Book excerpt: Available online: https://pub.norden.org/temanord2020-507/ Abstract [en] This Nordic report builds on ideas brought forward in the OECD Detailed Review Paper (DRP) 178 on screening and testing methods and endpoints for evaluating endocrine disruptors. It was identified that the retinoid system is not presently included in any OECD test guideline, and that it is urgent to cover this knowledge gap. The present TemaNord report is a contribution to an upcoming OECD DRP on retinoids. An overview of retinoid biology in female and male reproduction is presented. The report identifies that, in general, there is a lack of studies investigating effects of chemicals on reproduction, while simultaneously examining effects on retinoid related parameters. This is a major data gap. However, selected in vitro and/or in silico retinoid-related endpoints, presented in this report, could possibly be part of a broader screening test battery aimed at reproductive toxicity.

Book excerpt: The first demonstration of the existence of a vitamin and the full recognition of this fact are often attributed to the work of McCollum, who found that a sub stance in butterfat and cod-liver oil was necessary for growth and health of ani mals fed purified diets. It became obvious that an organic substance present in microconcentrations was vital to growth and reproduction of animals. Following the coining of the word vitamine by Funk, McCollum named this fat-soluble sub stance vitamin A. We can, therefore, state that vitamin A was certainly one of the first known vitamins, yet its function and the function of the other fat-soluble vitamins had remained largely unknown until recent years. However, there has been an explosion of investigation and new information in this field, which had remained quiescent for at least two or three decades. It is now obvious that the fat-soluble vitamins function quite differently from their water-soluble counter parts. We have learned that vitamin D functions by virtue of its being converted in the kidney to a hormone that functions to regulate calcium and phosphorus metabolism. This new endocrine system is in the process of being elucidated in detail, and in addition, the medical use of these hormonal forms of vitamin D in the treatment of a variety of metabolic bone diseases has excited the medical com munity.

Book excerpt: Scientific Frontiers in Developmental Toxicology and Risk Assessment reviews advances made during the last 10-15 years in fields such as developmental biology, molecular biology, and genetics. It describes a novel approach for how these advances might be used in combination with existing methodologies to further the understanding of mechanisms of developmental toxicity, to improve the assessment of chemicals for their ability to cause developmental toxicity, and to improve risk assessment for developmental defects. For example, based on the recent advances, even the smallest, simplest laboratory animals such as the fruit fly, roundworm, and zebrafish might be able to serve as developmental toxicological models for human biological systems. Use of such organisms might allow for rapid and inexpensive testing of large numbers of chemicals for their potential to cause developmental toxicity; presently, there are little or no developmental toxicity data available for the majority of natural and manufactured chemicals in use. This new approach to developmental toxicology and risk assessment will require simultaneous research on several fronts by experts from multiple scientific disciplines, including developmental toxicologists, developmental biologists, geneticists, epidemiologists, and biostatisticians.

As part of its efforts to reduce or eliminate exposure of Naval personnel and their families to reproductive and developmental toxicants, the Navy requested that the National Research Council (NRC) recommend an approach that can be used to evaluate chemicals and physical agents for their potential to cause reproductive and developmental toxicity. The NRC assigned this project to the Committee on Toxicology, which convened the Subcommittee on Reproductive and Developmental Toxicology, to prepare this report. In this report, the subcommittee recommends an approach for evaluating agents for potential reproductive and developmental toxicity and demonstrates how that approach can be used by the Navy. This report has been reviewed in draft form by individuals chosen for their diverse perspectives and technical expertise, in accordance with procedures approved by the NRC's Report Review Committee. The purpose of this independent review is to provide candid and critical comments that will assist the institution in making its published report as sound as possible and to ensure that the report meets institutional standards for objectivity, evidence, and responsiveness to the study charge. The review comments and draft manuscript remain confidential to protect the integrity of the deliberative process. It highlights the need to develop a regulatory framework to govern the use of insects for food security. And it presents case studies and examples from around the world. Book excerpt: The foundation of quantitative genetics theory was developed during the last century and facilitated many successful breeding programs for cultivated plants and t- restrial livestock. The results have been almost universally impressive, and today nearly all agricultural production utilises genetically improved seed and animals. The aquaculture industry can learn a great deal from these experiences, because the basic theory behind selective breeding is the same for all species. The ?rst published selection experiments in aquaculture started in 1920 s to improve disease resistance in ?sh, but it was not before the 1970 s that the ?rst family based breeding program was initiated for Atlantic salmon in Norway by AKVAFORSK. Unfortunately, the subsequent implementation of selective breeding on a wider scale in aquaculture has been slow, and despite the dramatic gains that have been demonstrated in a number of species, less than 10% of world aquaculture production is currently based on improved stocks. For the long-term sustainability of aquaculture production, there is an urgent need to develop and implement e- cient breeding programs for all species under commercial production. The ability for aquaculture to successfully meet the demands of an ever increasing human p- ulation, will rely on genetically improved stocks that utilise feed, water and land resources in an ef?cient way. Technological advances like genome sequences of aquaculture species, and advanced molecular methods means that there are new and exciting prospects for building on these well-established methods into the future.
