In order to celebrate this landmark, a symposium in the form of a series of Monday evening lectures was held in his honor at the Marine Biological Labora tory throughout the summer of 1980. The selection of speakers was made from among those investigators who had been either his students or co-workers. One intent of the symposium was to examine the current status of knowledge of those areas of interest in excitable membrane structure and function that owe their initiation or encouragement to Kacy Cole. The papers assembled in this volume represent a large majority of the presentations given during the 1980 Cole Symposium. It seems clear on examination of these papers that Kacy's interests in membrane impedance, ion channel conductances, channel fluctuation phenomena, excitation, and the development of membrane biophysical methodology are all being actively pursued. It is also clear that many of his suggestions have borne fruit. Of these, his invention of the voltage v vi Preface clamp method has been most productive. It is hoped that these papers will provide new directions for investigations into the nature of excitable membrane phenomena. The organizers of the symposium and the editors of this volume wish to express their thanks to the Marine Biological Laboratory for making available the facilities for the symposium. Book excerpt: The predecessor to this book was A Guide to the Laboratory Use of the Squid Loligo pealei published by the Marine Biological Laboratory, Woods Hole, Massachusetts in 1974. The revision of this long out of date guide, with the approval of the Marine Biological Laboratory, is an attempt to introduce students and researchers to the cephalopods and particularly the squid as an object of biological research. Therefore, we have decided to expand on its original theme, which was to present important practical aspects for using the squid as experimental animals. There are twenty two chapters instead of the original eight. The material in the original eight chapters has been completely revised. Since more than one method can be used for accomplishing a given task, some duplication of methods was considered desirable in the various chapters. Thus, the methodology can be chosen which is best suited for each reader's requirements. Each subject also contains a mini-review which can serve as an introduction to the various topics. Thus, the volume is not just a laboratory manual, but can also be used as an introduction to squid biology. The book is intended for laboratory technicians, advanced undergraduate students, graduate students, researchers, and all others who want to learn the purpose, methods, and techniques of using squid as experimental animals. This is the reason why the name has been changed to its present title. Preceding the chapters is a list of many of the abbreviations, prefixes, and suffixes used in this volume.

Calorimetry, electron spin and nuclear magnetic resonance, X-ray diffraction, and freeze-cleavage electron microscopy, reinforced by biochemical analyses and enzymatic studies, have led to universal acceptance of a generalized membrane model. All membrane biologists would agree that a major element of all biological membranes is a bilayer of phospholipids which, in some instances, also contains other lipids, notably sterols and glycolipids. The fatty acid com position of the lipids of most membranes is such that the lipids are above their transition temperatures in their normal environment so that the bilayer is fluid. The microviscosity of the fatty acyl groups decreases pro gressively down the chain so that, at the hydrocarbon interior of the bilayer, the lipid phase has a viscosity approximating that of olive oil at room temperature. As a consequence of this membrane fluidity, a phospholipid molecule is very mobile within the plane of the membrane (moving a distance of about 1-2 fLm in 1 s) but the movement of a phospholipid molecule from one side of the membrane bilayer to the other (flip-flop) is very slow. The lipid bilayer is an essentially inert and rather impermeable struc ture, as shown by many studies with model systems. Proteins, of course, provide the catalytic components of the membranes. as well as playing a significant structural role.

Book excerpt: This thesis focuses on the dynamics of autonomous Boolean networks, on the basis of Boolean logic functions in continuous time without external clocking. These networks are realized with integrated circuits on an electronic chip as a field programmable gate array (FPGA) with roughly 100,000 logic gates, offering an extremely flexible model system. It allows fast and cheap design cycles and large networks with arbitrary topologies and coupling delays. The author presents pioneering results on theoretical modeling, experimental realization, and selected applications. In this regard, three classes of novel dynamic behavior are investigated: (i) Chaotic Boolean networks are proposed as high-speed physical random number generators with high bit rates. (ii) Networks of periodic Boolean oscillators are home to long-living transient chimera states, i.e., novel patterns of coexisting domains of spatially coherent (synchronized) and incoherent (desynchronized) dynamics. (iii) Excitable networks exhibit cluster synchronization and can be used as fast artificial Boolean neurons whose spiking patterns can be controlled. This work presents the first experimental platform for large complex networks, which will facilitate exciting future developments.
