Book excerpt: In spite of a long history of intense investigation the transmissible spongiform encephalopathies remain a poorly understood family of neurodegenerative diseases. This group of diseases has been described in a wide variety of animal species and includes kuru, Creutzfeldt-Jakob disease, and Gerstmann-Straussler syndrome in humans, and scrapie, bovine spongiform encephalopathy, and related syndromes in ruminants and rodents. In all cases spongiform degeneration and astrocytosis are seen in specimens of brain and a filterable transmissible agent is present in the brain and some other tissues of affected individuals. However, the precise nature of this agent remains unknown. Agent infectivity, which can so far only be assayed by serial transmission to new individuals, be remarkably resistant to inactivation has been shown to by heat, chemicals, and irradiation. These properties create significant biohazard possibilities during exposure to infected tissues. Transmission between humans was originally reco gnized in the unique epidemiology of kuru in New Guinea tribesmen, and concern about transmission from animals to humans has re-emerged as a result of the current epidemic of bovine spongiform encephalopathy in dairy cattle in Great Britain. Although interspecies transmission has often been achieved experimentally, its efficiency is highly variable. There fore, the possibility of spread of bovine spongiform encephalopathy from cattle to humans or various animal populations cannot be accurately predicted at this time. This volume presents a comprehensive update of know ledge concerning the transmissible spongiform encephalo pathies.

The meeting was of great interest because, for the first time, a group of highly qualified scientists specializing in different fields were gathered together to discuss the problem of demyelinating diseases. In thi. context molecular biologists, immunologists, virologists, and lipid chemists could discuss with clinicians for four days every aspect of demyelinating diseases. They shared results of the more recent research with the hope that more knowledge of the myelinating process will eventually succeed in finding new treatments for these diseases. The meeting was successful, perhaps in part because discussions during lunches and dinners provided an opportunity for the exchange of new ideas. This was apparent in the discussions held after the presen tation of each paper and was summarized in the very lively round table which was held on the last day. Book excerpt: Four decades ago, lymphocytic choriomeningitis virus was discovered, at about the same time and independently, in 3 different localities in the U.S.A. Armstrong and Lillie encountered the agent in a monkey when they passaged a recent isolate of the St. Louis encephalitis virus; Rivers and Scott isolated 5 strains from patients with meningitis; and Traub revealed the virus in a colony of albino mice. Already in these first isolations mice were incriminated, and later observa tions proved beyond doubt that M. musculus is the principal reservoir of the virus in nature. For some time LCM virus was regarded as the sole etiologic agent of Wallgren's "meningite aseptique aigue". Soon, however, it became clear that Wallgren's syndrome had a multitude of causes, among which the LCM virus was of little relevance, and in subsequent years it disappeared from the sight of most viro logists. Indeed, it might have fallen into oblivion had it not been for Erich Traub who, practically all by himself, continued to investigate the intricate relationship between this virus and its natural host, the common house mouse.
