Book excerpt: (Written for the original Russian edition) Out of all the functional groups of protein molecules that be long to the side chains of amino acid residues, the sulfhydryl or thiol (SH) groups of cysteine residues and the disulfide (S-S) groups of cystine have for long attracted particular attention from chemists and biochemists. This is due, on the one hand, to the high chemical reactivity of these groups, which enter into many, and often very characteristic and selective, reactions with several types of compound. On the other hand, this attention is connected with the great significance of SH and S-S groups for the specific functions of a number of enzymes, hormones, and other biological ly active proteins, and consequently for the normal course of many physiological processes. The varied functions of sulfhydryl and disulfide groups in en zymes are particularly interesting; their role can consist of bind ing substrates and cofactors, in direct participation in the catalytic act, or in maintaining (in ways not so far sufficiently studied) the native, catalytically active conformation of the protein. It is entirely natural that the detailed study of the chemical behavior and biological role of these sulfur-containing groups in proteins (and in such low-molecular cofactors as glutathione, co enzyme A, lipoiC acid, etc. ) has, in addition to its academic, gen eral biological interest, great Significance for many applied branches of medicine and biology (toxicology, radiobiology, diagnostics and therapeutics, biochemical engineering, plant growing, etc. ).

Book excerpt: The number of disulfide (SS) bonds in Glycinin-the major soybean protein-and their rate of cleavage by dithiothreitol (DTT) were determined at various concentrations of urea. A progressive increase in SS bond scission was observed with increasing urea concentration reaching the maximum of 20 SS bonds at 8M urea. In addition, a linear relationship was obtained for the rate of SS cleavage as a function of urea concentration. These results indicate that most of the disulfide bridges are buried in the interior of the protein molecule. Upon exposure to alkaline denaturation, a maximum of 9.2 sulfhydryl (SH) groups were obtained per mole of glycinin at pH 11.9.

They included volumes entitled Bioorganic Mechanisms, I and II by T.C. Bruice and S.J. Benkovic, published in 1965, the volume entitled Catalysis in Chemistry and Enzymology by W.P. Jencks in 1969, and the volume entitled Enzymatic Reaction Mechanisms by C.T. Walsh in 1979. The Walsh book was based on the course taught by W.P. Jencks and R.H. Abeles at Brandeis University in the 1960's and 1970's. By the late 1970's, much more could be included about the structures of enzymes and the kinetics and mechanisms of enzymatic reactions themselves, and less emphasis was placed on chemical models. Walshs book was widely used in courses on enzymatic mechanisms for many years. Much has happened in the field of mechanistic enzymology in the past 15 to 20 years. Walshs book is both out-of-date and out-of-focus in todays world of enzymatic mechanisms. There is no longer a single volume or a small collection of volumes to which students can be directed to obtain a clear understanding of the state of knowledge regarding the chemicals mechanisms by which enzymes catalyze biological reactions. There is no single volume to which medicinal chemists and biotechnologists can refer on the subject of enzymatic mechanisms. Practitioners in the field have recognized a need for a new book on enzymatic mechanisms for more than ten years, and several, including Walsh, have considered undertaking to modernize Walshs book. However, these good intentions have been abandoned for one reason or another. The great size of the knowledge base in mechanistic enzymology has been a deterrent. It seems too large a subject for a single author, and it is difficult for several authors to coordinate their work to mutual satisfaction. This text by Perry A. Frey and Adrian D. Hegeman accomplishes this feat, producing the long-awaited replacement for Walshs classic text.

(Nucleic acids, carbohydrates, and other biopolymers are correspondingly affected.) Since proteins are ubiquitous, the consequences of their crosslinking are widespread and often profound. Scientists from many disciplines including organic chemistry, bio chemistry, protein chemistry, food science, nutrition, radiation biology, pharmacology, physiology, medicine, and dentistry are, therefore, minutely interested in protein crosslinking reactions and their implications.
