Book Quantitative Aspects of Chemical Pharmacology

Download or read book Quantitative Aspects of Chemical Pharmacology written by R.B. Barlow and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 257 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book gathers together chemical ideas which are important for understanding how drugs act and how new drugs may be developed. Students meet some of these ideas in courses in chemistry, biochemistry and pharmacology but in my experience they find it difficult to put together information often acquired in different years and from different departments; they need it set out in a book. I believe also that it helps if they can see how these ideas may be applied to numerical problems so I have included examples (with answers), many of which are taken from research work. These should therefore provide a chance for the student both to test his or her own understanding and to see the sort of results which form the bricks of which any experimental science is made. In all the problems the arithmetic takes less than ten minutes with a small calculator which has exponential functions and logarithms.

Book Chemical Structure Biological Activity Relationships Quantitative Approaches

Download or read book Chemical Structure Biological Activity Relationships Quantitative Approaches written by F. Darvas and published by Elsevier. This book was released on 2013-10-22 with total page 401 pages. Available in PDF, EPUB and Kindle. Book excerpt: Chemical Structure-Biological Activity Relationships: Quantitative Approaches, Volume III, documents the proceedings of the 3rd Congress of the Hungarian Pharmacological Society held in Budapest, 1979. This volume focuses on the methodological aspects of QSAR. It also aims to inform the reader about the QSAR research conducted in East-European countries. This volume contains 38 presentations organized into six sections. Several lecturers deal with ""real prediction"" cases, i.e. activity estimation prior to the synthesis of the compounds. A particularly abundant section is devoted to the question of how the receptor models can be built up by means of QSAR calculations. Other sections present mathematical models and algorithms which could be applied to improve further the effectiveness of QSAR calculations. As another unique feature, a separate section treats the quantitative aspects of peptide structure-activity relationships—a field seemingly backward despite its evident importance. Hydrophobicity and its influence on biological potency along with the relationship of steric properties and biological activity are also discussed.

The book begins with a review of chemical principles as they apply to drug molecules, focusing mainly on those for commonly prescribed drugs. The authors use drug structures to illustrate the chemical concepts learned in general and organic chemistry courses. They cover the dynamics of receptors in mediating the pharmacological effects of drugs. They clarify theories, drawn from the scientific literature, which explain drug-receptor interactions and the quantitative relationship between drug binding and its effects at the cellular level. The authors’ extensive use of drug structures for teaching chemical and molecular pharmacology principles, and their emphasis on the relevance of these principles in future professional life makes this book unique. It provides the framework for better understanding of advanced pharmacology and therapeutics topics. Blending medicinal chemistry and pharmacodynamics aspects, this textbook clearly elucidates the essential concepts that form the cornerstone for further work in pharmacology.

Book The Emerging Discipline of Quantitative Systems Pharmacology

Download or read book The Emerging Discipline of Quantitative Systems Pharmacology written by Tarek A. Leil and published by Frontiers Media SA. This book was released on 2015-09-07 with total page 99 pages. Available in PDF, EPUB and Kindle. Book excerpt: In 2011, the National Institutes of Health (NIH), in collaboration with leaders from the pharmaceutical industry and the academic community, published a white paper describing the emerging discipline of Quantitative Systems Pharmacology (QSP), and recommended the establishment of NIH-supported interdisciplinary research and training programs for QSP. QSP is still in its infancy, but has tremendous potential to change the way we approach biomedical research. QSP is really the integration of two disciplines that have been increasingly useful in biomedical research; “Systems Biology” and “Quantitative Pharmacology”. Systems Biology is the field of biomedical research that seeks to understand the relationships between genes and biologically active molecules to develop qualitative models of these systems; and Quantitative Pharmacology is the field of biomedical research that seeks to use computer aided modeling and simulation to increase our understanding of the pharmacokinetics (PK) and pharmacodynamics (PD) of drugs, and to aid in the design of pre-clinical and clinical experiments. The purpose of QSP modeling is to develop quantitative computer models of biological systems and disease processes, and the effects of drug PK and PD on those systems. QSP models allow testing of numerous potential experiments “in-silico” to eliminate those associated with a low probability of success, avoiding the potential costs of evaluating all of those failed experiments in the real world. At the same time, QSP models allow us to develop our understanding of the interaction between drugs and biological systems in a more systematic and rigorous manner. As the need to be more cost-efficient in the use of research funding increases, biomedical researchers will be required to gain the maximum insight from each experiment that is conducted. This need is even more acute in the pharmaceutical industry, where there is tremendous competition to develop innovative therapies in a highly regulated environment, combined with very high research and development (R&D) costs for bringing new drugs to market (~$1.3 billion/drug). Analogous modeling & simulation approaches have been successfully integrated into other disciplines to improve the fundamental understanding of the science and to improve the efficiency of R&D (e.g., physics, engineering, economics, etc.). The biomedical research community has been slow to integrate computer aided modeling & simulation for many reasons: including the perception that biology and pharmacology are “too complex” and “too variable” to be modeled with mathematical equations; a lack of adequate graduate training programs; and the lack of support from government agencies that fund biomedical research. However, there is an active community of researchers in the pharmaceutical industry, the academic community, and government agencies that develop QSP and quantitative systems biology models and apply them both to better characterize and predict drug pharmacology and disease processes; as well as to improve efficiency and productivity in pharmaceutical R&D.

Book Quantitative Analysis of Cellular Drug Transport Disposition and Delivery

Download or read book Quantitative Analysis of Cellular Drug Transport Disposition and Delivery written by Gus R. Rosania and published by . This book was released on 2021 with total page 453 pages. Available in PDF, EPUB and Kindle. Book excerpt: This thorough book explores some of the most important methods and concepts affecting the quantitative analysis of the transport, targeting, and disposition of chemicals within cells, which in turn impact the macroscopic pharmacokinetics of chemical agents in the whole organism. The first half of the volume focuses on small organic molecules with drug-like characteristics, while the second half delves into the cellular pharmacokinetics of biologics and other macromolecules, including peptide therapeutics, cyclotides, antibodies, as well as nanoparticles, thus creating a comprehensive treatise that approaches cellular pharmacokinetics from the different perspectives of pharmaceutical scientists, chemical biologists, medicinal chemists, and protein engineers dealing with very different chemical agents spanning a wide range of sizes, physicochemical properties, and targeting mechanisms. Written for the Methods in Pharmacology and Toxicology series, chapters provide the kind of key detail and expert implementation advice that leads to excellent results in the lab. Synthetic biologists, biophysicists, and bioengineers are amongst the long list of scientists who could benefit from reading this book or from using it as a textbook. Authoritative and practical, Quantitative Analysis of Cellular Drug Transport, Disposition, and Delivery builds on a long history of drug development and the adding of quantitative methods at the cellular scale in order to inspire new approaches to drug development that are better able to take advantage of phenomena such as soluble-to-insoluble phase transitions or bispecific targeting, which could ultimately be exploited for the development of more effective drug delivery systems and therapeutic agents.

Book Quantum Pharmacology

    Book Details:
  • Author : W. G. Richards
  • Publisher : Elsevier
  • Release : 2013-10-22
  • ISBN : 1483163679
  • Pages : 288 pages

Download or read book Quantum Pharmacology written by W. G. Richards and published by Elsevier. This book was released on 2013-10-22 with total page 288 pages. Available in PDF, EPUB and Kindle. Book excerpt: Quantum Pharmacology, Second Edition discusses molecular pharmacology, molecular quantum mechanics, and the applications of theory to experiment. The book reviews molecular and quantitative pharmacology, as well as acetylcholines, catecholamines, and histamines. The text also discusses the central nervous system in relation to monoamines, acetylcholines, amino acids, and peptides. The book explains wave functions and orbitals, secular equations, matrix elements, and self-consistent molecular orbitals. One way to improve wave functions as a result of a self-consistent field or as an approximate calculation, is to permit different configurations to interact. Linear mixing will lead to improved wave functions. The investigator can compute wave functions and energies of molecules for any set of nuclear coordinates. Quantum chemical calculations treat conformation as similar with geometry; calculations are performed for a series of positions in one part of a molecule relative to another. The investigator can then compare the energies found in each position. The book points out that calculations of conformational internal energy surfaces for flexible pharmacological molecules treating the molecule or ion as isolated in space fit in with observed results of crystallographic experiments. The text is suitable for pharmacologists, and scientists involved in molecular pharmacology, medicinal chemistry, and quantum chemistry.
