Book excerpt: "Carcinoembryonic antigen (CEA), a widely used clinical tumor marker, functions in vitro as an adhesion molecule and is the prototype for a family of glycoproteins, which includes nonspecific crossreacting antigen (NCA) and biliary glycoprotein (BGP). This study examines the control processes responsible for the overproduction of the CEA family in carcinomas and identifies the trans-acting factors which mediate CEA and BGP gene transcription. During differentiation and $ gamma$-interferon treatment, CEA and NCA are regulated at both transcriptional and post-transcriptional levels and transcriptional control is markedly different between these two corresponding genes. Characterization of the CEA gene promoter revealed both negative and positive regulatory elements and tissue-specific, differentiation-dependent interactions with USF, Sp1, Sp1-like, AP-2-like and novel silencer transcription factors. Characterization of the BGP promoter identified one positively-acting element binding both USF and HNF-4/LF-A1, thereby contributing to distinct BGP gene expression. The Sp1, Sp1-like and silencer factors are specific to CEA gene transcription." --

This volume will be of interest to researchers in the fields of intercellular adhesion, evolution, and molecular recognition, as well as clinicians interested in the recent knowledge regarding a tumor marker commonly utilised in the clinic, and companies wishing to exploit this cancer-related family for novel commercial applications in the biomedical sphere.

Book excerpt: "Human nonspecific cross-reacting antigen (NCA), a close relative of the tumour marker human carcinoembryonic antigen (CEA), is also an in vitro homotypic intercellular adhesion molecule capable of inhibiting differentiation when expressed ectopically in myoblasts. Moreover, NCA appears to be overexpressed at the transcriptional level to a greater extent and more frequently in colorectal carcinomas than CEA. This study examines the transcriptional control mechanisms responsible for orchestrating NCA expression. The region within 283 bp upstream of the translational start site of the NCA gene was found to be capable of directing high levels of expression in functional promoter assays. Footprinting experiments identified three cis-acting elements and mobility-shift assays revealed that the first of these elements is bound by USF1 and USF2, and the second two are bound by Sp1 and Sp3. No cis-acting elements corresponding to CEA FP4 or the silencer CEA FP5 of the CEA promoter were detected in the NCA promoter, which may contribute to the differential expression of NCA versus CEA in tumourigenesis." --

Book excerpt: "Carcinoembryonic antigen (CEA) is a tumor marker used clinically to assess post-operative recurrences of breast, lung and colon cancers. The CEA gene family, which is part of the immunoglobulin superfamily, is composed of several proteins crossreactive with anti-CEA antibodies. Carcinoembryonic antigen appears to function in the development of the gastro-intestinal tract as well in tumor formation; it is capable of mediating cell-cell adhesion in vitro which is consistent with its putative role of maintaining tissue architecture in vivo. CEA gene family members have also been identified in various tissues of the mouse. Two of the mouse proteins, mmCGMla and mmCGMlb, have been characterized. By sequence homology, mmCGMla and mmCGMlb are the mouse homologues of human biliary glycoprotein and of rat hepatocyte ecto-ATPase. Both of the mouse CEA related proteins function as adhesion molecules when expressed on the cell surface of transfectant cells; however, mmCGMla, unlike mmCGMlb, mediates cellular aggregation irrespective of calcium concentration or temperature. Sequence comparison of mmCGMla, mmCGMlb, and other cDNAs isolated by polymerase chain reaction techniques, demonstrates that there are at least eight possible transcripts encoding CEA-related proteins and that these transcripts are all produced by alternative splicing of one precursor messenger RNA." --
