Keeping abreast of the enormous advances in neuroscience in the five years since the first edition was published, the authors have revised all their chapters in the second edition. What was formerly the first chapter has been expanded substantially and divided into two separate chapters to emphasize the cell biology of neurons and glia, and their commonalities with other kinds of cells. The section on intracellular communication has also been expanded and reorganized. Levitan and Kaczmarek introduce the concept of ion channels as specialized membrane proteins at an early stage, making the idea of selective membrane permeability more accessible in terms of the properties of specific ion channel proteins. In addition, they emphasize the astonishing diversity of voltage-dependent ion channels that has become evident in recent years, and discuss the implications of this diversity for neuronal physiology. In the section on intercellular communication, the chapter on neurotransmitter secretion has also been rewritten to reflect the new level of understanding of secretion that has resulted from the identification of many of the molecular players in vesicle fusion and exocytosis. The other chapters in this section have also been fully revised to incorporate new information resulting from the cloning and characterization of the multitude of glutamate receptors as well as to describe novel elements of intracellular signaling pathways in neurons and other cells. Finally, the last section has been substantially updated to reflect the recent successes of molecular studies of development and plasticity. As more and more of the molecular entities that are essential for neuronal development and adult plasticity are identified and characterized, phenomena that previously could be studied only at the descriptive level can now be explained in greater depth.

But what kind of government is it—a dictatorship or a democracy? Nervous systems consist of circuits of interconnected nerve cells (neurons) that transmit and receive information via electrical signals. Every moment, each neuron adds up stimulating and inhibiting inputs from many other neurons to determine whether to send an electrical signal to its recipients. Some circuits are dominated by a single “dictator” neuron that gathers information from many sources and then issues commands, such as the Mauthner neuron that triggers escape in fish. In other more “democratic” circuits, such as those mediating eye movements in monkeys, the outcome is determined by a tally of “votes” from a large population of neurons. Rhythmic movements like breathing and locomotion are generated by “government programs” within the central nervous system, but modified by a soup of chemicals and by free market–like feedback from sensory neurons. Nervous systems also use sophisticated surveillance of the surrounding environment and keep track of their own decisions in order to avoid internal conflicts. Nervous systems are not restricted to using one set of procedures at a time. They have evolved over long periods to control behaviors in whichever ways are most effective, and they essentially combine multiple forms of government simultaneously. Book excerpt: Serotonin signaling is implicated in a variety of psychiatric conditions and regulates behaviors through both synaptic and extra-synaptic neurotransmission. Two serotonin receptors, the 5-HT[subscript]1B and 5-HT6 receptors, are noted for their mediation of extra-synaptic serotonin signaling and unique localization. 5-HT[subscript]1B receptors localize to axon terminals and inhibit the release of neurotransmitters. 5-HT6 receptors localize to neuronal primary cilia where they serve as excitatory receptors and are particularly abundant in the direct and indirect pathway neurons of the striatum. While many aspects of the serotonergic system have been well studied, the ways in which these extra-synaptic serotonin receptors modulate neuronal signaling and circuit-specific behaviors are poorly understood. We first describe the creation of neuronal cell lines with stable expression of 5-HT[subscript]1B receptors and loss of either isoform of [Beta]-arrestin in order to elucidate the intracellular signaling mechanisms of 5-HT1B receptors. We next use these cell lines to show that 5-HT[subscript]1B receptors activate ERK1/2 signaling in a G[alpha][subscript]s- and [Beta]-arrestin-dependent manner. Finally, we demonstrate that 5-HT6 receptors modulate cocaine reinforcement in a pathway-specific manner; specifically, increased 5-HT6 receptor signaling in the indirect pathway of the nucleus accumbens reduces cocaine taking by increasing the reinforcing properties of the drug. Together, these findings increase our understanding of extra-synaptic serotonin receptors and the mechanisms by which they influence intracellular signaling cascades and circuit-specific behaviors.
