Book excerpt: Abstract: The nuclear envelope (NE) is composed of the inner nuclear membrane (INM) and the outer nuclear membrane (ONM) which is contiguous with the endoplasmic reticulum (ER). The appropriate location of NE proteins is important in cells. Integral INM proteins are proposed to be synthesized at the ER and then translocated through the nuclear pore complex (NPC). In contrast, peripherally associated INM proteins are proposed to follow a targeting mechanism to the nucleus that is similar to nucleoplasmic proteins. Our research aims to understand the mechanism of targeting of peripherally associated proteins to the INM. We employed yeast as a genetic model and the tRNA modification enzyme, Trm1-II, as a reporter. We screened a collection of temperature sensitive (ts) mutants for defects in galactose-inducible Trm1-II-GFP (Gal-Trm1-II-GFP) INM localization. We found that the majority (46%) of the ts mutations affecting Gal-Trm1-II-GFP localization were in genes that encode proteins involved in ER-Golgi homeostasis. Interestingly, about 35% of the mutated essential genes encode components of the Spindle Pole Body (SPB). In the SPB ts mutants, at the non-permissive temperature, Gal-Trm1-II-GFP accumulates as a spot that localizes to the ER, rather than being evenly distributed around the entire INM as in wild-type cells. Following the dynamics of Gal-Trm1-II-GFP we learned that its inappropriate distribution results from a failure to move from the initial contact with the NE (ONM) throughout the INM. Gal-Trm1-GFP accumulates to the ER with time, suggesting that this might be the initial Trm1-II tethering site. Surprisingly, SPB defects also affect targeting of an integral INM protein, but not a soluble nucleoplasmic protein which indicates that there is no defect in import and that appropriate SPBs are required for INM targeting of both integral and peripheral INM proteins, but not nucleoplasmic proteins. Our evidence suggests Gal-Trm1-II-GFP is alternatively transported via soluble mechanism when unable to tether to the ER. We propose a novel mechanism for peripherally associated INM proteins that combines targeting mechanisms for both integral and soluble proteins. We also learned that INM maintenance of Gal-Trm1-II-GFP was altered in SPB defective cells, which suggests that a general defect at the membrane that forms the ER and the NE occurs in SPB defective cells. The possible role of the SPB based in INM targeting and maintenance is discussed.

Cell biological approaches, advanced genetics, biophysics and biochemistry have greatly contributed to an increase in our knowledge in this field.lt is obvious however, that the three major membrane constituents - lipids, proteins and carbohydrates- are studied, in most cases separately and that a coherent overview of the various aspects of membrane biogenesis is not readily available. The NATO Advanced Study Institute on "New Perspectives in the Dynamics of Assembly of Biomembranes" intended to provide such an overview: it was set up to teach students and specialists the achievements obtained in the various research areas and to try and integrate the numerous aspects of membrane assembly into a coherent framework. The articles in here reflect this. Statting with detailed contributions on phospholipid structure, dynamics, organization and biogenesis, an up to date overview of the basic, lipidic backbone of biomembranes is given. Extensive progress is made in the research on membrane protein biosynthesis. In particular the post- and co-translational modification processes of proteins, the mechanisms of protein translocation and the sorting mechanisms which are necessary to direct proteins to their final, intra - or extracellular destination have been characterized in detail. Modern genetic approaches were indispensable in this research area: gene cloning, hybrid protein construction, site directed mutagenesis and sequencing techniques elucidated many functional aspects of specific nucleic acid and amino acid sequences.

Book excerpt: The fluid-mosaic model of membrane structure formulated by Singer and Nicolson in the early 1970s has proven to be a durable concept in terms of the principles governing the organization of the constituent lipids and proteins. During the past 30 or so years a great deal of information has accumulated on the composition of various cell membranes and how this is related to the dif ferent functions that membranes perform. Nevertheless, the task of explaining particular functions at the molecular level has been hampered by lack of struc tural detail at the atomic level. The reason for this is primarily the difficulty of crystallizing membrane proteins which require strategies that differ from those used to crystallize soluble proteins. The unique exception is bacteriorhodopsin of the purple membrane of Halobacterium halobium which is interpolated into a membrane that is neither fluid nor in a mosaic configuration. To date only 50 or so membrane proteins have been characterised to atomic resolution by diffraction methods, in contrast to the vast data accumulated on soluble proteins. Another factor that has been difficult to explain is the reason why the lipid compliment of membranes is often extremely complex. Many hundreds of different molecular species of lipid can be identified in some membranes. Remarkably, the particular composition of each membrane appears to be main tained within relatively narrow limits and its identity distinguished from other morphologically-distinct membranes.

Book excerpt: The endoplasmic reticulum is a continuous membrane network in the cytosol, which encloses its internal compartment, the endoplasmic reticulum lumen. Several metabolic pathways are compartmentalised within the ER lumen, for example hydrolysis of glucose 6-phosphate, glucuronidation of endo- xenobiotics, posttranslational modification of proteins including redox reactions required for oxidative folding, oxidoreduction of steroid hormones, synthesis of ascorbate. Therefore, enzyme activities of these pathways depend on the special luminal microenvironment, on access to substrates and on release of products. However, in spite of great efforts, the molecular mechanism for the generation and maintenance of this special microenvironment still remains to be elucidated. Beside the well-known functions of the endoplasmic reticulum, such as calcium signaling and the synthesis of secretory proteins, recent findings underlined the importance of the intraluminal redox biochemistry and the role of membrane transporters. The field is currently undergoing extensive reappraisal, new transporters have been identified either molecular or functional level.The local synthesis and the membrane transport of redox active compounds (pro- and antioxidants) seem to be important not only in the disulfide bond formation, but also in the generation of intracellular proliferative/apoptotic signals. The different points of views in this publication help highlight the potential importance of several recently described phenomena, whose significance needs elucidation.
