Available in PDF, EPUB and Kindle. Book excerpt: A reaction sequence leading to the core ring system of Dragmacidin E was proposed. The only prior total synthesis occurred in 25 steps. This study investigated a reaction sequence proposed to have fewer overall steps with a lower cost for the formal synthesis. Experimental results elucidated that attempted substitutions at C(3) of the indole did not provide the intended products. However, a Vilsmeier formylation successfully added a formyl group to C(3) of 4-bromoindole. A new scheme leading to a 6-5-7 indole ring system starting with the Vilsmeier formylation of 4-bromoindole was envisioned. A formal synthesis could be initiated with 4-bromotryptophan, leading to a functionalized C(3)-C(4) bridged indole.

Particular emphasis is placed both on how photochemistry contributes to these disciplines and on what the current developments are. The book starts with a general description of the interaction between light and matter, which provides the general background to photochemistry for non-specialists. The following chapters develop the general synthetic and mechanistic aspects of photochemistry as applied to both organic and inorganic materials, together with types of materials which are useful as light absorbers, emitters, sensitisers, etc. for a wide variety of applications. A detailed discussion is presented on the photochemical processes occurring in the Earth’s atmosphere, including discussion of important current aspects such as ozone depletion. Two important distinct, but interconnected, applications of photochemistry are in photocatalytic treatment of wastes and in solar energy conversion. Semiconductor photochemistry plays an important role in these and is discussed with reference to both of these areas. Free radicals and reactive oxygen species are of major importance in many chemical, biological and medical applications of photochemistry, and are discussed in depth. The following chapters discuss the relevance of using light in medicine, both with various types of phototherapy and in medical diagnostics. The development of optical sensors and probes is closely related to diagnostics, but is also relevant to many other applications, and is discussed separately. Important aspects of applied photochemistry in electronics and imaging, through processes such as photolithography, are discussed and it is shown how this is allowing the increasing miniaturisation of semiconductor devices for a wide variety of electronics applications and the development of nanometer scale devices. The final two chapters provide the basic ideas necessary to set up a photochemical laboratory and to characterise excited states. This book is aimed at those in science, engineering and medicine who are interested in applying photochemistry in a broad spectrum of areas. Each chapter has the basic theories and methods for its particular applications and directs the reader to the current, important literature in the field, making Applied Photochemistry suitable for both the novice and the experienced photochemist.

A promising therapeutic target, its synthetic challenge is attributed to its heptacyclic core. In this study, we propose the synthesis of a Dragmacidin E heptacyclic core precursor, mediated through a Lewis acid (LA) mediated cyclization of an acceptor-acceptor-donor (AAD) cyclopropane. Utilizing a model study, alkoxy AAD cyclopropanes were investigated to develop a protocol for precursor synthesis. After generating various ethyl-a-diazobenzoyl acetate derivatives, the metal catalyzed cyclopropanation reaction of these compounds was studied. With vinyl acetate and Rh2esp2, acetoxy AAD cyclopropanes were synthesized in yields ranging from 12 % - 53 %. These cyclopropanes were successfully generated and rearrangement into dihydrofuran products was avoided. To complete our model study, LA cyclization of acetoxy AAD cyclopropanes was studied. Using stoichiometric quantities of TiCl4, naphthol derivatives were synthesized in one step.

In the first part of the thesis, the author introduces the research background and reviews the research progress in total synthesis of Maoecrystal V. In the second part, the author develops a novel and concise approach for the stereo selective construction of the tetracyclic model system of Maoecrystal V. The model system is accomplished in 8 steps with 20% yield. In the third part, the author describes the first successful total synthesis of Maoecrystal V and investigates four strategies for constructing the key tetrahydrofuran oxa-bridge skeleton. The total synthesis starts from a known compound and is accomplished in 17 steps with 1.2% yield. The successful total synthesis of Maoecrystal V will contribute to the development of efficient synthetic strategies for natural products and other compounds with complex structures.
