Book excerpt: This dissertation explores quantitative two-photon absorption spectroscopy to relate molecular structure with optical properties of organic chromophores. The dissertation describes an advanced fluorescence-based technique for reliable measurements of the two-photon spectra and cross sections. To facilitate the measurements it establishes a set of reference compounds measured with a 15% absolute accuracy covering a broad range of excitation and fluorescence wavelengths. The dissertation shows that in many cases the few-essential-levels model can be successfully applied for the description and interpretation of two-photon absorption spectra and cross sections, at least for the low-energy transitions. The dissertation presents examples of applications of two-photon absorption for volumetric optical storage and cancer tumor detection. It describes the basic principles of the two-photon absorption-based optical memory and limitations imposed on two-photon sensitivity of photochromic materials by a necessity of fast access to the data. It also proposes a novel technique for sensitive detection of cancer cells by using two-photon excitation of near-IR fluorescence of a commercial dye and discusses the mechanisms responsible for differentiation between the normal and the cancer cells. The methods described in this dissertation can be applied to understanding the relations between structure and two-photon absorption strength of individual transitions of organic and biological chromophores, which can be used for design of new materials, maximally adapted for particular applications.

Book excerpt: This project explored structure-property relationships relevant to enhancement of two-photon absorption (2PA) in organic dendritic molecules for optical power limiting. We synthesized and studied two series of novel monodisperse dendrimers built of identical blocks of thriphenylamine-based chromophores, BDPAS and BDPADSB. With both series, we observed strong cooperative enhancement of two- and three-photon absorption. We showed that this effect is due to conjugation between constituent branches. For lower generation dendriemers, the peak 2PA cross section was found to increase faster than linearly with the number of pi-electrons. For the largest dendrimer, 4SG2, the largest absolute values of molecular 2PA- and 3PA cross section was shown, 1.1 x 10(exp-46) cm(exp 4)s s and 10(exp-79) cm(exp 6)s(exp2), respectively. We compared the 2PA cross section of the two dendrimer series, and concluded that enlarging the conjugation system between the adjacent nitrogen atoms can create remarkably large sigma value. We developed a new method for quantitative determination of the extent of conjugation by comparing one- and three-photon absorption measurements. Coherent domain structure was deduced from 3PA measurements in the BDPAS series. It was found that, in terms of largest coherence domain size, 4SG0 is the optimum dendrimer generation, which corresponds to an effective maximum number of 150 conjugated pi-electrons.

One such nonlinear process, two-photon absorption (TPA), has had a wildly successful adoption into the field of biological imaging and microscopy. As far and as fast as this field is progressing, limitations stemming from the use of ultrafast lasers are starting to appear. In this work, an alternative nonclassical light source will be motivated for the application of low photon-flux two-photon microscopy. The origin and properties of the chosen nonclassical source, spontaneous parametric downconversion (SPDC), will be discussed along with the spatial and spectral properties modelled. Nonlinear processes such as TPA and sum frequency generation (SFG) will be viewed as "ultrafast coincidence measurements" of two photons arriving at a molecule within the time window of excitation. These ultrafast coincidence measurements will be viewed in an alternative manner: in terms of the second-order coherence from a light source. This degree of second-order coherence can be subdivided into two categories arising from different combinations of correlations within and between entangled photon pairs. Of interest, the energy-time correlations within the photon pair allow for enhancements in ultrafast coincidence rates over coherent light sources. The makings of an experimental setup to demonstrate enhanced rates from ultrafast two-photon coincidences taking place in SFG in a nonlinear crystal will be discussed.
