Book excerpt: Concern is often expressed that our environment may include an increasingly large variety of mutagens, but the extent of the potential hazard they pose has yet to be fully evaluated. A variety of empirical procedures has been devised with which to estimate the mutagenic potency of suspect agents, and the relative merits of different tests are currently under debate. Although such tests are of great value, and are indeed indispensable, they are not, nevertheless, sufficient. In the long term, accurate estimation of hazard will also require a better understanding of the various mechanisms of mutagenesis, and in many instances these remain remarkably elusive. Our knowledge and appreciation of the problem has increased substantially over the last few years, but the precise way in which many mutagens cause mutations is not yet known. The aims of this conference were therefore two-fold. The first was to survey present information about mutagenic mechanisms, drawing together data from work with various experimental approaches and organisms, in order to discern the principles governing the action of different mutagens. The second was to examine the implications of such principles for the execution and evaluation of test procedures, and critically assess the research areas that need further attention in order to improve the interpretation of test results. Book excerpt: In the four years since the 1978 Symposium on the Application of Short-Term Bioassays in the Fractionation and Analysis of Complex Environmental Mixtures the use of short-term bioassays to evaluate potential health hazards of complex environmental mixtures has substantially increased. Increased research activity has been particularly noticeable in mobile source emissions, where initial observations on the mutagenic activity of diesel particulate extracts reported at the 1978 symposium stimulated the development of major research programs in government and industry. In the absence of appropriate reference materials, the U.S. Environmental Protection Agency initiated comparative genotoxicity studies to determine the relative mutagenic and carcinogenic activity and, ultimately, the potential human health risk due to exposure to various complex emission products. Among the materials investigated were those of known health risk, such as coke oven and roofing tar emissions and cigarette smoke condensates, and those of unknown hazard, such as exhaust from diesel-and gasoline-powered vehicles. Studies on diesel emission products proved useful in short term bioassay development, as the diesel exhaust extracts were genetically active with low cellular toxicity and could be obtained in relatively large quantities. Availability of such samples aided chemical characterization, and it was eventually determined that the nitro-polynuclear aromatic hydrocarbons were among the mutagenic components of diesel exhaust particulate.

Book excerpt: I have been privileged to witness and participate in the great growth of knowledge on chemical carcinogenesis and mutagenesis since 1939 when I entered graduate school in biochemistry at the University of Wisconsin Madison. I immediately started to work with the carcinogenic aminoazo dyes un der the direction of Professor CARL BAUMANN. In 1942 I joined a fellow graduate student, ELIZABETH CA VERT, in marriage and we soon commenced a joyous part nership in research on chemical carcinogenesis at the McArdle Laboratory for Cancer Research in the University of Wisconsin Medical School in Madison. This collaboration lasted 45 years. I am very grateful that this volume is dedi cated to the memory of Elizabeth. The important and varied topics that are reviewed here attest to the continued growth of the fields of chemical car cinogenesis and mutagenesis, including their recent and fruitful union with viral oncology. I feel very optimistic about the application of knowledge in these fields to the eventual solution of numerous problems, including the detection and estimation of the risks to humans of environmental chemical carcinogens and re lated factors.

The aim of the Course was to present different scientific ap proaches and technical advices in order to get dose-effect relation ships which are the basis for risk evaluation. The scientific back ground which is behind this approach was extensively discussed. Emphasis has been given to the use of human cells or human data in order to attempt to have a correct and realistic evaluation of the damage in humans. There are many criticisms on the use of animal data for human risk evaluation because of differences between species and between strains within the same species: differences in metabolism, activa tion processes and DNA repair ability makes uncertain the extrapola tion of animal data to humans. Also data obtained using specific strains or highly inbred strains in order to reduce the variance are not applicable due to the heterogeneity of the human population connected with individual responses. In this respect only the use of human cells enable us to detect the individual variability and to identify sensitive subpopu lations that would be at greater risk. Book excerpt: During the early 1930s, when I was a graduate student and later a post-doctoral researcher at the National Research Council for the University of Wisconsin at Madison, we had the opportunity to get acquainted with many graduate students from China who were sent to the University for training in modern basic sciences as well as social sciences. The University of Wisconsin continues to graduate a large number of Chinese students. Economic conditions in the 1930s were very precarious for the United States and other parts of the world. Many of us students grew closer together because we were living on similarly tight budgets. As a matter of fact, we subleased a part of our apartment in Madison to some Chinese graduate students. This was a very nice opportunity for us to learn about the scientific and cultural back ground of our Chinese friends. Many of them came from the interior of China and had had very little opportunity to become acquainted with people from a western culture. Living with these students was a very pleasant and educational experience which gave us a good pic ture of the cultural life and educational system of China at that time--an intimate picture that one normally would not see without travelling in that country.

Book excerpt: It is delightful but humbling to find my face at the start of these Proceedings--there are innumerable other faces which could equally weIl stand there, from among the band who have fore gathered at every gerontology conference since the subject was launched in its present form; but I deeply appreciate being there. Gerontology d. id not grow by accident. Its present standing is the fruit of careful planning, undertaken by European and American scientists back in the 1950's. In those days it was still a "fringe" science, and the conspirators had much the standing of the 1920's Interplanetary Society. The United States itself is the offspring of conspiracy, for when the results of conspiracy are beneficent, the conspirators become Founding Fathers. This has been the case with gerontology. The present meeting is especially gratifying because the papers have been recitals of normal, hard-science investigation. We had to get through the rigors of a long period of semantic argument and a long period of one-shot general theories before this kind of meeting, normal in all other research fields, could take place. It was also necesssary to breed in the menagerie a generation of excellent investigators aware of the theoretical background but unintimidated by it, who share our conviction that human aging is comprehensible and probably controllable, and who go into the laboratory to attack specifics.

Book excerpt: This volume is based on the proceedings of an Advanced Study Institute (ASI) sponsored by the North Atlantic Treaty Organization (NATO) held October 1984 in Corfu, Greece. The meeting received finan cial support from the United States Department of Energy and the United States National Cancer Institute. A plethora of recent developments in the molecular biology of DNA are leading to new ideas concerning how DNA alterations might be involved in the mechanism of radiation carcinogenesis. Evidence is accumulating that genetic sequences, known as oncogenes, are involved in the translation of DNA molecular alterations into phenotypic changes associated with malignant cells. For example, a chromosome break often occurs at or near the loca tion of a specific oncogene in Burkitt's lymphoma. Such breaks could represent initial lesions in a translocation process that activates the oncogene by inserting it at a new location, eg., near an active pro moter. Since breakage of the DNA is one of the principal ways that ion izing radiation affects mammalian cells, these new molecular ideas sug gest ways that radiation-induced DNA breaks might be involved as initial events in carcinogenesis. While the possible involvement of oncogenes in radiation carcino genesis is an exciting new development, a direct sequential connection between early molecular changes in DNA and later tumor development has yet to be established. Accordingly, there is a tremendous need for experimental studies of how DNA alterations might convert normal cells to cancer cells.
