However, all epithelial cells share some common characteristics. The cells possess a morphological polarity (an-apical and basolateral surface), and are interconnected by tight junctions. The epithelial cells also possess the capacity to transport select solutes across the monolayer. Transport systems localized on either the apical or basolateral surface are respon sible for this vectorial transport. Such characteristics of epithelial cells can be examined in the tissue culture situation. This volume discusses the use of cell culture techniques to study these fundamental properties of epithelial cells. Ma jor questions concerning epithelia which may be examined in culture are ad dressed. The approaches which are taken to answer these questions are described in detail with regards to kidney cell cultures. Similar investigations may be done with epithelial cell cultures derived from other tissues, following the kidney cell culture paradigm.

Such assays are particularly important in situations where a large number of foods/bioactives need to be screened rapidly and in a cost effective manner in order to ultimately identify lead foods/bioactives that can be the subject of in vivo assays. The book is an asset to researchers wishing to study the health benefits of their foods and food bioactives of interest and highlights which in vitro/ex vivo assays are of greatest relevance to their goals, what sort of outputs/data can be generated and, as noted above, highlight the strengths and weaknesses of the various assays. The cross-fertilization produced by this interdisciplinary communica tion was invaluable to those trying to understand and overcome diagnostic problems posed by hereditary metabolic disease. Cell culture methods and cryopreser vation techniques have allowed the wide distribution of biochemically abnor mal cells and their study over long periods of time. It is also evident that when a defect which produces severe metabolic disorder in man can be studied in the laboratory using isolated cell cultures a wide variety of investigative procedures can be focused on to the cellular defect without distress or discomfort to the patient or relatives.

Whereas the technique might have been considered an "art" after its introduction in 1976, it developed into a relatively simple method after it became possible to obtain high-resistance (several gigaohm) membrane-pipette seals. In the summer of 1982, a course on the technique was held at the Ettore Majorana Center for Scientific Culture in Erice, Sicily. It brought together people from most of the laboratories involved in patch clamping at that time. During the course, it became apparent that the technique had reached a state of maturity. Repeatedly, the opinion was expressed that a detailed description of all the aspects of the technique including representative examples of results should be available. We therefore asked the course instructors, as well as several other colleagues, to provide chapters on selected topics in order to produce this volume. The different variants of patch clamping were described quite extensively in an article by Hamill, Marty, Neher, Sakmann, and Sigworth (Pflugers Archiv 391:85) in 1981. Rather than repeating this survey in an introductory chapter, we chose to reprint that article in the Appendix of this volume (by permission of Springer-Verlag). The methods section will, therefore, go straight into detailed aspects of the technol ogy.
