The fluidity of these bilayers allows them to respond to different environmental cues by changing their local molecular composition as well as their shape and topology. On the nanometer scale, this multi-responsive behavior can be studied by molecular dynamics simulations, which provide both snapshots and movies of the bilayer conformations. The general conceptual framework for these simulations is provided by the theory of curvature elasticity. The latter theory also explains the behavior of giant vesicles as observed by optical microscopy on the micrometer scale. The present volume describes new insights as obtained from recent developments in analytical theory, computer simulations, and experimental approaches. The seven chapters of the volume are arranged in a bottom-up manner from smaller to larger scales. Book excerpt: Giant unilamellar vesicles (GUVs) can be described as a topologically closed membrane with a diameter of 10-50 [mu]m, compartmentalizing pico-liter quantities of the aqueous core from the surrounding bulk. One of the fundamental motif of the GUV study is its simplicity. Without any external constraint vesicle will exhibit spherical morphology as sphere configures the lowest bending energy in a topologically closed system. Albeit to sustain mere sphere morphology, GUV can be locked into various morphologies utilizing relatively low bending rigidities and the semi-permeable nature of the lipid membrane, under the presence of concentration gradient across the membrane. In the presence of an impermeable solute with concentration gradient across the membrane, GUV experiences osmotic potential as water endeavors to diffuse against concentration gradient of the solute through the membrane and the encapsulated volume of the GUV deviates as a result. Fluctuation in encapsulated volume insists deformation to the GUV and due to unique combination of elastic properties, which include large volume compressibility (∼109 to 1010 N/m2) and area expansion moduli (102 to 103 mN/m), and low bending rigidities (10−19 Nm)1−2, GUV undergoes bending-dominated shape deformation. Furthermore, the associated bending energy of typical lipid (k[subscript b] ~ 10-100 kT) is far less in scale compared to osmotic potential that the encapsulated volume will be secured under the deformation. Manipulating the surrounding concentration of GUV can ultimately control the encapsulated volume while the bounding surface area remains constant. Since the sphere encapsulates the highest volume with given bounding area, we focus in the reduction of the encapsulated volume. Depleting encapsulated volume forces GUV to move out of spherical shape and find a new equilibrium state within the reduced volume constrain enforced by the osmotic potential. By carefully designing, we were able to control the surrounding concentration temporally and precisely, seizing an opportunity to witness not only the morphology in the equilibrium states but also to monitor the pathway from the initial sphere morphology to the new equilibrium state. This dissertation describes dynamic analysis of shape deformation in lipid bilayer vesicles using a microfluidic system. It includes 1) design, preparation and characterization of microsystem not only for confining a vesicle but also for manipulating surrounding environment of GUV; 2) verification and validation of feasibility of microfluidic devices for a dynamic study of vesicle shape transformation through multiple sets of proof-of-concept experiments; and 3) generation of vesicle fission via well-controlled osmotic cycles.

Book excerpt: We investigate isothermal diffusion and growth of micron-scale liquid domains within membranes of free-floating giant unilamellar vesicles (GUVs) with diameters between 80μm and 250μm. Domains appear after a rapid temperature quench, when the membrane is cooled through a miscibility phase transition such that coexisting liquid phases form. In membranes quenched far from a miscibility critical point, circular domains nucleate and then progress within seconds to "late stage" coarsening in which domains grow via two mechanisms: (1) collision and coalescence of liquid domains, and (2) Ostwald ripening. Both mechanisms are expected to yield the same growth exponent, [alpha] = 1/3, where domain radius grows as time[superscript alpha]. We measure [alpha] = 0.28 ± 0.05, in excellent agreement. In membranes close to a miscibility critical point, the two liquid phases in the membrane are bi-continuous. A quench near the critical composition results in rapid changes in morphology of elongated domains. In this case, we measure [alpha] = 0.50 ± 0.16, consistent with theory and simulation.

Book excerpt: Dynamics of phospholipids in unilamellar vesicles (ULV) is of interest in biology, medical, and food sciences since these molecules are widely used as biocompatible agents and a mimic of cell membrane systems. We have investigated the nanoscopic dynamics of 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) phospholipid in ULV as a function of temperature using elastic and quasielastic neutron scattering (QENS). The dependence of the signal on the scattering momentum transfer, which is a critical advantage of neutron scattering techniques, allows the detailed analysis of the lipid motions that cannot be carried out by other means. In agreement with a differential scanning calorimetry measurement, a sharp rise in the elastic scattering intensity below ca. 296 K indicates a phase transition from the high-temperature fluid phase to the low-temperature solid gel phase. The microscopic lipid dynamics exhibits qualitative differences between the solid gel phase (in a measurement at 280 K) and the fluid phase (in a measurement at a physiological temperature of 310 K). The data analysis invariably shows the presence of two distinct motions: the whole lipid molecule motion within a monolayer, or lateral diffusion, and the relatively faster internal motion of the DMPC molecule. The lateral diffusion of the whole lipid molecule is found to be Fickian in character, whereas the internal lipid motions are of localized character, consistent with the structure of the vesicles. The lateral motion slows down by an order of magnitude in the solid gel phase, whereas for the internal motion not only the time scale, but also the character of the motion changes upon the phase transition. In the solid gel phase, the lipids are more ordered and undergo uniaxial rotational motion. However, in the fluid phase, the hydrogen atoms of the lipid tails undergo confined translation diffusion rather than uniaxial rotational diffusion. The localized translational diffusion of the hydrogen atoms of the lipid tails is a manifestation of the flexibility of the chains acquired in the fluid phase. Because of this flexibility, both the local diffusivity and the confinement volume for the hydrogen atoms increase linearly from near the lipid s polar head group to the end of its hydrophobic tail. Our results present a quantitative and detailed picture of the effect of the gel-fluid phase transition on the nanoscopic lipid dynamics in ULV. Lastly, the data analysis approach developed here has a potential for probing the dynamic response of lipids to the presence of additional cell membrane components.

Book excerpt: The present thesis introduces a novel sensitive technique based on TSM resonators that provides quantitative information about the dynamic properties of biological cells and artificial lipid systems. In order to support and complement results obtained by this method supplementary measurements based on ECIS technique were carried out. The first part (chapters 3 and 4) deals with artificial lipid systems. In chapter 3 ECIS measurements were used to monitor the adsorption of giant unilamellar vesicles as well as their thermal fluctuations. From dynamic Monte Carlo Simulations the rate constant of vesicle adsorption was determined. Furthermore, analysis of fluctuation measurements reveals Brownian motion reflecting membrane undulations of the adherent liposomes. In chapter 4 QCM-based fluctuation measurements were applied to quantify nanoscopically small deformations of giant unilamellar vesicles with an external electrical field applied simultaneously. The response of liposomes to an external voltage with shape changes was monitored as a function of cholesterol content and adhesion force. In the second part (chapters 5 - 8) attention was given to cell motility. It was shown for the first time, that QCM can be applied to monitor the dynamics of living adherent cells in real time. QCM turned out to be a highly sensitive tool to detect the vertical motility of adherent cells with a time resolution in the millisecond regime. The response of cells to environmental changes such as temperature or osmotic stress could be quantified. Furthermore, the impact of cytochalasin D (inhibits actin polymerization) and taxol (facilitate polymerization of microtubules) as well as nocodazole (depolymerizes microtubules) on the dynamic properties of cells was scrutinized. Each drug provoked a significant reduction of the monitored cell shape fluctuations as expected from their biochemical potential.
