Book excerpt: There has been a convergence in recent years of people from the physical and biological sciences and from various engineering disciplines who are interested in analyzing the electrical activity of nerve and muscle quantita tively. Various courses have been established at the graduate level or final-year undergraduate level in many universities to teach this subject matter, yet no satisfactory short text has existed. The present book is an attempt to fill this gap, and arises from my experience in teaching this material over the past fifteen years to students on both sides of the Atlantic. Although covering a wide range of biophysi cal topics from the level of single molecules to that of complex systems, I have attempted to keep the text relatively short by considering only examples of the most general interest. Problems are included whenever possible at the end of each chapter so the reader may test his understand ing of the material presented and consider other examples which have not been included in the text.

Not only the fundamentals of the modern membrane theory of biopotentials, but also the vast factual material collected in the last decades by the study of the biophysical and pharmacological properties of the ionic permeability pores of the cell membrane, are described in the book in an authoritative yet readable form. Book excerpt: One of the most active and productive areas of biological science in the past decade has been the study of the biochemical and biophysical prop erties of cell membranes. There is little doubt that membranes are essen tial components of all cellular systems and that each type of membrane manifests specific and characteristic cellular functions. In the nervous system, important events such as neurotransmission, receptor binding, ion transport, axonal transport, and cell uptake are all known to take place within the neural cell membrane. Phospholipids, one of the major components of membranes, not only provide the membrane with its structural integrity and physical proper ties, but also play an important role in regulating membrane function. Attention has recently been focused on the asymmetric localization of these molecules, the identification of discrete metabolic pools of phospholipids within the membrane matrix, and their involvement in sig nal transmission. Although synaptic membranes generally lack an active mechanism for the de novo biosynthesis of phospholipids, a number of enzymic routes are present for their interconversions and for facilitating metabolic turnover. Metabolites generated during the interconversion reactions may also exert a great influence in modulating membrane func tions. The phosphogylcerides of neural membranes are especially enriched in polyunsaturated fatty acids. However, only very small amounts of these fatty acids are present in the free form, and they are maintained in dynamic equilibrium with the membrane phospholipids.

Book excerpt: This is a book about how Cl- crosses the cell membranes of nerve, muscle, and glial cells. Not so very many years ago, a pamphlet rather than book might have resulted from such an endeavor! One might ask why Cl-, the most abundant biological anion, attracted so little attention from investigators. The main reason was that the prevailing paradigm for cellular ion homeostasis in the 1950s and 1960s assigned Cl- a ther modynamically passive and unspecialized role. This view was particularly prominent among muscle and neuroscience investigators. In searching for reasons for such a negative (no pun intended) viewpoint, it seems to us that it stemmed from two key experimental observations. First, work on frog skeletal muscle showed that Cl- was passively distributed between the cytoplasm and the extracellular fluid. Second, work on Cl- transport in red blood cells confirmed that the Cl- transmembrane distribution was thermodynamically passive and, in addition, showed that Cl- crossed the mem brane extremely rapidly. This latter finding [for a long time interpreted as being the result of a high passive chloride electrical permeability(? CI)] made it quite likely that Cl- would remain at thermodynamic equilibrium. These two observations were gener alized and virtually all cells were thought to have a very high P Cl and a ther modynamically passive Cl- transmembrane distribution. These concepts can still be found in some physiology and neuroscience textbooks.

By emphasizing the experimental approach, it illustrates the tools that have re cently become available for investigating the nervous system. Also, perhaps even more than other volumes, it illustrates the multidisciplinary nature of the field, requiring multidisciplinary methodology. It is now recognized that the availability of methodology is often the rate-limiting determinant of studies and that improvements or innovations in instrumentation can open up new avenues. A new improved method, although opening up new possibilities and being crucial to making advances, is only a tool whose use will determine its use fulness. If we do not recognize its possibilities, its use will be limited; if we do not recognize its limitations, it will mislead us. It is the possibilities and limitations and the results obtained that are illustrated here.
