Book excerpt: Whereas platinum compounds are already clinically used as anticancer agents, tin compounds exhibiting high enough antitumour activity have not yet been found to enter the clinical phase. This is paradoxical with the observation that 50% of the tested compounds showed some activity, which is an abnormally high percentage. This is probably due to the fact that research of this type with tin compounds started more recently than with platinum compounds and that almost exclusively known compounds coming from laboratories working in the field of organotin compounds have been blindly tested. Some research directed towards the development of organotin compounds with some biologically favourable properties, for instance derivatives with hydrophilic or lipophilic, or with increased bio-availability characteristics, has started only recently. We thought that was now the appropriate time to bring together experts from different disciplines (biochemists, pharmacologists, organotin chemists, oncologists, etc.) working in the field of tin-based antitumour drugs, to contribute to the critical assessment of existing knowledge in this new important topic, to identify the directions for future research and to promote close working relations between the different countries and professional experiences gathered in this workshop.

Finally, lipophilic organotin compounds have been prepared that can be hydrolyzed into hydrophilic derivatives [5]. Lipophilicity could be an important factor that allows them to cross the cell membrances and their hydrolysis into hydrophilic derivatives might be necessary for them to enter the cell to reach the possible target, DNA. All these compounds have been tested either in vitro or in vivo or both. The synthesis of these organotin molecules, their characterization by NMR, their structure by x-ray diffraction and the results of the screnings will be discussed.

Cisplatin now occupies an important place in the armamentarium of the oncologist due to its effectiveness in the treatment of testicular cancer. A second generation analogue, carbo platin, offers reduced toxicity together with therapeutic activity, which gives it a place in the front-line therapy of genitourinary cancers. These and other successes have encouraged the search for novel metal-based drugs for cancer therapy. Research has shown that metal compounds have potential for activity not only as cytotoxic antitumour agents, but also in areas such as adjuvant therapy, diagnosis and immunotherapy. Then, following 21 years' service with the International Tin Research Institute, it was a great pleasure for me when the wheel turned full circle and, in 1994, Alwyn - now an Emeritus Professor - asked me to return to UCL as an Honorary Research Fellow in the Chemistry Department. One of my first tasks was when I received an invitation from Blackie A&P to edit the second edition of the Chemistry of Tin, which I was delighted to accept, since it enabled me to continued my life-long interest in tin chemistry and to maintain contact with my former friends and colleagues, many of whom have contributed to this book.
