Book excerpt: Aedes aegypti is a naturalized invasive mosquito in United States that is responsible for transmitting diseases such as yellow fever, dengue, and chickungunya around the globe. In Chapter I, I examined the phylogeography of the mosquito in south Florida using a 418 base pair partial ND5 mitochondrial DNA marker. I sampled larvae from multiple vases within a cemetery, multiple cemeteries within a city, and cities along the east and west coast of south Florida. Aedes aegypti haplotypes assemble into two clades suggesting at least two separate introductions. The relatively high genetic diversity of A. aegypti in south Florida compared to genetic diversity in other invaded areas such as Southeast Asia and Australia provides additional evidence for multiple introductions from multiple source populations into south Florida. Genetic structure of these populations did not yield evidence for deviation from stable population sizes, despite eradication measures of the 1960's or the population fluctuations that might be expected to result from vector control efforts across both coasts. There was a significant east-west split based on differences in haplotype frequencies. Haplotype and nucleotide diversities of A. aegypti populations in the west coast were higher than those for populations in the east coast, suggesting that the west coast populations were established earlier than the east coast populations. There was evidence for extensive gene flow within the west coast and a moderate gene flow within the east coast and between the two coasts. Postulated landscape barriers (rivers and bays) in the west coast of Florida did not appear to be barriers to dispersal of A. aegypti. In Chapter II, I tested whether successful human-aided A. aegypti dispersal was different for two habitat types within Tampa and Miami in South Florida. I postulated that tires and cemetery vases are larval habitats that differ in the extent of human activity, with tires often moved by humans, resulting in displacement of the larvae and eggs contained within, and cemetery vases infrequently moved by humans. Hence, I postulated that dispersal of tire site A. aegypti populations will likely be more impacted by human activity ('human-aided dispersal') when compared with cemetery populations, while the natural dispersal of A. aegypti would be similar in both habitats. I used both mitochondrial (773 base pair partial ND4 and partial ND5 mitochondrial DNA fragments) and microsatellite markers (six polymorphic loci). I sampled multiple tire sites and cemetery sites within each city. Contrary to my hypothesis, I found no evidence for greater human-aided dispersal in tire habitats compared to cemeteries, and gene flow from both habitats was similar. There was, however, a difference between the two cities, with Miami having higher proportion of dispersers than Tampa. Most of the dispersers from Miami tended to migrate within the city. The extent of migration from Tampa to Miami was greater than vice versa. My research has confirmed the existence of long-distance dispersal between A. aegypti populations in south Florida, which could be one contributor to successful establishment and persistence of this invasive species in Florida.
