Book excerpt: Excerpt from The Journal of Biological Chemistry, 1917, Vol. 30 The accompanying table gives the result of the analyses for calcium of fifty-four samples of human blood, determinations of the total non-coagulable nitrogen also being done on the pathological cases. It is hoped that these figures may help to establish a base line for further study of the metabolism of this metal. The chief point brought out seems to be that the calcium varies but little in a great majority of subjects. Even in cases of advanced uremia, where the total nitrogen was extremely high, the calcium was not increased. On the other hand, the hemophiliac and the case of purpura hemorrhagica were not lower than what we may regard as normal. The average for the males, with the exception of No. 36, was 6.1 mg. of calcium per 100 cc. of blood; while that for females was 7.1, a difference sufficiently great to stimulate further investigation. In No. 36 there seems to be no reason for the extremely low figure shown. Duplicate determinations were of course performed and there is no doubt that this represents the correct calcium content. Unfortunately, as the patient soon left the hospital, a second specimen could not be obtained. No. 8, on the other hand, was taken from a healthy young woman whose history gives us no clue to the explanation of the high result - 9.6 mg. per 100 cc. Determination of calcium was carried out according to the new method of the author,1 while the total nitrogens were done by the direct Nesslerization process of Folin and Denis. The former figures probably represent hydrolyzed hippuric acid since van de Velde and Stokvis (31) showed that if urine were collected directly from the ureters no free benzoic acid was present. In order to make use of the excretion of hippuric acid during the 6 hour period as a measure of the rate of synthesis of glycine, it must be shown that the excretion of hippuric acid depends upon the amount of glycine available and not upon the rate of absorption of benzoate from the intestine, nor upon the rate of formation of hippuric acid, nor upon the rate of elimination of hippuric acid by the kidneys. Book excerpt: Excerpt from The Journal of Biological Chemistry, 1918, Vol. 33 The finding of occult blood in stools is of great importance in the diagnosis of ulcer and cancer of the stomach. However, the usual chemical methods are so unreliable in their results that they are at present discredited by many surgeons and internists. A review of the literature of this subject shows not only that the number of reagents proposed is very large, but that there also exists a considerable number of procedures for the use of these reagents. The two most widely used are gum guaiac and benzidine. The phenolphthalein reagent is difficult to prepare and moreover is entirely too sensitive for ordinary use. In a previous paper we have shown how the benzidine reagent may give erroneous results unless great care is exercised in cleaning all the apparatus as well as in the use of reagents of definite concentrations. In the routine examination of stools we have found it difficult to get good controls with benzidine; moreover, it is too sensitive. An alcoholic solution of gum guaiac possesses the following disadvantages: (1) Not being of fixed composition or purity, a solution of definite concentration cannot be readily prepared. (2) Gums from different sources vary greatly in their sensitiveness as well as in the stability of the blue compound formed. (3) For the best results, solutions must be freshly prepared.
