For these reasons, the study of bioactive marine metabolites should rely on the development of chemical synthesis and synthetic strategies aimed at the preparation of pure compounds and analogs both for structural confirmation and/or for the large-scale preparation necessary for future applications. Moreover, natural products can be a crucial starting point for the preparation of molecules structurally inspired by the latter, opening the path to new classes of biologically active compounds with pharmacological potential. This book collects original research articles regarding synthetic strategies for secondary marine metabolites and/or analogs that favor applications of these molecules and/or solve structural challenges common in the field of natural substances.

Book excerpt: Biotechnology may be defined as the application of scientific and engineering principles to the processing of materials by biological agents to provide goods and services (Bullet al. , 1982, p. 21) or as any technique that uses living organisms (or parts of organisms) to make or modify products, to improve plants or animals, or to develop microorganisms for specific use (OTC, 1988). In line with these broad definitions we can consider marine biotechnology as the use of marine organisms or their constituents for useful purposes in a controlled fashion. This series will explore a range of scientific advances in support of marine biotechnology. It will provide information on advances in three categories: (1) basic knowledge, (2) ap plied research and development, and (3) commercial and institutional issues. We hope the presentation of the topics will generate interest and interaction among readers in the academic world, government, and industry. This first volume examines chemical and biological properties of some natural products that are useful or potentially useful in research and in the chemical and pharmaceutical industries. One chapter describes a system for producing such substances on a large scale. Biotechnology incorporates molecular biology in order to go beyond tradi tional biochemical technology such as the production of antibiotic drugs from bacterial cultures in bioreactors. Development of the technology for production of antibiotics in this way resulted from fundamental advances in chemistry, phar macology, microbiology, and biochemical engineering.

Little wonder then, that synthetic methodology has achieved a high degree of predictability and total synthesis of natural products has been successfully directed toward ever more challenging targets. As for the second point, that of single authorship, multiple authorship would invariably have led to gaps and overlaps, thus making it difficult to assemble and assess recent research in a systematic and comprehens ive fashion.

The Andersen lab examines compounds from marine sources that can be used as lead structures for drug discovery. Synthetic studies, structure-activity relationships (SAR) and biological findings of two such compounds are described in this thesis. The first is pelorol, a meroterpene isolated from a tropical sponge Dactylospongia elegans. Pelorol is a small molecule activator of SHIP 1, a phosphatase that is a negative regulator of the P13K pathway in hematopoetic cells. Using a synthetic route from a previous co-worker, Lu Yang, a series of SHIP 1 activating compounds based on pelorol were synthesized. These compounds were evaluated for selectivity, potency, and efficacy in a series of biological studies, leading to the discovery of 2.27 as a preclinical lead compound. Water-soluble prodrugs of the SHIP 1-activating compounds were also synthesized and their properties reported. The second compound examined is ceratamine A, an alkaloid isolated from the sponge Pseudoceratina sp. from Papua New Guinea. Ceratamines A and B are microtubule stabilizing antimitotic agents that may be useful in cancer chemotherapy. The core imidazo[4,5,d]azepine heterocycle of the ceratamines has no precedent among known synthetic or natural compounds. The relatively simple structure of the ceratamines and the novel antimitotic phenotype they generate makes them attractive targets. Desbromo ceratamine A (3.44) was synthesized by an efficient and scaleable route, confirming the structure of ceratamine A and validating the biological activity of the core pharmacophore. Synthetic efforts towards ceratamine A were ultimately thwarted by the inability to install the bromine atoms present in the natural product. A significant finding is that the bromine atoms in ceratamine A contribute significantly to the antimitotic potency of the compound necessitating a bioisosteric approach to more potent antimitotic ceratami.

Book excerpt: Marine natural products (MNPs) are, undoubtedly, highly valuable starting points for drug-development programs. Generally, MNPs show a well-defined spatial orientation that have evolved for thousands of years to interact with their biological target and the special and unique conditions present in seas, lakes, oceans and rivers allow the biosynthesis of highly potent compunds. Nowadays, the processes to discover a new drug from MNPs are bio-assay guided, giving more importance to the biological results rather than to the molecular structure. After a molecular entity has shown to be biologically relevant the next step is to determine the structural and stereochemical information of such molecule. It is at this step where the project described in this doctoral thesis started. Phormidolides B, C and D (PM B-D) is a family of complex molecules that were isolated in 2010 by the pharmaceutical company Pharmamar S.A. during an expedition to collect samples in the Pemba Island (Madagascar). The extracts rendered a mixture of PM B-D that, once purified, were tested on an IC50 assay against three tumour cell lines. The results showed activity in the low micromolar range with a still unknown mechanism of action. After extensive HRMS, 1D-NMR and 2D-NMR techniques a possible structure was proposed. Following previous UB doctoral theses (Dr. Adriana Lorente and Dr. Janire Lamariano) this doctoral thesis describes, as a compendium of publications, several synthetic studies with the final objective of achieving a robust total synthesis to confirm the structural information of PM B-D by chemical synthesis. After a general introduction (Chapter 1), Chapter 2 (Org. Lett. 2015, 17, 6246.) describes an optimization for the synthesis of the macrocyclic core present in PM B-D. Chapter 3 (Chem. Eur. J. 2016, 21, 7033.) deals, for the first time, with the synthesis and introduction of the most challenging part of the molecule: the bromo-methoxy-diene (BMD) moiety present at the end of the polyhydroxylated chain. After this finding, Chapter 4 (Org. Lett., 2016, 18, 4485.) illustrates the enantioselective synthesis of the polyol chain present in Oscillariolide and Phormidolides, including the previously mentioned BMD moiety. Chapter 5 reports the preparation of the fatty acids present in PM C and D. Finally, after the previous synthetic optimizations, Chapter 6 describes the first synthetic approach for the construction of this large family of natural products using a highly convergent approach. Finally, as an annex a review article entitled "Role of the Nozaki-Hiayama-Kishi-Takai coupling in the synthesis of natural products" (Chem. Rev., 2017, 117, 8420.) is included to deeply explain the uses of this reaction used, as well, for the synthesis of phormidolide B-D. A section with the general conclusions extracted from this thesis is also included.

Several marine based pharmaceuticals are under active commercial development, ecosystem health is high on the public's list of concerns, and aquaculture is providing an ever greater proportion of the seafood on our tables. Nevertheless, marine biotechnology has not yet caught the public's, or investor's, attention. Two workshops, held in October 1999 and November 2001 at the National Academies, were successful in highlighting new developments and opportunities in environmental and biomedical applications of marine biotechnology, and also in identifying factors that are impeding commercial exploitation of these products. This report includes a synthesis of the 2001 sessions addressing drug discovery and development, applications of genomics and proteomics to marine biotechnology, biomaterials and bioengineering, and public policy and essays contributed by the workshop speakers.
