Book excerpt: The flexible filamentous plant viruses are responsible for more than half of all agricultural loss worldwide. Potexvirus is one of the two most important flexible filamentous plant viruses. Bamboo mosaic virus (BaMV), a single-stranded positive-sense RNA virus, is a member of the Potexvirus genus of Alphaflexiviridae. It can infect at least 12 species of bamboo, causing a huge economic impact on the bamboo industry in Taiwan. The study of BaMV did not start extensively until the completion of the full-length sequencing of genomic RNA of BaMV and generation of the BaMV infectious cDNA clone in the early 1990s. Since then, BaMV has been extensively studied at the molecular, cellular and ecological level, covering both basic and applied researches, by a group of researchers in Taiwan. In this eBook, the content comprises 6 reviews and 4 articles. Seven of them are involved in the infection of BaMV covering viral RNA replication, viral RNA trafficking, and the host factors. Two of them are related to the vector transmission and the ecology of BaMV. The last one is the application of using BaMV as a viral vector to produce vaccines in plants.

Whereas in animals the well-defined, protected cells of the germ line separate early, germ cells in plants differentiate from somatic cells only after many cycles of mitotic division. Therefore somatic mutations in plants can be transmitted via the germ cells to the progeny. There is thus a clear need for somatic tissues to maintain their genetic integrity in the face of environmental challenges, and two types of interactions have been shown to play important roles in the conservation as well as flexibility of plant genomes: homologous recombination of repeated sequences and silencing of multiplied genes. Sensitive methods have been developed that allow greater insights into the dynamics of the genome. This book summarizes current knowledge and working hypotheses about the frequencies and mechanisms of mitochondrial, plastid, nuclear and viral recombination and the inactivation of repeated genes in plants. Despite rapid developments in the field, it is often not possible to provide final answers. Thus, it is an additional task of this book to define the open questions and future challenges. The book is addressed to scientists working on plant biology and recombination, to newcomers in the field and to advanced biology students.

Between 1961 and 1966 my colleagues and I deciphered the genetic code in Escherichia coli and showed that the genetic code is the same in E. coli, Xenopus laevis, and guinea pig tissues. These results showed that the code has been c- served during evolution and strongly suggested that the code appeared very early during biological evolution, that all forms of life on earth descended from a c- mon ancestor, and thus that all forms of life on this planet are related to one another. The problem of biological time was solved by encoding information in DNA and retrieving the information for each new generation, for it is easier to make a new organism than it is to repair an aging, malfunctioning one. Subsequently, small modi?cations of the standard genetic code were found in certain organisms and in mitochondria. Mitochondrial DNA only encodes about 10–13 proteins, so some modi?cations of the genetic code are tolerated that pr- ably would be lethal if applied to the thousands of kinds of proteins encoded by genomic DNA.
