Book excerpt: Stripe rust (Puccinia striiformis f. sp. tritici) has been receiving increased attention in the USA since the appearance of more virulent races detected in the past decade. These races caused yield losses even in areas where the disease previously was rarely detected. Host plant resistance is the most cost effective and environmentally friendly means of controlling stripe rust. Deployment of single, major genes for resistance has been compromised due to the genetic variability of the pathogen, emphasizing the need to breed for durable resistance. 'Stephens' wheat (Triticum aestivum L.) has been grown commercially in the Pacific Northwest region of the USA for 30 years and shows resistance to P. striiformis f. sp. tritici "old" and "new" races. The durable resistance of Stephens to stripe rust is believed to be due to a combination of seedling and adult plant resistance genes. Multilocation field trials and molecular genotyping were used to study the genetics of the resistance response in 'Stephens'. Disease severity, based on the percentage of leaf area infected in field plots, was recorded in eight locations: five in 2008 and three in 2009. Locations could be divided into two mega-environments based on time of appearance of disease and QTL identified. Quantitative trait loci (QTL) analysis identified 11 chromosomal regions associated with resistance to stripe rust in the resistant parent ('Stephens') and two regions derived from the susceptible parent ('Platte'). Many QTL locations coincided to those of previous reports. Furthermore, this study suggests that a combination of additive resistance genes acting at different plant stages is responsible for the durable resistance of 'Stephens'. Significant QTL x environment interactions were found, suggesting that specificity to plant stage, race, and/or temperature are responsible for different disease responses.

Book excerpt: As a result of a recent spread of a new and highly virulent race of Puccinia graminis f. sp. tritici called Ug99, stem rust is becoming a serious threat to wheat production in Ethiopia and other east African countries. Therefore, there is an urgent need to analyse the virulence spectrum of Pgt populations in Ethiopia. Wheat stem rust samples were collected in 2006 and 2007 in Arsi, Bale, Shewa and the Northwest regions of Ethiopia to determine the virulence diversity and race distribution in P. graminis f. sp. tritici populations. Stem rust incidence was high in Arsi, Bale and East Shewa. In Northwest Ethiopia, and North and West Shewa, stem rust was prevalent at low levels. A total of 152 isolates was analyzed and 22 races were identified. Races TTKSR (Ug99), TTHSR and RRTTR were predominant with frequencies of 26.6, 17.7 and 11.1%, respectively. These races were also detected in all regions. It turned out that the highly virulent race designated as Ug99 was present throughout the country and dominated in all regions except Northwest Ethiopia. A variant of Ug99, which is virulent against Sr24 was not detected in this study. Four stem rust resistance genes (Sr13, Sr30, Sr36 and SrTmp) were found to confer resistance to most of the races prevalent in Ethiopia. With the exception of Sr30, which is not effective to Ug99, the other three genes can be used in breeding for resistance to stem rust in Ethiopia.

F. sp. tritici), is an economically significant pathogen affecting durum wheat (Triticum turgidum var. durum L.) crops worldwide. Genetic resistance is one of the most effective and environmentally friendly methods to control leaf rust in wheat. New sources of resistance genes need to be identified due to the threat of new Pt races to durum wheat production. Marker assisted selection (MAS) is a highly efficient method to select resistance genes in breeding programs particularly to pyramid multiple resistance genes in new varieties. The objective of this study was to characterize and map leaf rust resistance genes in a Canadian durum wheat Strongfield. A double haploid (DH) mapping population of 87 DH lines was developed from the cross Strongfield/Blackbird. Seedling rust tests with Pt isolates 12-3 MBDS, 06-1-1 TDBG, 128-1 MBRJ, 74-2 MGBJ, and 77-2 TJBJ revealed a single hypersensitive leaf rust resistance gene. Three genes segregated for resistance to isolate 1-1 BBBD at the seedling stage, one of which controlled resistance to the other five Pt isolates. Blackbird contributed one of the seedling resistance genes effective against isolate 1-1 BBBD. Parental lines and 87 DH lines were genotyped using the Illumina Infinium assay with the iSelect 90K wheat SNP array. A database search using the DNA sequences of linked markers provided a putative location in the Chinese Spring reference genome sequence. The gene conferring resistance to the six isolates used in the study mapped to the long arm of chromosome 3A and was temporarily designated as LrStr_3A. No leaf rust resistance gene has been detected in this region previously. Quantitative trait locus (QTL) analysis identified eight QTL controlling leaf rust resistance in field leaf rust nurseries. One of these QTL mapped to chromosome 3AL as the same region of LrStr_3A. Another QTL mapped to the expected location of the adult plant resistance (APR) gene Lr46 based upon the marker csLV46G22. Kompetitive allele-specific PCR (KASP) markers were developed for LrStr_3A that will be useful for MAS.
