Book excerpt: Oral protein antigen administration induces a state of immunologic tolerance termed oral tolerance. In the first part of the dissertation, we characterized systemic immune tolerance in adoptive transfer recipient (ATR) mice. Adoptive transfer of TCR transgenic cells from DO11.10 mice provides a means for following antigen-specific T cell responses in vivo. In ATR mice, oral protein activated antigen-specific T cells in the gut associated lymphoid tissue (GALT) and periphery. T cell activation accompanied systemic reduction of IL-2 production and selective induction of IFN-gamma in GALT. Oral antigen induced the homing molecules alpha4beta7, alpha4beta1, and P-selectin ligands on Tg + T cells in Peyer's patches and mesenteric lymph nodes. That suggested tolerant T cells migrate outside GALT. Oral tolerance was demonstrated by reduced T cell responses to peptide antigen administration in adjuvant. OVA feeding blunted Tg+ T cell expansion. T cell effector functions (DTH and antibody responses) and cytokine productions (IFN-gamma, IL-4, and TGF-beta) were reduced. To test the importance of IFN-gamma induction by oral antigen, we blocked IFN-gamma by neutralizing antibodies or utilized IFN-gamma-deficient mice. The absence of IFN-gamma did not interfere with blunted T cell expansion, reduction in cytokines, or the tolerance in antibody responses. In contrast, IFN-gamma inhibition abrogated oral antigeninduced reductions in DTH responses. Furthermore, T cell accumulation at cutaneous sites of inflammation was markedly increased in anti-IFN-gamma mAb-treated mice. These observations suggest that the role of IFN-gamma in oral tolerance may be to limit T cell migration to the sites of effector immune responses.

The impact of caffeine consumption on various aspects of health, including cardiovascular disease, reproduction, bone mineral density, and fluid homeostasis are reviewed.
