Oxidants, whether intentionally produced or by-products of normal metabolism can either mediate a variety of critical biological processes but when present inappropriately cause extensive damage to biological molecules (DNA, proteins, and lipids). These effects can lead to either damage that is a major contributor to aging and degenerative diseases (or to other diseases such as cancer, cardiovascular disease, immune-system decline, brain dysfunction, and cataracts) or normal physiological function- tissue repair, defense against pathogens and cellular proliferation. On the other hand the body is equipped with a complex antioxidant/oxidant handling system which includes both enzymatic and nonenzymatic (i.e. small molecules such as flavonoids, ascorbate, tocopherol, and carotenoids) produced endogenously or derived from the diet. This book focuses on how the same molecules can have favorable or noxious effects depending on location, level and timing. Each chapter focuses on one particular molecule or oxidant/antioxidant system and provides a state of the art review of the current understanding regarding both positive and negative actions of the system under review.

Book excerpt: Viral vectors are superior tools for gene therapy and as a genetic vaccine platform because viruses have evolved to efficiently infect and transfer their genomes to cells. Several impressive successes in viral vector-based gene therapies have been reported in humans, including restoration of vision in patients with Leber’s congenital amaurosis by retinal gene transfer and cures for severe immune deficiencies by gene transfer to hematopoietic stem cells. However, the mammalian immune system has evolved in parallel to fend off invading pathogens such as viruses. Innate and antigen-specific adaptive immune responses against viral vectors and therapeutic transgene products pose serious hurdles for successful gene therapy. Pre-existing immunity in humans, resulting from prior exposure to the parent virus that forms the basis for the gene transfer vehicle may be derived from, often prevents efficient gene transfer. This problem also reduces our ability to use certain vectors for genetic vaccination or in anti-cancer therapy. For these reasons, the gene transfer community has been extensively studying the mechanisms of immune responses against viral vectors and has started to develop strategies and protocols to block or circumvent such responses. Choice, design and engineering of a vector as well as the route of administration/target tissue can be optimized/ altered to minimize immune responses or evade pre-existing immunity. Immune suppression and modulation strategies are being developed in order to minimize inflammation, prevent antibody or T cell responses against vectors, and to promote tolerance to therapeutic gene products. Combinations of these approaches will likely facilitate clinical applications of gene therapy for many target diseases and also aid in vaccine development.

Indeed, our evolving concepts have, like so many political parties, been forced into dramatic "V-turns" and contortions. Within our working lives, we have had to debate whether radicals made any con tribution to any pathology, whilst now it is difficult not to entertain the view that every physiological process is pivotally controlled by exquisitely sensitive radical reactions. Inflammation is, of course, an example of pathology evolving from physiology, and in this book we have called upon both scientists and clinicians who have research interests in the complex switching mechanisms that sustain these transi tions. The book as a whole explores, from a physiological standpoint, how deter ministic radical systems sensitive to their initial conditions can interdigitate, iterate and feed back to control diverse cellular processes that create the inflammatory response. Whilst systems such as these to a mathematician would provide the basis for a chaotic response, one is forced to marvel how, for all stages of an inflammatory reaction, this system appears exquisitely controlled, making therapeutic manipula tion both possible and, to some extent, predictable.
