes as the main means of transport between the long internodal cells.
