Book excerpt: Among heterocyclic compounds, quinoline is a privileged scaffold that appears as an important construction motif for the development of new drugs. Quinoline nucleus is endowed with a variety of therapeutic activities, and new quinolone derivatives are known to be biologically active compounds possessing several pharmacological activities. Many new therapeutic agents have been developed by using quinoline nucleus. Hence, quinoline and its derivatives form an important class of heterocyclic compounds for the new drug development. Numerous synthetic routes have been developed for the synthesis of quinoline and its derivatives due to its wide range of biological and pharmacological activities. The article covers the synthesis as well as biological activities of quinoline derivatives such as antimalarial, anticancer, antibacterial, anthelmintic, antiviral, antifungal, anti-inflammatory, analgesic, cardiovascular, central nervous system, hypoglycemic, and miscellaneous activities.

Book excerpt: Quinoline heterocycles found to have diverse biological activities and 3-functionalised quinolines are important intermediates for the synthesis of various quinoline derivatives. 3-acetyl-4-hydroxy-quinolin-2(1H)-one and 6-chloro-3-acetyl-4-phenyl-2-methyl quinoline were chosen as intermediates for the synthesis of some novel quinoline derivatives. In particular, only a few methods are available for the synthesis of 3-acetyl-4-hydroxy-quinolin-2(1H)-one, which involve either two step or multistep reaction protocols. In the present work, a rapid, efficient and one pot procedure for the synthesis of 3-acetyl-4-hydroxy-quinolin-2(1H)-ones has been developed both by conventional and microwave assisted synthesis. The 3-acetyl quinolines were converted into chalcones which in turn converted to quinoline-3-yl pyrazolines and quinoline-3-yl isoxazolines and a few into 3-(2, 4-substitutedstyryl)isoxazolo[4,5-c]quinolin-4(5H)-ones. These compounds were explored for their anti-microbial and cyctotoxicity studies.

Book excerpt: Acetoxy drug: Protein transacetylase catalyses the transfer of acetyl groups from acetylated polyphenolic peracetates (PAs) to the receptor proteins and modulates the activity of proteins and alters the effect of various enzymes like Glutathione S-transferase, NADPH Cytochrome c Reductase and Nitric Oxide Synthase (NOS). Accordingly, Polyphenolic acetates were found effective in the inhibition of ADP induced platelet aggregation, because of which, the acetoxy derivatives of several classes of polyphenols were found to be the substrates of TAase. Our group has synthesized a number of heterocyclic compounds e.g. benzofurans and quinolones etc. These compounds were screened for various biological activities e.g. antioxidant, antiplatelet aggregation, and antimicrobial activities.

Among those medicinal preparations are the natural and synthetic origins of quinazolinone-4 derivatives. Quinazolinone derivatives are reported to be physiologically and pharmacologically active. They also exhibit a wide range of activities such as anticonvulsant, antiinflammatory, antifungal, antimalarial, and sedative properties. Some of these compounds are identified as drugs used as diuretics, vasodilators, and antihypertensive agents. Moreover, sulfonamide derivatives have been widely used as bacteriostatic agents. Prompted by the above-mentioned facts and in conjunction with our ongoing program on the utility of readily obtainable starting material for the synthesis of heterocyclic systems of biological interest, we have decided to synthesize a series of quinazolinone derivatives having sulfonamide moiety with a potentially wide spectrum of biological responses.
