Book excerpt: In the Davies group there are three research areas: methodology development, the total synthesis of natural products, and catalyst design & development. Donor/acceptor substituted rhodium carbenoids have been shown by the Davies' group to have exceptional chemoselectivity, regioselectivity, and stereoselectivity in intermolecular cyclopropanation reactions. The first major section of this thesis was devoted to explore the scope and limitations of the acceptor (the electron withdrawing group) part of the donor/acceptor substituted diazo compounds in asymmetric intermolecular cyclopropanation by Rh (II) catalysis. In the context of these projects, Rh 2 (S -PTAD) 4 was found to be an exceptional chiral catalyst for intermolecular cyclopropanation when the acceptor group was either a trifluoromethyl, nitrile, or ketone group. The highly strained cyclopropanes could be synthesized in good yields, with high diastereoselectivity, and with high enantioselectivity, when Rh 2 (S -PTAD) 4 was employed. The second major section (Chapter three) describes a collaborative investigation between the Davies and the Williams groups toward the first asymmetric synthesis of vibsanin E and related natural products. An asymmetric method to the key tricyclic core utilizing the Davies' [4+3] cyclopropanation/Cope methodology, which contains 3 of the 5 continues stereocenters of vibsanin E, was developed utilizing Rh 2 (S -PTAD) 4 . The Williams group then used the enantiomerically pure tricycle to finish the synthesis of vibsanin E and is currently working on several other similar natural products in the vibsanin family. The last major section (Chapter four) of this thesis focuses on catalyst design & development. A new route to Rh 2 (S -PTAD) 4 was developed via a asymmetric Strecker approach, which provided gram quantities of this catalyst. Also within the chapter is described the synthesis of several hydrocarbon-soluble catalysts based on the Rh 2 (S -PTAD) 4 scaffold. These catalysts are fully soluble in hexanes and/or 2,2-dimethylbutane to -78 °C and are active in the decomposition of donor/acceptor diazo compounds.

The Davies group is interested in the development of chiral dirhodium catalysts and their utilization in donor/acceptor diazo decomposition reactions. Rh 2 (S -DOSP) 4 catalyzed intermolecular carbenoid transformations lead to diverse products with excellent selectivity. The first chapter of this thesis explores the limitations of the currently available dirhodium catalysts and the design and synthesis of novel phthalamide-based dirhodium catalysts. Applications of these new catalysts in various carbenoid reactions are discussed. The second chapter focuses on the development of an asymmetric methodology for the synthesis of tropanes by rhodium-catalyzed [4+3] cycloaddition. This methodology has been extended to a range of electronically and structurally diverse pyrroles and furans. This asymmetric tandem cyclopropanation Cope rearrangement was then successfully applied in a key step of the enantioselective synthesis of ( - )-isostemofoline. The third chapter describes the development of asymmetric amination methodology using chiral dirhodium catalysts and their applications in both intramolecular as well as intermolecular amination reactions.

Book excerpt: The development of new methodology for the asymmetric synthesis of chiral organic compounds is a major focus in modem organic chemistry. The use of chiral catalysts is replacing chiral auxiliaries as a new tool for synthetic chemists. An efficient chiral catalyst allows for large quantities of optically active product to be obtained on use of relatively small amount of enantiopure material, without the need for the removal and recovery of a chiral auxiliary. Furthermore, the most practical catalytic methods utilize an inexpensive and readily available chiral ligand that can provide high and predictable enantioselectivity across a wide range of substrates. In our project, two type of versatile, upgraded chiral ligands have been designed and synthesized. Their application in Simmons-Smith type cyclopropanation is investigated, and the pleasing results suggest that they are the potential catalytic enantioselective candidates to build C-C bonds.
