Book excerpt: The chemistry of cyclometalated gold(III) has been relatively slow to emerge, but recent advances in catalyst and material design have driven a renewed interest. Reported here are synthetic, structural, and optical studies of cyclometalated gold(III) aryl complexes. These new complexes show ligand-centered luminescence that is perturbed by the heavy-atom effect of gold. Traditionally, metal-carbon bond formation relied on organometallic complexes of electropositive elements, including lithium and magnesium. The easily reducible nature of gold(III) often interferes with these conventional reagents, thus resulting in the use of toxic metals like mercury or thallium. Reported here is the use of palladium-catalyzed Suzuki-Miyaura cross-coupling for the attachment of aryl ligands to cyclometalated gold(III) chloride complexes. The resulting di- and mono-arylated species were luminescent. Several complexes have been crystallographically characterized. Luminescent derivatives of a previously published gold(III) chloro precursor, initially reported for catalytic purposes, were prepared through boron transmetalation reactions. Complexes bearing both aryl and cyanide ligands were synthesized. These luminescent materials are diamagnetic and were be characterized by multinuclear NMR, absorption and emission spectroscopies (including time-resolved emission), and by X-ray diffraction crystallography. The synthesized gold(I) complexes were designed to display reverse saturable absorption and be a two photon acceptor. The BTF (BTF = 2-(9,9’-diethyl-9H-fluoren-2-yl)-benzothiazole) ligand was chosen as it has been previously demonstrated to have favorable photophysical effects on platinum(II) and ruthenium(II) complexes for applications in photodynamic therapy and optical power limiting (OPL). Gold(I) complexes with the BTF ligand are sought for potential OPL applications. Synthesized complexes are luminescent and were characterized by multinuclear NMR, absorption and emission spectroscopies, photoluminescent quantum yields, time-resolved emission. Two complexes were crystallographically characterized.

In modern times, gold has been used in the treatment of a number of different human diseases including rheumatoid arthritis, cancer, and viral infections. This dissertation will focus on the development of gold complexes for the purpose of selective inhibition of HIV NCp7, a 55 amino acid zinc finger protein with two Cys3His zinc binding domains. NCp7 is involved in a number of viral life cycle processes including activation of reverse transcription, integration, DNA recognition, RNA packaging, and formation of the viral envelope. The diversity in the roles of NCp7 across the viral life cycle make it a highly attractive target for chemical intervention. Ejection of the tetrahedrally coordinated zinc atoms, modification or deletion of the zinc coordinating amino acids, or modification or deletion of the "essential" tryptophan residue can result in the loss of viral infectivity. This is due to the inability of NCp7 to recognize its "natural" substrate -- polynucleic acids. Previous studies have investigated the differences in the manner in which platinum(II), gold(I), and gold(III) complexes interact with NCp7. The platinum(II) complex [Pt(dien)(9-EtGua)]2 interacts with the C-terminal zinc finger of NCp7 in a non-covalent manner, through a [pi]-[pi] stacking interaction between the platinated 9-EtGua and the "essential" tryptophan residue. The isostructural and isoelectronic complex [Au(dien)(9-EtGua)]3 ejects the tetrahedrally coordinated zinc atom and replaces it with a gold atom, forming "gold fingers". This result is consistent with the interactions of the gold(I) complex [(PPh3)Au(9-EtGua)]+. In order to complete the series of isoelectronic and isostructural platinum(II), palladium(II), and gold(III) complexes with N-heterocyclic ligands and diethylenetriamine chelates, the complexes [Au(dien)(1-MeCyt)]3+ and [Au(N-Medien)(1-MeCyt)]3+ were synthesized. These complexes were found to dimerize the C-terminal zinc finger once the central zinc atom is ejected. This is likely the result of a charge transfer from the 1-methylcytosine ligand to the tryptophan residue, and is a product that was not seen as a result of interaction with the previously published 4-dimethylaminopyridine and 9-ethylguanine analogs. The 1-methylcytosine complexes also stabilize the gold(III) oxidation state and associate with N-acetyltryptophan in a manner consistent with the previously studied gold(III) analogs. Finally, in order to address concerns arising from the inner filter effect, a proof of concept study using 1H-NMR spectroscopy was utilized to show that the complex [Au(dien)(1-MeCyt)]3+ likely has a lower association constant with N-acetyltryptophan than the value determined by fluorescence quenching. The impact of the incorporation of additional steric hindrance on the gold(III) chelate was studied using the di-(2-picolyl)amine ligand. The gold(III) chlorides incorporating this ligand and the centrally methylated analog were found to eject zinc from the C-terminal zinc finger of NCp7, and the electronegativity differences between the gold(III) and platinum(II) metal centers were highlighted. The attempts to incorporate an N-heterocyclic ligand into these complexes were unsuccessful due to the steric and electronic demands of the chelate. The use of an organometallic chelating ligand led to the investigation of the ability of gold(III) complexes to catalyze the arylation of zinc-coordinating cysteine residues. The complex [AuCl2(dampa)], which had been formerly investigated as a chemotherapeutic agent due to its structural similarities to cisplatin, was found to arylate N-acetylcysteine, glutathione, and NCp7. The arylation was not found to be dependent on the cis- chloride ligand, as blocking that site with the ligand triphenylphosphine did not prevent the arylation of NCp7. The X-Ray crystal structure of the complex [AuCl(dampa)(PPh3)](PF6) was also solved. Using the advancement of the knowledge of how the electronic and structural properties of gold(III) complexes described herein impact interactions with NCp7, it is possible that a coordination complex that is a selective inhibitor of NCp7 may eventually be developed.

Various aspects of devices based on polyparaphenylene vinylenes are discussed in chapters by N.C. Greenham and R.H. Friend and by H. Chayet, V. Savvateeyv, D. Davidov and R. Neumann. Chapters by S. Tasch, W. Graupner, and G. Leising and by Y. Z. Wang, D. Gebler, and A. J. Epstein describe OLEDs based on poly(paraphenylene) and poly(pyridine), respectively. This book is intended to reminisce this event, to take inventory, and to point out new lines of development in this field. Divided in 6 sections and 22 chapters, the book provides an up-to-date account on topics such as - the chemistry and biochemistry of cisplatin, - the clinical status of Pt anticancer drugs, - the impact of cisplatin on inorganic and coordination chemistry, - new developments in drug design, testing and delivery. It also includes a chapter describing the historical development of the discovery of cisplatin. The ultimate question - How does cisplatin kill a cell? - is yet to be answered, but there are now new links suggesting how Pt binding to DNA may trigger a cascade of cellular reactions that eventually result in apoptosis. p53 and a series of damage recognition proteins of the HMG-domain family appear to be involved. The book addresses the problem of mutagenicity of Pt drugs and raises the question of the possible relevance of the minor DNA adducts, e.g. of interstrand cross-links, and the possible use of trans-(NH3)2Pt(II)-modified oligonucleotides in antisense and antigene strategies. Our present understanding of reactions of cisplatin with DNA is based upon numerous model studies (from isolated model nucleobases to short DNA fragments) and application of a large body of spectroscopic and other physico-chemical techniques. Thanks to these efforts there is presently no other metal ion whose reactions with nucleic acids are better understood than Pt. In a series of chapters, basic studies on the interactions of Pt electrophiles with nucleobases, oligonucleotides, DNA, amino acids, peptides and proteins are reported, which use, among others, sophisticated NMR techniques or X-ray crystallography, to get remarkable understanding of details on such reactions. Reactivity of cisplatin, once bound to DNA and formerly believed to be inert enough to stay, is an emerging phenomenon. It has (not yet) widely been studied but is potentially extremely important. Medicinal bioinorganic chemistry - the role of metal compounds in medicine - has received an enormous boost from cisplatin, and so has bioinorganic chemistry as a whole. There is hardly a better example than cisplatin to demonstrate what bioinorganic chemistry is all about: The marriage between classic inorganic (coordination) chemistry and the other life sciences - medicine, pharmacy, biology, biochemistry. Cisplatin has left its mark also on areas that are generally considered largely inorganic. The subject of mixed-valance Pt compounds is an example: From the sleeping beauty it made its way to the headlines of scientific journals, thanks to a class of novel Pt antitumor agents, the so-called "platinum pyrimidine blues". In the aftermath diplatinum (III) compounds were recognized and studies in large numbers, and now an organometalic chemistry of these diplatinum (III) species is beginning to emerge. The final section of the book is concerned with new developments such as novel di- and trinuclear Pt(II) drugs with DNA binding properties different from those of cisplatin, with orally active Pt(IV) drugs which are presently in clinical studies, and with attempts to modify combinatorial chemistry in such a way that it may become applicable to fast screening of Pt antitumor drugs. The potential of including computational methods in solving questions of Pt-DNA interactions is critically dealt with in the concluding chapter.
