Book excerpt: RNA helicases are essential in all steps of RNA maturation, beginning with transcription and ending with RNA decay. They catalyze the separation of nucleic acid double strands and thereby facilitate structural remodeling. Their cellular importance is reflected in severe diseases caused by RNA helicase dysregulation. The largest group of RNA helicases is confined by the so called DEAD-box helicases of superfamily 2, characterized by the signature sequence D-E-A-D. DEAD-box helicases share a structurally conserved core of two RecA-like domains that carry signature motifs involved in ATP-binding, ATP-hydrolysis, RNA-binding and RNA-remodeling. An exceptional member of the DEAD-box protein family is the human RNA helicase DDX1(DEAD-box helicase1). In contrast to all other family members, DDX1 harbors a SPRY domain insertion in between the signature motifs of the helicase core. DDX1 is involved in a plethora of different RNA maturation processes and has been associated with tumor progression. Moreover due to its versatile function in RNA processing, it is hijacked by viruses for their replication. This medical relevance makes DDX1 a potential target for the development of pharmaceutics.

Book excerpt: Helicases are the proteins that bind to double- or single-stranded DNA and/or RNA chains to unwind higher order structures, usually consuming energy from the hydrolysis of ATP molecules. The biological roles of helicases are associated with a variety of DNA and/or RNA metabolisms, including DNA-replication, -repair, -recombination, RNA processing, and transcription. Dysfunctions of helicases cause various diseases, such as xeroderma pigmentosum (XP), premature aging syndrome, cancer and immunodeficiency, in humans. Moreover, recent genetic analyses revealed that mutations in helicase-encoding genes are frequently found in patients of specific diseases. Some helicases regulate cellular senescence by controlling integrity of genomes, and others play a role in neuromuscular functions presumably by modulating processing of mRNAs. However, the molecular mechanisms of how helicases are regulated in order to maintain our health are not yet fully understood. In this research topic, we will focus on the expression and functions of helicases and their encoding genes, reviewing recent research progresses that provide new insights into development of clinical and pharmaceutical treatments targeting helicases.

Considerable resources and efforts have been directed at understanding the interaction between these retroviruses and their host which may provide clues as to how the infection can be controlled or prevented. Among the key scientific successes is the identification of intracellular “restriction factors” that have evolved as obstacles to the replication of pathogens including infectious retroviruses. The discovery of APOBEC, which are strong mutagens of retroviral genomes and intracellular retroelements, began a new era of intense research activities into the spectrum of intrinsic anti-HIV activity, leading to the identification of TRIM5a, BST2/Tetherin, and SAMHD1. In response, HIV has evolved several accessory genes as weaponries to evade these intracellular restriction activities. The intracellular antiretroviral defenses evolved in response to endogenous retroelements that make up more than 40% of the entire mammalian genome, and which are regarded as ancestors of infectious retroviruses. LTR-type retroelements are present in all higher eukaryotes, representing about 8% of the human genome. Non-LTR retroelements can be found at extremely high copy numbers also, with a significant portion of mammalin genomes consisting of LINEs. Mammalian genomes are modified by LINEs through insertions, but also by the indirect replication of non-autonomous retrotransposons such as SINEs. LINEs insertion was shown to have played, and continue to play important roles in genomic evolution and somatic genome mosaicism-mediated physiology. And, because retrotransposition can confer genetic diversity that is beneficial to the host, the vertebrate intrinsic immunity has evolved to support a balance between retroelement insertions that confer beneficial and those that cause deleterious gene disruptions. The articles published in this Research Topic should serve not only as valuable references for the field, but provide future topics of research for investigators that should further our understanding of the retrovirus, retroelements and their restrictions.

Interestingly, an increasing number of RNA helicases have been shown to be required for more than one cellular pathway. Therefore, protein cofactors have been suggested to recruit multifunctional RNA helicases to their substrates and regulate their activity. The best characterised family of helicase cofactors are G-patch proteins, which all contain a glycine-riche domain. This work focuses on the characterisation of the yeast RNA helicase Prp43 and its G-patch protein cofactors. Additionally, a new G-patch protein cofactor of Prp43 was identified and characterised. Together, the data suggest that protein cofactors can regulate the distribution and activity of RNA helicases in different pathways.
