Book excerpt: A study was made of contact microradiographic techniques for the x-ray evaluation of tin specimens or selected small regions in large specimens. Extraneous absorbers such as air and the film holder were removed to permit the use of low-energy x rays, and high-resolution plates were used which permit magnifications up to 500x. Microradiographs are presented for graphite-coated uranium carbide particles, nickel screen, thin graphite section, and various fibrous materials. It is concluded that the contact microradiographic technique is a very useful tool. (D.L.C.).

Book excerpt: The compound optical microscope, in its various modern forms, is probably the most familiar of all laboratory instruments and the electron microscope, once an exotic rarity, has now become a standard tool in biological and materials research. Both instruments are often used effectively with little knowledge of the relevant theory, or even of how a particular type of microscope functions. Eventually however, proper use, interpretation of images and choices of specific applications demand an understanding of fundamental principles. This book describes the principles of operation of each type of microscope currently available and of use to biomedical and materials scientists. It explains the mechanisms of image formation, contrast and its enhancement, accounts for ultimate limits on the size of observable details (resolving power and resolution) and finally provides an account of Fourier optical theory. Principles behind the photographic methods used in microscopy are also described and there is some discussion of image processing methods. The book will appeal to graduate students and researchers in the biomedical sciences, and it will be helpful to students taking a course involving the principles of microscopy.

Book excerpt: Recent developments in techniques for low-voltage radiography have found application for the examination and evaluation of minute details in small or thin specimens. Utilization of x-ray tubes and helium chambers with thin windows for maximum transmission of soft x rays and bare film with dark-room exposure techniques have allowed very high contrast radiographs to be produced on low-contrast materials. These conditions, coupled with very high resolution photographic emulsion detectors, have resulted in the observation of detail approximately 1 {micron} in size. Useful examination of the plates and subsequent reproductions have been made at magnifications as high as 500X. This contact system offers the advantage of evaluation of a relatively large area, as compared to the restricted field of observation of projection microradiography. Included among the subjects have been graphite-coated UC2 particles, graphite, beryllium, paper, and other organic specimens.

The rapid evolution which microscopy in its broadest sense has since undergone and which has contributed greatly to our insight in many fields of biological science and medicine has, however, lead to a progressive specialisation. Both experienced investigators in clinical and biological laboratories and post graduate students, confronted with a limited number of microscopic tech niques in their daily research work, have increasing difficulty in keeping (or obtaining) a general idea of the many time-honoured and new possibilities which microscopy has to offer. This book has been written with the aim of presenting general informa tion on light microscopic techniques, at a level somewhere in between booklets like those provided by microscope manufacturers (which are often too much focussed on the production program of a particular make) and very advanced treatises with a thorough mathematical treatment of all phenomena concerned. The physically oriented texts moreover often do not sufficiently take into account the practical situation in a medical or biolog ical laboratory; on the other hand, the value of really understanding what one is doing in using a microscopic technique is often underestimated. At tempt has been made, therefore, to present sufficient background informa tion necessary for a rational application of the different microscopical tech niques in their mutual relationship.

Book excerpt: In 1979, a conference on x-ray microscopy was organized by the New York Academy of Sciences, and in 1983, the Second Interna tional Symposium on X-ray Imaging was organized by the Akademie der Wissenschaften in Gottingen, Federal Republic of Germany. This volume contains the contributions to the symposium "X-ray Microscopy '86", held in Taipei, Taiwan, the Republic of China in August 1986. This is the first volume which intends to provide up-to date information on x-ray imaging to biologists, therefore, emphasis was given to specimen preparation techniques and image interpreta tion. Specimen preparation represents a major part of every microscopy work, therefore, it should be strongly emphasized in this emerging field of x-ray microscopy. Theoretically, x-ray microscopy offers the potential for the study of unfixed, hydrated biological ma terials. Since very few biological system can be directly observed without specimen preparation, we would like to emphasize that new information on biological specimens can only be obtained if the speci men is properly prepared. In the past decade, many of the published x-ray images were obtained from poorly prepared biological speci mens, mainly air-dried materials. Therefore, one of the goals of this conference is to bring the importance of specimen preparation to the attention of x-ray microscopy community. X-ray microscopy can be subdivided into several major areas. They are the classic x-ray projection microscope, x-ray contact imag ing (microradiography) and the more recent x-ray scanning micro scope, x-ray photoelectron microscope and x-ray imaging microscope.
