Book excerpt: The project report describes a two year experiment to control wetland drainage to the San Joaquin River of California from the San Luis National Wildlife Refuge using a decision support system for real-time water quality management. This system required the installation and operation of one inlet and three drainage flow and water quality monitoring stations which allowed a simple mass balance model to be developed of the seasonally managed wetlands in the study area. Remote sensing methods were developed to document long-term trends in wetland moist soil vegetation and soil salinity in response to management options such as delaying the initiation of seasonal wetland drainage. These environmental management tools provide wetland managers with some of the tools necessary to improve salinity conditions in the San Joaquin River and improve compliance with State mandated salinity objectives without inflicting long-term harm on the wild fowl habitat resource.

Book excerpt: This report documents work at the U.S. Geological Sur- vey (USGS) National Water Quality Laboratory (NWQL) to validate enzymatic reduction, colorimetric determinative meth- ods for nitrate + nitrite in filtered water by automated discrete analysis. In these standard- and low-level methods (USGS I-2547-11 and I-2548-11), nitrate is reduced to nitrite with nontoxic, soluble nitrate reductase rather than toxic, granular, copperized cadmium used in the longstanding USGS auto- mated continuous-flow analyzer methods I-2545-90 (NWQL laboratory code 1975) and I-2546-91 (NWQL laboratory code 1979). Colorimetric reagents used to determine resulting nitrite in aforementioned enzymatic- and cadmium-reduction meth- ods are identical. The enzyme used in these discrete analyzer methods, designated AtNaR2 by its manufacturer, is produced by recombinant expression of the nitrate reductase gene from wall cress (Arabidopsis thaliana) in the yeast Pichia pastoris. Unlike other commercially available nitrate reductases we evaluated, AtNaR2 maintains high activity at 37°C and is not inhibited by high-phenolic-content humic acids at reaction temperatures in the range of 20°C to 37°C. These previously unrecognized AtNaR2 characteristics are essential for success- ful performance of discrete analyzer nitrate + nitrite assays (henceforth, DA-AtNaR2) described here.

In its Ecology Program, one mode of initiating research is to en courage the development of new ideas through advisory workshops. The NSF is specifically directed to strengthen our nation's research potential. In addition, stimulating new approaches to research will continue to be prominent in the coming years as federal attention is given to increasing the innovativeness and competitiveness of the U. S. in science and engineering. A decision to initiate a workshop does not arise de novo in the Ecology Program. Rather, it emerges from panel discussions, conversations with in vestigators at meetings or on the phone, and from discussions between pro gram officers in the Division of Biotic Systems and Resources. This workshop was developed to provide advice to the NSF and the lim nological community. Some NSF perceptions on future funding for ecolog ical research on lake communities are presented here. Researchers often mentioned a paucity of innovative lake ecology at the community level. This perception was accompanied by a certain frustration since lakes probably have the best empirical data base of any natural environment and should continue to lead in the development of ecological concepts. Members of NSF advisory panels sometimes expressed similar concerns during consid eration of proposals for lake research.
