Book excerpt: In front of you is the finished product of your work, the text of your contributions to the 2003 Dayton International Symposium on Cell Volume and Signal Transduction. As we all recall, this symposium brought together the Doyens of Cellular and Molecular Physiology as well as aspiring young investigators and students in this field. It became a memorable event in an illustrious series of International Symposia on Cell Volume and Signaling. This series, started by Professors Vladimir Strbák, Florian Lang and Monte Greer in Smolenice, Slovakia in 1997 and continued by Professors Rolf Kinne, Florian Lang and Frank Wehner in Berlin in 2000, is projected for 2005 in Copenhagen to be hosted by our colleague, Professor Else Hoffmann and her team. We dearly miss Monte Greer to whom this symposium was dedicated and addressed so eloquently by Vladimir Strbák in his Dedication to Monte. Monte and I became friends in Smolenice and had begun to discuss the 2003 meeting only a few days before his tragic accident in 2002. There are others who were not with us, and we missed them, too. We would not have been able to succeed in this event without the unflagging support of our higher administration at Wright State University, the NIDDKD of the National Institute of Health, and the Fuji Medical System (see Acknowledgments).

This has been made quite clear by the fluid-mosaic model which considers the cell membrane as a two-dimensional solution of a mosaic of integral membrane proteins and glycoproteins firmly embedded in a fluid lipid bilayer matrix. Such a model has several virtues, chief among which is that it allows membrane components to diffuse in the plane of the membrane and orient asymmetrically across the membrane. The model is also remarkable since it provokes the right sort of questions. Two such examples are: Does membrane fluidity influence enzyme activity? Does cholesterol regulate fluidity? However, it does not go far enough. As it turns out, there is now another version of this model, the so-called post-fluid mosaic model which incorporates two concepts, namely the existence in the membrane of discrete domains in which specific lipid-lipid, lipid-protein and protein-protein interactions occur and ordered regions that are in motion but remain separate from less ordered regions. We must admit that both are intriguing problems and of importance in guiding our thinking as to what the next model might be. We have chosen not to include the subject of membrane transport in the present volume. This obviously represents a break with convention. However, the intention is to have the topic covered subsequent volumes relating to organ systems. It would be right to regard this as an attempt to strengthen the integrated approach to the teaching of medicine.

The interaction of an extracellular agonist (First Messenger) with its plasma membrane receptor leads to the transmission of a signal across the cell membrane and results in the production and/or activation of other signalling molecules (Second Messengers). These Second Messengers control the action of many protein kinases and protein phosphatases and so lead to cellular responses. Although the biochemical basis of the transduction of signals in the main signalling systems in eukaryotic cells is probably largely known, intensified research is ongoing in the following areas: the discovery of specific substrates for many protein kinases, elucidation of the biological significance of the differential tissue expression and heterogeneity of many signalling proteins, and the unravelling of diverse interactions (such as signal potentiation, synergism, antagonism and neuronal co-transmission) between signalling systems. As knowledge from such studies accumulates, it is becoming clear that the `cross talk' interactions between signalling systems are important features of dynamic cell regulation.
