Book excerpt: According to Valentin (1833) and Luschka (1862), the first description of the structure now known as the carotid body must be ascribed to a Swiss physiolo gist - Albrecht von Haller - who, in 1762, called it the ganglion exiguum. This claim, however, may be erroneous, for Tauber (1743) described a struc ture at the bifurcation on the common carotid artery and called it the ganglion minutum. Andersch (1797) reprinted the text of a study made by his father between 1751 and 1755. The original printing of this work had apparently been sold as waste paper! Andersch called the organ the ganglion intercaroticum on account of its location. He also specifically stated that the sympathetic chain, the glossopharyngeal and the vagus nerves sent branches into the organ. For a while the carotid body remained forgotten, to be rediscovered in 1833 by Mayer of Bonn who again remarked upon the branches of the sympathetic, glossopharyngeal and vagus nerves as sources of a nerve plexus which innervated the ganglion intercaroticurtl. . Valentin (1833) clearly regarded the structure as part of the sympathetic nervous system, although he too recognised that the vagus and glossopharyngeal nerves contributed conspicuously to its innervation. Thus it is evident that the anatomists of the eighteenth and early nineteenth centuries regarded the structure in the carotid bifurcation as one of the many ganglia which are interspersed in the course of the sympathetic nervous system.

In 1966 a meeting was organized by R. W. Torrance in Oxford and in 1973 by M. J. Purves in Bristol; in 1974 A. S. Paintal organized a satellite symposium of the Physiological Congress in Kashmir. The organizers of these meetings are to be commended for their efforts in publishing both the papers and discussions. At these meetings it has become apparent that the direction of research is becoming more sharpely focused on the cellular mechanisms of chemoreception. During the meeting in Dortmund the papers dealt mostly with the different cell types in the carotid body and their environment, i. e. , local P0 and local flow. These included 2 light and electron microscopic studies of the morphometric and histochemical pro perties of the different cells, microelectrode studies of the glomoid tissue to understand the conversion of a chemical stimulation into nervous activity, as well as strictly bio chemical and physiologic investigations concerning the dependence of the chemo receptive process on O consumption and the turnover of catecholamines. In spite 2 of the variety in methodology, it was apparent that all contributors had a common interest: to understand the mechanisms of chemoreception. Although at the meeting itself there was ample time to fully discuss the various papers, it has become necessary here to shorten the papers and discussions; otherwise the cost of publication would have been prohibitive.

Book excerpt: Carotid bodies (CB) are paired, oxygen-sensing organs located in the bifurcation of the carotid artery that act as peripheral chemoreceptors in the detection of hypoxic, hypercapnic and acidotic levels in the arterial blood. CBs respond to these fluctuations in blood gases by initiating firing of the carotid sinus nerve. This ultimately results in the appropriate ventilatory change to restore blood gases to their physiological levels. Studies have shown that the hypoxic response of the carotid body in juvenile mammals is low, but as maturation occurs this response is strengthened and clearly exhibited in adults. One theory suggests mitochondria play a vital role in the oxygen sensitivity of Type I cells in carotid bodies. The experimental hypothesis of this project is that mitochondria in the oxygen sensing Type I cells of rat carotid bodies change in number and /or distribution during the development of the acute hypoxic ventilatory response. Type I cells were isolated from juvenile (4-6 days) and mature (14-16 days) rat carotid bodies and loaded with mitotracker to quantify mitochondrial content. Data obtained demonstrated a significant reduction in the total volume of mitochondria in Type I cells during development which was not paralleled by a fall in metabolic rate, indicating an increase in the rate of mitochondrial oxidative phosphorylation. These findings suggest that change in mitochondrial content during the development of the hypoxic response might be crucial for this mechanism's development.

In all there were 87 presentations by 108 scientists from 18 countries. Authors making multiple presentations at Park City combined their results into single, longer papers for this volume. As a result this vol~me offers the reader 63 contributions of state-of-the-art research in this important and exciting field. Inasmuch as oxygen is the substrate sine qua non for the survival of all higher organisms, it is quite understandable that considerable interest sur rounds investigations into mechanisms responsible for detecting dwindling oxygen supplies in the organism. This interest has intensified as the newer techniques of cell, sub-cell, and molecular biology have become available. As detectors of insufficient oxygen in the arterial blood the arterial che moreceptors (carotid and aortic bodies) initiate many cardiopulmonary reflexes geared toward maintaining constant the delivery of oxygen to the tissues. These chemoreceptors, which also trigger secretions from the ad renal glands, are located near the carotid sinus and in the arch of the aorta.

ISAC was established with the aim of providing a framework to support the increasing number of investigators from a wide variety of disciplines (anatomists, pathologists, respiratory physiologists and clinicians, high altitude physiologists, biochemists, biophysicists, physiologists and pharmacologists) who share a common interest in arterial chemoreception. ISAC took over the co-ordination of the international chemoreceptor meetings, with the membership deciding the venue for forthcoming meetings. During the Park City symposium Dublin was selected to host the 1993 meeting, under the Presidency of Professor Ronan O'Regan. The 12th International Meeting on Arterial Chemoreception, which was held in Dublin in August 1993, was acclaimed as a great success by all those present. The delegates not only shared in a wide-ranging feast of chemoreceptor based science, they had plenty of opportunity during the meeting for renewing acquaintances and establishing new friendships. The location for the meeting at University College Dublin's modem Belfield campus helped to promote such interaction, and the social programme was outstanding.
