Book excerpt: This book examines Au (I, III) complexes that selectively attack and inhibit zinc finger proteins (ZnFs) for potential therapeutic use. The author explores gold(I)-phosphine, gold(III) complexes with N^N and C^N donors as inhibitors of the HIV-1 nucleocapsid protein (NCp7), in comparison to the human transcription factor Sp1. To determine the coordination sphere of the gold adducts formed by interaction with ZnFs, two innovative approaches are used, based on Travelling-Wave Ion Mobility coupled with Mass Spectrometry (TWIM-MS), and X-ray Absorption Spectroscopy. Both approaches are proven to yield valuable structural information regarding the coordination sphere of gold in the adducts. In addition, the organometallic compound [Au (bnpy)Cl2] is evaluated. The system is shown to be capable of inhibiting ZnFs by means of C–S coupling.

Over the years gold(I) and gold(III) compounds have been used and designed to target rheumatoid arthritis, cancer, and viral diseases. New drug targets have been found for gold compounds that give insight into their mechanisms of action. Here we focus on the synthesis of Au(III) compounds designed to selectively target zinc finger (ZF) proteins. ZF proteins exhibit a variety of functions, including transcription, DNA repair, and apoptosis. Displacement of the central zinc ion, along with mutation of coordinated amino acids can result in a loss of biological function. Synthesis of complexes that selectively target zinc finger proteins, in turn inhibiting DNA/ZF interactions and therefore resulting in loss of protein function, is of great interest. Of particular interest here is the Cys3His (Cys = cysteine, His = histidine) HIV nucleocapsid zinc finger protein, NCp7. NCp7 is involved in multiple steps of the HIV life cycle, thus making it a desirable drug target. Previous studies from our group show platinated nucleobases such as [Pt(dien)(9-EtG)]2+ (dien = diethylenetriamine; 9-EtG = 9-ethylguanine) to stack effectively in a non-covalent manner with tryptophan of the C-terminal finger of HIV Nucleocapsid, NCp7(F2), a key residue involved in nucleic acid recognition. Due to the isoelectronic and isostructural relationship of Au(III) to Pt(II), we have expanded this system to Au(III)-(nucleobase/N-heterocycle) compounds. Novel Au(III)(dien)(N-heterocycle) compounds, including the first Au(III)N3(N-purine) examples, were synthesized. As previously reported for [AuCl(dien)]Cl2, these compounds exhibit pH dependency of the 1H NMR chemical shifts of the dien ligand. The acidity of the dien ligand is affected by the nature of the fourth ligand as a leaving group. The presence of an inert nitrogen donor, compared to that of the more labile Cl-, as the leaving group stabilizes the Au(III) metal center towards reduction, resulting in significant enhancement of [pi]-[pi] stacking interactions with tryptophan relative to platinum(II) and palladium(II) compounds. The presence of a more inert N-donor as the leaving group slows down the reaction with the sulfur-containing amino acid N-Acetylmethionine (N-AcMet); essentially no reaction was observed for the Au(III)-N-heterocycle compounds. All compounds react readily with N-Acetylcysteine (N-AcCys), however lack of N-heterocycle ligand dissociation indicates, to our knowledge, the first long-lived N-heterocycle-Au-S species in solution. Electrospray ionization mass spectrometry (ESI-MS) studies with NCp7(F2) indicate [Au(dien)(DMAP)]3+ (DMAP = 4-dimethylaminopyridine) to be the least reactive of the Au(III) compounds studied, showing the presence of intact NCp7(F2) zinc finger at initial reaction times. Reactivity of the Au-compounds was compared with that of Sp1(F3), a Cys2His2 ZF; in contrast, no intact ZF was observed for any of the compounds studied, suggesting the mode of action of these compounds is dependent on the nature of the zinc binding core. ESI-MS studies were expanded to that of the full HIV NCp7 zinc finger. [Au(dien)(9-EtG)]3+ reacts quickly with NCp7, resulting in immediate zinc ejection and replacement with up to three gold ions. Unlike with [Au(dien)(DMAP)]3+, no intact NCp7 was observed. Addition of [Au(dien)(9-EtG)]3+ to preformed NC-SL2 complex results in release of free RNA; based on EMSA (electrophoretic mobility shift assay) studies, [Au(dien)(9-EtG)]3+ disrupts the NCp7-RNA complex with an IC50 of ~450 [micro]M. It is possible that this HIV nucleocapsid-nucleic acid antagonism may result in a loss of viral activity.

In modern times, gold has been used in the treatment of a number of different human diseases including rheumatoid arthritis, cancer, and viral infections. This dissertation will focus on the development of gold complexes for the purpose of selective inhibition of HIV NCp7, a 55 amino acid zinc finger protein with two Cys3His zinc binding domains. NCp7 is involved in a number of viral life cycle processes including activation of reverse transcription, integration, DNA recognition, RNA packaging, and formation of the viral envelope. The diversity in the roles of NCp7 across the viral life cycle make it a highly attractive target for chemical intervention. Ejection of the tetrahedrally coordinated zinc atoms, modification or deletion of the zinc coordinating amino acids, or modification or deletion of the "essential" tryptophan residue can result in the loss of viral infectivity. This is due to the inability of NCp7 to recognize its "natural" substrate -- polynucleic acids. Previous studies have investigated the differences in the manner in which platinum(II), gold(I), and gold(III) complexes interact with NCp7. The platinum(II) complex [Pt(dien)(9-EtGua)]2 interacts with the C-terminal zinc finger of NCp7 in a non-covalent manner, through a [pi]-[pi] stacking interaction between the platinated 9-EtGua and the "essential" tryptophan residue. The isostructural and isoelectronic complex [Au(dien)(9-EtGua)]3 ejects the tetrahedrally coordinated zinc atom and replaces it with a gold atom, forming "gold fingers". This result is consistent with the interactions of the gold(I) complex [(PPh3)Au(9-EtGua)]+. In order to complete the series of isoelectronic and isostructural platinum(II), palladium(II), and gold(III) complexes with N-heterocyclic ligands and diethylenetriamine chelates, the complexes [Au(dien)(1-MeCyt)]3+ and [Au(N-Medien)(1-MeCyt)]3+ were synthesized. These complexes were found to dimerize the C-terminal zinc finger once the central zinc atom is ejected. This is likely the result of a charge transfer from the 1-methylcytosine ligand to the tryptophan residue, and is a product that was not seen as a result of interaction with the previously published 4-dimethylaminopyridine and 9-ethylguanine analogs. The 1-methylcytosine complexes also stabilize the gold(III) oxidation state and associate with N-acetyltryptophan in a manner consistent with the previously studied gold(III) analogs. Finally, in order to address concerns arising from the inner filter effect, a proof of concept study using 1H-NMR spectroscopy was utilized to show that the complex [Au(dien)(1-MeCyt)]3+ likely has a lower association constant with N-acetyltryptophan than the value determined by fluorescence quenching. The impact of the incorporation of additional steric hindrance on the gold(III) chelate was studied using the di-(2-picolyl)amine ligand. The gold(III) chlorides incorporating this ligand and the centrally methylated analog were found to eject zinc from the C-terminal zinc finger of NCp7, and the electronegativity differences between the gold(III) and platinum(II) metal centers were highlighted. The attempts to incorporate an N-heterocyclic ligand into these complexes were unsuccessful due to the steric and electronic demands of the chelate. The use of an organometallic chelating ligand led to the investigation of the ability of gold(III) complexes to catalyze the arylation of zinc-coordinating cysteine residues. The complex [AuCl2(dampa)], which had been formerly investigated as a chemotherapeutic agent due to its structural similarities to cisplatin, was found to arylate N-acetylcysteine, glutathione, and NCp7. The arylation was not found to be dependent on the cis- chloride ligand, as blocking that site with the ligand triphenylphosphine did not prevent the arylation of NCp7. The X-Ray crystal structure of the complex [AuCl(dampa)(PPh3)](PF6) was also solved. Using the advancement of the knowledge of how the electronic and structural properties of gold(III) complexes described herein impact interactions with NCp7, it is possible that a coordination complex that is a selective inhibitor of NCp7 may eventually be developed.

These metal complexes contain N-heterocyclic carbene (NHC) ligands which are able to form strong M-C(NHC) bonds, conferring high stability and favorable lipophilicity, reactivity and binding specificity of metal complexes on biomolecules. The author demonstrates significant advances made in anti-cancer gold(III), gold(I) and platinum(II) complexes. Detailed chemical synthesis, in vitro and/or in vivo anti-cancer activities are clearly presented including: (i) a class of Au(III) complexes containing a highly fluorescent N^N^N ligand and NHC ligand that simultaneously act as fluorescent thiol “switch-on” probes and anti-cancer agents; (ii) a dinuclear gold(I) complex with a mixed diphosphine and bis(NHC) ligand displaying favorable stability and showing significant inhibition of tumor growth in two independent mice models with no observable side effects; and (iii) a panel of stable luminescent cyclometalated platinum(II) complexes exhibiting high specificity to localize to the endoplasmic reticulum (ER) domain, inducing ER stress and cell apoptosis. Cisplatin and follow-up drugs carboplatin (ParaplatinTM) and oxaliplatin (EloxatinTM) have been the gold standard for metallodrugs in clinical settings as antineoplastic agents. While effective, these drugs (either alone or in combination therapy) have faced a number of clinical challenges resulting from their limited spectrum of activity, high toxicity leading to significant side effects, resistance, poor water solubility, low bioavailability and short circulating time. In the past 10 years, various unconventional non-platinum metal-based agents have emerged as a potential alternative for cancer treatment. These compounds are highly effective and selective in cancers resistant to cisplatin and other chemotherapeutic agents. Research in this area has recently exploded with a relevant number of patents and clinical trials, in addition to reports in scientific journals. Furthermore, in parallel to the synthesis of coordination and organometallic compounds comprising many different metals and unconventional platinum-based derivatives, researchers are focused on optimizing mechanistic and pharmacological features of promising drug candidates. This Special Issue aims to highlight the latest advances in anticancer metallodrugs with a focus on unconventional anticancer agents, as well as novel activation, targeting and delivery strategies aimed at improving their pharmacological profile.
