Book excerpt: From May 3-7,1997, the NATO Advanced Research Workshop on 'Biological Electron Transfer Chains' was organized in Tomar, Portugal. In the application for support the choice of the topic was justified as follows: "[Until recently efforts] have concentrated on the study of the structure and function of individual redox enzymes and proteins. Enough information is now available to make a start with the study of biological electron transfer (E1) at the next higher level of organization, that of the complete ET chain." The interest in the workshop was high: the majority of participants had registered before the workshop was formally announced, which illustrates the popularity of the topic within the biochemical and biophysical communities. The present volume contains a number of reports based on the lectures presented by the key speakers during the meeting. The workshop dealt with the following three themes: a) Electron transfer, which is the subject of Chapter 1. Enzymes involved in direct oxygen activation are oxidases and oxygenases. Multi-copper oxidases are an important class of oxidases reducing dioxygen in a four-electron reduction to water with concomitant one-electron oxidation of the reducing substrate. The progress in the characterization and understanding of the structure and function of these enzymes has advanced so tremendously over the last ten years that the publication of a book documenting these achievements has been overdue.Especially the recent discovery of a key role of the FET3 protein of Saccharomyces cerevisae, a multi-copper oxidase, in iron metabolism of this eukaryote has underpinned the function of the plasma multi-copper oxidase ceruloplasmin in vetebrate iron transport. Human diseases such as thalassemia, immunodeficiency, and leukemia represent defects in this process. Approaches to treat these disorders have required a basic understanding of the biology of blood cells. For instance, hemapoietic stem cell replacement or bone marrow transplantation has been used to ameliorate disease. This volume focuses on hematopoiesis at a cellular and molecular level, and establishes the basis for clinical manipulation of hematopoietic cells for therapeutic benefit. In Part I, the cellular characteristics of progenitors and stem cells are explored. Emphasis is placed on purification of stem cells and both in vitro and in vivo assays. The regulation of normal and leukemis stem cells is illustrated. An excellent discussion of potential use of these cells for gene therapy concludes this section. Hemapoiesis is easily studied during embryogenesis. Part II develops the concept of the waves of hemapoiesis during development. Comparative hematology is making a major comeback as a field in the 1990's. One hope is that general principles of hematopoiesis will be established by studying many models and systems. Part III delves into critical factors that regulate hematopoiesis, including both intracellular and extracellular signals. Part IV and V describe lineage programs for myeloid and lymphoid lineages. These chapters are meant to be illustrative of the different cell fates, but are not exhaustive. Part VI examines the genetics of hematopoisis, particularly in animal models. The hematopoietic system is in constant contact with stromal cells and endothelial cells during development and in the adult. Evidence suggests that endothelial cells and blood cells may arise from a common progenitor, the hemangioblast. Part VII and VIII discuss the stromal and endothelial cells with the emphasis on their interaction with hematopoietic cells.

Book excerpt: The interplay between host and pathogen is a complex co-evolutionary battle of surveillance and evasion. The pathogen continuously develops mechanisms to subvert the immune response in order to establish infection while the immune system responds with novel mechanisms of detection. Because the majority of Lyme disease pathology is due to an over-exuberant immune response, much research in Borrelia burgdorferi pathogenesis has been devoted to understanding the mammalian host response to the bacterium. Immunological studies continue to be an active area of research employing emerging techniques, such as intra-vital imaging. These studies have furthered our understanding of inflammatory processes during long-term infection and provided some surprising insights, such as the continued presence of bacterial products after clearance. The field of Lyme disease has long debated the etiology of long-term inflammation and recent studies in the murine host have shed light on relevant cell types and inflammatory mediators that participate in the pathology of Lyme arthritis. Live imaging and bioluminescent studies have allowed for a novel view of the bacterial life cycle, including the tick mid-gut, tick-to-mammal transmission and dissemination throughout a mouse. A number of tick and bacterial proteins have been shown to participate in the completion of the enzootic cycle. Novel mechanisms of gene regulation are continuously being identified. However, B. burgdorferi lacks many traditional virulence factors, such as toxins or specialized secretion systems. Many genes in the B. burgdorferi genome have no known homolog in other bacteria. Therefore, studies focusing on host-pathogen interactions have therefore been limited by an incomplete understanding of the repertoire of bacterial virulence factors. Questions such as how the pathogen causes disease, colonizes the tick and evades host immune-surveillance have been difficult to address. Genetic studies involving single gene deletions have identified a number of important bacterial proteins, but a large-scale genomics approach to identify virulence factors has not been attempted until recently. The generation of a site-directed mutagenesis library is an important step towards a detailed analysis of the B. burgdorferi genome and pathogenome. Using this library, high-throughput genomic studies, utilizing techniques such as massively parallel sequencing have been promising and could be used to identify novel virulence determinants of disease in the mammalian host or persistence in the tick vector. Continued research on this unique pathogen and its specific interaction with host and vector may have far reaching consequences and provide insights for diverse disciplines including ecology, infectious disease, and immunology. Here, several reviews will discuss the most recent advances and future studies to be undertaken in the field of B. burgdorferi biology.

The aim of this Research Topic is to highlight recent research on amphibian microbiomes that addresses relevant questions on the ecology of amphibian-microbe interactions. The publications gathered in this Research Topic have expanded our knowledge on the role of microbial symbionts of amphibians and have revealed novel insights that can direct the next set of research questions. We suggest that soon the field will move from the basic (and necessary) descriptions of microbial communities to more experimental approaches that include the use of omics methods and a variety of novel analytic and multivariate approaches. In addition to providing more insights into the microbial and disease ecology of amphibians, these studies may lead to effective ways to manipulate the microbiome to achieve protection from diseases, such as chytridiomycosis.
