Book Regulation of Autoimmune Disease by B7 CD28 CTLA 4 mediated Costimulation

Download or read book Regulation of Autoimmune Disease by B7 CD28 CTLA 4 mediated Costimulation written by Nitin Jayant Karandikar and published by . This book was released on 1997 with total page 326 pages. Available in PDF, EPUB and Kindle. Book excerpt: The present study was conducted to understand the role of costimulatory interplay in regulating autoimmune pathogenesis in R-EAE. Interference with CTLA-4-mediated interactions, using monoclonal antibodies to CTLA-4 or their F(ab) fragments, enhanced proliferation, cytokine production and disease transfer by $\rm PLP\sb{139-151}$-primed lymph node cells. In vivo treatment with anti-CTLA-4 mAb at various stages of the disease resulted in exacerbation of the disease course and acceleration of epitope spreading with enhancement of responses to $\rm PLP\sb{139-151},\ PLP\sb{178-191}$ and $\rm MBP\sb{84-104}.$ Thus, we conclude that CTLA-4 plays an important downregulatory role in the spread of immune responses during ongoing autoimmune destruction.

Available in PDF, EPUB and Kindle. Book excerpt: Leukocyte culture conferences have a long pedigree. This volume records some of the scientific highlights of the 16th such annual con ference, and is a witness to the continuing evolution and popularity of leukocyte culture and of immunology. There is strong evidence of the widening horizons of immunology, both technically, with the obviously major impact of molecular biology into our understanding of cellular processes, and also conceptually. Traditionally, the 'proceedings' of these conferences have been published. But have the books produced really recorded the major part of the conference, the informal, friendly, but intense and some times heated exchanges that take place between workers in tackling very similar problems and systems and which are at the heart of every successful conference? Unfortunately this essence cannot be incorpo rated by soliciting manuscripts. For this reason, we have changed the format of publication, retaining published versions of the symposium papers, but requesting the workshop chairmen to produce a summary of the major new observations and areas of controversy highlighted in their sessions, as a vehicle for defining current areas of interest and debate. Not an easy task, as the workshop topics were culled from the abstracts submitted by the participants, rather than being on predefined topics. The unseasonal warmth in Cambridge was reflected in the atmos phere of the conference, the organization of which benefited from the administrative skills of Jean Bacon, Philippa Wells, Mr. Peter Irving, and Mrs.

Book T Cell Costimulation in Anti tumor Immunity and Autoimmunity

Download or read book T Cell Costimulation in Anti tumor Immunity and Autoimmunity written by Kenneth F. May and published by . This book was released on 2004 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Abstract: Costimulatory molecules, including 4-1BB, CTLA-4, and B7, play a critical role in the activation, sustenance, and regulation of T cell immune responses. Manipulation of these pathways holds promise for the development of therapies for cancer and autoimmunity. Anti-4-1BB monoclonal antibody (mAb) has been demonstrated to boost anti-tumor immunity in animal models. Using a model of tumor-specific CD8 T cell adoptive immunotherapy, we demonstrate that anti-4-1BB mAb can mediate the rejection of large established tumors in the absence of CD4 T cell help. Anti-4-1BB mAb increases populations of tumor-specific CD8 T cells in peripheral blood by reduction of activation-induced cell death, but not increased T cell proliferation. The use of anti-CTLA-4 mAb has also been shown to enhance anti-tumor immunity. Here we employ two novel "humanized" mouse models to screen anti-human CTLA-4 mAb for translation to human cancer therapy. Using the hu-PBL-SCID model of Epstein-Barr virus (EBV)-associated lymphoproliferative disease, we show that anti-human CTLA-4 mAb promotes the in vivo expansion of human CD8 and CD4 T cells, and the generation of antigen specific CD8 T cell responses to EBV lymphoma. This correlates with reduced levels of the oncogenic EBV protein LMP-1, and increased survival in these mice. We also characterize the creation of a knock-in mouse model in which mouse T cells express the human CTLA-4 molecule. Preliminary testing in this mouse model supports the use of this model to screen anti-human CTLA-4 mAb for clinical use. Understanding the role of B7/CD28/CTLA-4 interaction in immune activation and tolerance in autoimmune disease is fundamental to successful intervention targeted at costimulatory molecules. We describe the spontaneous development of whole-body alopecia, lymphadenopathy, and skin disease in mice lacking B7 molecules. This disease is mediated by autoimmune CD4 T cells, which induce multi-organ inflammation when transferred to mice expressing B7 molecules. This disease may result from impaired development of CD4+CD25+ regulatory T cells (Treg) in B7-deficient mice. Since provision of Treg can abrogate the multi-organ inflammation despite lack of B7 molecules on the auto-pathogenic T cells, interaction between CTLA-4 on Treg and B7-1/2 on effector T cells is not essential for Treg function.

  • Author : Jenni Punt
  • Publisher : Macmillan Higher Education
  • Release : 2018-10-16
  • ISBN : 1319172989
  • Pages : 2997 pages

Download or read book Kuby Immunology written by Jenni Punt and published by Macmillan Higher Education. This book was released on 2018-10-16 with total page 2997 pages. Available in PDF, EPUB and Kindle. Book excerpt: Janis Kuby’s groundbreaking introduction to immunology was the first textbook for the course actually written to be a textbook. Like no other text, it combined an experimental emphasis with extensive pedagogical features to help students grasp basic concepts. Now in a thoroughly updated new edition, Kuby Immunology remains the only undergraduate introduction to immunology written by teachers of the course. In the Kuby tradition, authors Jenni Punt, Sharon Stranford, Patricia Jones, and Judy Owen present the most current topics in an experimental context, conveying the excitement of scientific discovery, and highlight important advances, but do so with the focus on the big picture of the study of immune response, enhanced by unsurpassed pedagogical support for the first-time learner. Punt, Stranford, Jones, and Owen bring an enormous range of teaching and research experiences to the text, as well as a dedication to continue the experiment-based, pedagogical-driven approach of Janis Kuby. For this edition, they have worked chapter by chapter to streamline the coverage, to address topics that students have the most trouble grasping, and to continually remind students where the topic at hand fits in the study of immunology as a whole.

Book B Cell Receptor Signaling

Download or read book B Cell Receptor Signaling written by Tomohiro Kurosaki and published by Springer. This book was released on 2015-12-26 with total page 231 pages. Available in PDF, EPUB and Kindle. Book excerpt: This volume details our current understanding of the architecture and signaling capabilities of the B cell antigen receptor (BCR) in health and disease. The first chapters review new insights into the assembly of BCR components and their organization on the cell surface. Subsequent contributions focus on the molecular interactions that connect the BCR with major intracellular signaling pathways such as Ca2+ mobilization, membrane phospholipid metabolism, nuclear translocation of NF-kB or the activation of Bruton’s Tyrosine Kinase and MAP kinases. These elements orchestrate cytoplasmic and nuclear responses as well as cytoskeleton dynamics for antigen internalization. Furthermore, a key mechanism of how B cells remember their cognate antigen is discussed in detail. Altogether, the discoveries presented provide a better understanding of B cell biology and help to explain some B cell-mediated pathogenicities, like autoimmune phenomena or the formation of B cell tumors, while also paving the way for eventually combating these diseases.

Book CD4 CD25 Regulatory T Cells Origin Function and Therapeutic Potential

Download or read book CD4 CD25 Regulatory T Cells Origin Function and Therapeutic Potential written by B. Kyewski and published by Springer Science & Business Media. This book was released on 2006-01-09 with total page 331 pages. Available in PDF, EPUB and Kindle. Book excerpt: The vertebrate immune system defends the organism against invading pathogens while at the same time being self-tolerant to the body’s own constituents thus preserving its integrity. Multiple mechanisms work in concert to ensure self-tolerance. Apart from purging the T cell repertoire from auto-reactive T cells via negative selection in the thymus dominant tolerance exerted by regulatory T cells plays a major role in tolerance imposition and maintenance. Among the various regulatory/suppressive cells hitherto described, CD4+CD25+ regulatory T cells (Treg) and interleukin-10 producing T regulatory 1 (Tr1) cells have been studied in most detail and are the subject of most articles in this issue. Treg, also called "natural" regulatory T cells, will be traced from their intra-thymic origin to the site of their action in peripheral lymphoid organs and tissues. The repertoire of Treg is clearly biased towards recognition of self-antigens, thereby potentially preventing autoimmune diseases such as gastritis and oophoritis. Regulatory T cells, however also control infections, allergies and tolerance to transplanted tissues and this requires their induction in the periphery under conditions which are not yet fully understood. The concept of dominant tolerance, by far not novel, will offer new insights and hopefully tools for the successful treatment of autoimmune diseases, improved cancer immunotherapy and transplant survival. The fulfillment of these high expectations will, however, require their unambiguous identification and a better understanding of their mode of action.
