Book excerpt: Stimulation of brown adipose tissue (BAT) thermogenesis could have far-reaching health benefits in combatting obesity and obesity-related complications. Apolipoprotein A-IV (ApoA-IV), secreted by the brain and small intestine in the presence of dietary lipids, is a well-known short-term satiating protein. We hypothesized that a central administration of ApoA-IV into the third ventricle stimulates BAT thermogenesis through the sympathetic and sensory nerves. The effect of central ApoA-IV on BAT temperature and thermogenesis in mice with unilateral denervation of the intrascapular BAT were monitored using transponder probe implantation, qPCR, and immunoblots. Centrally administered ApoA-IV significantly increased the BAT temperature of WT mice compared to the injection of artificial cerebral spinal fluid (CSF). In addition, ApoA-IV treatment elevated fatty acid oxidation and induced UCP-1-dependent thermogenesis. In the study of the requirement of sympathetic and sensory nerves for ApoA-IV-induced thermogenesis, ApoA-IV elevated BAT temperature and thermogenesis in the innervated BAT. In contrast, ApoA-IV did not elevate BAT thermogenesis in the denervated BAT. There was no difference in plasma lipids and incretins between mice treated with ApoA-IV and CSF. Therefore, central administration of ApoA-IV requires sympathetic and sensory nerves to induce fatty acid oxidation and thermogenesis in BAT.
