Book excerpt: The meeting on "Molecular Mechanisms of Lipid and Protein Traffic", sponsored by NATO Scientific Affairs Division as an Advanced Study Institute and by the Federation of European Biochemical Societies as a Lecture Course was held in Cargese, France, in June 1997. The program included introductory lectures, specialized up-to-date contributions, poster sessions, tutorials and workshops. Emphasis was laid on the new developments in the field of membrane dynamics, in particular on the insertion and translocation of proteins; on intracellular protein, lipid and membrane traffic and sorting and on the mutual interactions between the various events occurring during membrane biogenesis. Mitochondrial biogenesis, chloroplast assembly and the insertion of proteins into peroxisomes were highlighted. Much progress in these research areas has been made in recent years and the ASI provided an excellent opportunity to illustrate this progress in comparison with previous meetings on a similar topic. Not only graduate students and postdocs took advantage from this program but also experienced scientists were given the opportunity to obtain a complete overview of recent progress and the remodeling of ideas and concepts.

Book Crosstalk and Integration of Membrane Trafficking Pathways

Download or read book Crosstalk and Integration of Membrane Trafficking Pathways written by Roberto Weigert and published by BoD – Books on Demand. This book was released on 2012-04-11 with total page 262 pages. Available in PDF, EPUB and Kindle. Book excerpt: Membrane traffic is a broad field that studies the complex exchange of membranes that occurs inside the cell. Protein, lipids and other molecules traffic among intracellular organelles, and are delivered to, or transported from the cell surface by virtue of membranous carriers generally referred as "transport intermediates". These carriers have different shapes and sizes, and their biogenesis, modality of transport, and delivery to the final destination are regulated by a multitude of very complex molecular machineries. A concept that has clearly emerged in the last decade is that each membrane pathway does not represent a close system, but is fully integrated with all the other trafficking pathways. The aim of this book is to provide a general overview of the extent of this crosstalk.

Download or read book Membrane Dynamics and Domains written by Peter J. Quinn and published by Springer Science & Business Media. This book was released on 2013-06-29 with total page 502 pages. Available in PDF, EPUB and Kindle. Book excerpt: The fluid-mosaic model of membrane structure formulated by Singer and Nicolson in the early 1970s has proven to be a durable concept in terms of the principles governing the organization of the constituent lipids and proteins. During the past 30 or so years a great deal of information has accumulated on the composition of various cell membranes and how this is related to the dif ferent functions that membranes perform. Nevertheless, the task of explaining particular functions at the molecular level has been hampered by lack of struc tural detail at the atomic level. The reason for this is primarily the difficulty of crystallizing membrane proteins which require strategies that differ from those used to crystallize soluble proteins. The unique exception is bacteriorhodopsin of the purple membrane of Halobacterium halobium which is interpolated into a membrane that is neither fluid nor in a mosaic configuration. To date only 50 or so membrane proteins have been characterised to atomic resolution by diffraction methods, in contrast to the vast data accumulated on soluble proteins. Another factor that has been difficult to explain is the reason why the lipid compliment of membranes is often extremely complex. Many hundreds of different molecular species of lipid can be identified in some membranes. Remarkably, the particular composition of each membrane appears to be main tained within relatively narrow limits and its identity distinguished from other morphologically-distinct membranes.

Available in PDF, EPUB and Kindle. Book excerpt: Since the discovery of the gene for green fluorescent protein (GFP), derived from jellyfish, this protein that emits a green glow has initiated a revolution in molecular biosciences. With this tool, it is now possible to visualize nearly any protein of interest in any cell or tissue of any species. Since the publication of the first edition, there have been tremendously significant technological advances, including development of new mutant variants. Proteins are now available in yellow and blue, and Novel Fluorescent Proteins (NFPs) have expanded their utility in developing biosensors, biological markers, and other biological applications. This updated, expanded new edition places emphasis on the rise of NFPs, including new chapters on NFP properties with detailed protocols, applications of GFPs and NFPs in industry research, and biosensors. This book provides a solid theoretical framework, along with detailed, practical guidance on use of GFPs and NFPs with discussion of potential pitfalls. The expert contributors provide real examples in showing how to tailor GFP/NFP to specific systems, maximize expression, and enhance detection.

Book Mobility and Proximity in Biological Membranes

Download or read book Mobility and Proximity in Biological Membranes written by S. Damjanovich and published by CRC Press. This book was released on 2018-01-18 with total page 673 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cell surface membranes have long been characterized as two-dimensional fluids whose mobile components are randomized by diffusion in the plane of the membrane bilayer. Recent research has indicated that cell surface membranes are highly organized and ordered and that important functional units of membranes appear as arrays of interacting molecules rather than as single, freely diffusing molecules. Mobility and Proximity in Biological Membranes provides an overview of the results obtained from biophysical methods for probing the organization of cell surface membranes. These results are presented in the context of detailed treatments of the theory and the technical demands of each of the methods. The book describes a versatile and easily applied mode for investigating molecular proximities in plasma membranes in a flow cytometer. Its analysis of lipid fluidity and viscosity of membranes and the rotational mobility of proteins offers intimate insight into the physical chemistry of biological membranes. The electrophysiology of lymphocytes is presented with focus on its importance in different diseases. New techniques are described, and new data, new possibilities, and future trends are presented by world experts. This book's chapters can serve both as guides to the existing literature and as starting points for new experiments and approaches associated with problems in membrane function.

Book Free Energy Calculations

Download or read book Free Energy Calculations written by Christophe Chipot and published by Springer Science & Business Media. This book was released on 2007-01-08 with total page 528 pages. Available in PDF, EPUB and Kindle. Book excerpt: Free energy constitutes the most important thermodynamic quantity to understand how chemical species recognize each other, associate or react. Examples of problems in which knowledge of the underlying free energy behaviour is required, include conformational equilibria and molecular association, partitioning between immiscible liquids, receptor-drug interaction, protein-protein and protein-DNA association, and protein stability. This volume sets out to present a coherent and comprehensive account of the concepts that underlie different approaches devised for the determination of free energies. The reader will gain the necessary insight into the theoretical and computational foundations of the subject and will be presented with relevant applications from molecular-level modelling and simulations of chemical and biological systems. Both formally accurate and approximate methods are covered using both classical and quantum mechanical descriptions. A central theme of the book is that the wide variety of free energy calculation techniques available today can be understood as different implementations of a few basic principles. The book is aimed at a broad readership of graduate students and researchers having a background in chemistry, physics, engineering and physical biology.

Book Membrane Biogenesis

  • Author : Jos A.F. Op den Kamp
  • Publisher : Springer Science & Business Media
  • Release : 2013-06-29
  • ISBN : 3642731848
  • Pages : 474 pages

Download or read book Membrane Biogenesis written by Jos A.F. Op den Kamp and published by Springer Science & Business Media. This book was released on 2013-06-29 with total page 474 pages. Available in PDF, EPUB and Kindle. Book excerpt: Many individual aspects of the dynamics and assembly of biological membranes have been studied in great detail. Cell biological approaches, advanced genetics, biophysics and biochemistry have greatly contributed to an increase in our knowledge in this field.lt is obvious however, that the three major membrane constituents - lipids, proteins and carbohydrates- are studied, in most cases separately and that a coherent overview of the various aspects of membrane biogenesis is not readily available. The NATO Advanced Study Institute on "New Perspectives in the Dynamics of Assembly of Biomembranes" intended to provide such an overview: it was set up to teach students and specialists the achievements obtained in the various research areas and to try and integrate the numerous aspects of membrane assembly into a coherent framework. The articles in here reflect this. Statting with detailed contributions on phospholipid structure, dynamics, organization and biogenesis, an up to date overview of the basic, lipidic backbone of biomembranes is given. Extensive progress is made in the research on membrane protein biosynthesis. In particular the post- and co-translational modification processes of proteins, the mechanisms of protein translocation and the sorting mechanisms which are necessary to direct proteins to their final, intra - or extracellular destination have been characterized in detail. Modern genetic approaches were indispensable in this research area: gene cloning, hybrid protein construction, site directed mutagenesis and sequencing techniques elucidated many functional aspects of specific nucleic acid and amino acid sequences.
