Book excerpt: Complex carbohydrates and glycoconjugates mediate a host of biologically and medicinally relevant processes. In this respect, multivalency has emerged as a recognition mechanism intimately associated with carbohydrate function. Selectin-ligand interactions provide an ideal system in which to explore this phenomenon, as high affinity and specificity are governed by distinct patterns of oligovalency. Toward this end, the chemoenzymatic synthesis of sialyl-trimeric-Lewis x was accomplished. This decasaccharide is exhibited on selectin glycoprotein ligands, and has been implicated in cell adhesion processes. Following the development of an efficient synthetic protocol, the target structure was assayed as an E-selectin ligand. The significance of the Lewis x repeats was assessed by comparison to the sialyl Lewis x tetrasaccharide. To investigate P-selectin interactions, the chemoenzymatic synthesis of complex sulfo-glycopeptides from the N-terminus of PSGL-1 was undertaken. The target structure represents both segments of a hetero-dimeric recognition element (tyrosine sulfate and a sLe x O -glycan) necessary for high affinity interactions with P-selectin. A synthetic method toward obtaining the sulfo-glycopeptide was first established, and the effect of the sulfate ester on glycosyltransferase-catalyzed construction of the O -glycan was then examined. Finally, multimerization also represents an avenue for the modulation of carbohydrate-RNA interactions. As such, the synthesis of aminoglycoside dimers as novel antibiotics that target the bacterial 16S rRNA A-site was achieved. RNA affinity and antibiotic potency were determined, and the ability of these unnatural bivalent molecules to inhibit aminoglycoside-modifying enzymes was assessed. Multivalency is a recognition mechanism employed in nature, and can be exploited synthetically to enhance ligand affinity for a receptor. Especially applicable to glycobiology, multivalency is emerging as a general characteristic of carbohydrate-ligand interactions. Research of this nature may provide an improved understanding of oligovalency in biological systems, as well as novel strategies for the design of pharmaceutical agents.
