Book excerpt: The present volume represents the proceedings of the symposium on npyridine Nucleotide-Dependent Dehydrogenases" which was held on the campus of the re cently established University of Konstanz, Germany, from September 15 to 20, 1969. The aim of the symposium was to provide a forum for discussion among the experts interested in the various aspects of pyridine nucleotide-dependent de hydrogenases and the pyridine coenzymes, so as to evaluate the state of the present knowledge and to stimulate further progress in this field. In order to facilitate discussion and personal contact it was necessary to restrict the number of participants to 90 including the invited speakers, who came from fifteen countries (Australia, Austria, Denmark, England, Finland, France, Germany, Hungary, Israel, Italy, Japan, Netherlands, Sweden, Switzerland, USA). The printed lectures in this volume differ only in minor respects from those circulated be fore the meeting. In some cases speakers presented new experimen tal material during the symposium which was added as an addendum to the papers. The discussions were not recorded. The participants were requested to provide a written report of what they considered worth including in the published re port. This, together with the answers of the speakers is presented in this vo lume. I whish to thank all the speakers and discussants for their cooperativity in preparing the manuscript. The symposium was sponsored by the International Union of Biochemistry whose President, Professor Theorell, was also present and who was acting as one of the speakers and chairmen.

The structure and function of lactate dehydrogen ase, alcohol dehydrogenase, and glyceraldehyde 3-phosphate dehydrogenase are compared. These are the relatively simple oligomeric dehydrogenases for which very extensive structural and mechanistic information is available. Kinetic studies are dealt with in general terms, with selected examples. Genetic and structural relationships revealed by primary structure are discussed. General features of the specificity of dehydrogen ase reactions are described. The pyridine nucleotide - disulphide oxidoreductases exemplify a type of multicomponent enzyme complex. Lipoamide dehydrogenase, glutathione reductase, and thioredoxin reduct ase are described and compared. The fatty acid synthesizing systems of some organ isms are nonaggregated, whereas in other organisms they are multienzyme complexes containing, in animal tissues for example, two closely related multifunctional polypeptide chains. The dehydrogenase activities of the fatty acid syn thesizing systems afford many points of interest, which are described and discussed. Hydroxymethylglutaryl-CoA reductase occupies a key position in the synthesis of isoprenoid compounds, including cholesterol. It probably occurs in all forms of life; in animal cells it is a microsomal enzyme. Its properties are reviewed.

Book excerpt: Inhaltsangabe:Introduction: Metabolic reduction is the counterpart to oxidative pathways and plays an important role in the phase-I metabolism of carbonyl group bearing substances. Carbonyl reduction means the formation of a hydroxy group from a reactive aldehyde or ketone moiety and is generally regarded as an inactivation or detoxification step since the resulting alcohol is easier to conjugate and to eliminate. Not only are these carbonyl-containing compounds widespread in the environment and enter the body as xenobiotics and environmental pollutants, but they can also be generated endogenously through normal catabolic oxidation and deamination reactions. Many endogenous compounds such as biogenic amines, steroids, prostaglandins and other hormones are metabolized through carbonyl intermediates. In addition, lipid peroxidation within the cell results in the production of reactive carbonyls such as acrolein, 4-hydroxynonenal, 4-oxononenal and malon-dialdehyde, while oxidative damage to DNA generates base propenals. Dietary sources of carbonyl-containing compounds are diverse and include aldehydes found in fruits as well as the breakdown product of ethanol, acetaldehyde. Pharmacologic drugs represent further sources of exposure to carbonyl-containing compounds. From the pharmacologist s point of view, carbonyl reduction has been shown to be of significance in various inactivation processes of drugs bearing a carbonyl group. On the other hand, the carbinols formed may retain therapeutic potency, thus prolonging the pharmacodynamic effect of the parent drug, or, in some instances, a compound gains activity through carbonyl reduction. From the toxicologist s point of view, carbonyl reduction plays an important role in the toxification of drugs such as daunorubicin and doxorubicin (cf. chapter 4), whereas numerous reports corroborate the concept of carbonyl-reducing enzymes being involved in detoxification processes of endogenous and xenobiotic reactive carbonyl compounds. Compared with the oxidative cytochrome P450 (CYP) system, carbonyl-reducing enzymes had, for a long time, received considerably less attention. However, the advancement of carbonyl reductase molecular biology has allowed the identification and characterization of several carbonyl-reducing enzymes, including pluripotent hydroxysteroid dehydrogenases that are involved in xenobiotic carbonyl compound metabolism, in addition to catalyzing the oxidoreduction of their physiologic [...]

Book excerpt: Prior to the start of the eighth meeting, I had the good sense to ask Professor Rosa Angela Canuto of Turin, Italy if she would help me organize the ninth meeting. She quickly suggested that both she and Dr. Guiliana Muzio, also of Turin, help plan the meet ing. Each of our previous eight meetings was a unique experience for the participants. The science was always outstanding and the presentations and discussions were excellent. By moving each meeting to a different part of the world we were able to experience exciting foods and cultural aspects of the world in addition to the science. The ninth meeting was no exception. We met from June 18 to 22 in the small mountain city of Varallo, Italy, the birth place of Dr. Canuto. Holding the scientific sessions in a several-hundred-year-old converted mansion and having an afternoon trip to either Lago Maggiore or Monte Rosa made some aspects of this meeting extremely memorable. An additional unique aspect of the social portion of the meeting was our ability to invite the townspeople to share with us a concert performed in an old church. Though the social and cultural aspects of the meeting were outstanding, the pur pose of the meeting was to exchange scientific information about the status of the three enzyme systems.

Prof. Keith Tipton, Chairman of the Biochemistry Department at Trinity College, kindly agreed to host the meeting. On behalf of all of us who attended I wish to extend our sincere thanks to the whole Tipton family for making us feel so welcome in Ireland. It has been a decade since the frrst workshop was held in Bern, Switzerland. The scope of the meetings reflected somewhat the changes that have occurred in biochemistry during the past decade. At the first meeting primarily enzymes and their properties were discussed. At this last meeting many of the talks centered on gene regulation as well as more traditional aspects of enzymology and metabolism. During the past decade site directed mutagenesis to probe for the active site of an enzyme has become part of traditional enzymology; this was virtually unheard of at our frrst meeting. Many of the presenters now used this tool to study some aspect of structure and function of one of the three carbonyl metabolizing enzymes.

At our 1992 meeting in Dublin we learned that the next ISBRA meeting would be held in Brisbane, of all our previous meetings, I was very concerned Australia. As the scientific organizer about holding a meeting in the Southern Hemisphere for fear that many of our potential participants would not travel that far. I am pleased to say that I was proven to be incorrect. Nearly 90 scientists from a dozen countries participated at our seventh conference. At this meeting, like at all our previous ones, much new information about the three enzyme systems was presented. Of equal importance was, like at all our previous meetings, the extreme openness of the participants to discuss ideas, future directions and unpublished data. On behalf of all the participants I wish to express our sincere thanks to our Massey University colleagues for the excellent organization of this Palmerston North, New Zealand meeting. These included Kathryn Kitson, Michael Hardman, Paul Buckley, Trevor Kitson and Len Blackwell. At this meeting a few new innovations were introduced. Though posters are common at many meetings, bush walks and visits to nature preserves to see kiwi birds Our hosts were able to secure support from the International Union of Biochemistry are not.
