Book Growth Factors and Alzheimer s Disease

Download or read book Growth Factors and Alzheimer s Disease written by Franz Hefti and published by Springer Science & Business Media. This book was released on 2013-03-08 with total page 259 pages. Available in PDF, EPUB and Kindle. Book excerpt: The study of the relationships between neurotrophic factors (NGF, FGF, amyloid) and neurodegenerative disorders (especially Alzheimer's disease) is of major importance not only for understanding the pathogenesis of the latter, but also for the pharmacological approach to this disease. Information on the subject stemming from the symposium in Strasbourg in April, 1990, organized by the Fondation IPSEN is presented in this book. It provides stimulating hypotheses about the possible role of growth factors in the generation of senile plaques, the process of degeneration and regeneration in Alzheimer's disease, the expression of the gene of NGF and the possible use of NGF in treatment.

Book excerpt: A distinguished panel of internationally recognized neuroscientists comprehensively review the involvement of and changes in glial cells both during the normal aging process and in the major disorders of old age. Topics range from the cellular and molecular changes that occur with aging-especially aging-associated activation of astrocytes and microglia and its relation to neuronal injury and repair-to neuron-glia intercommunication. The contributors show how glial signals may be modulated by hormones, growth factors, neurotransmitters, intracellular metabolism, and intercellular exchanges, as well as by aging of the blood-brain barrier.

Available in PDF, EPUB and Kindle. Book excerpt: Alzheimer's disease is one the foremost health problems facing every society fortunate enough to attain a level of medical care that ensures an average lifespan of over 70 years of age. The cause of the disease remains unknown, and no single therapeutic approach has yet been found highly efficacious. Indeed, as the complexity of its effects on brain neuronal systems becomes elucidated, the concept emerges that Alzheimer's disease may be an umbrella term for multiple Alzheimer's-type syndromes that can be differentiated based on etiology (hereditary versus sporadic), neurochemical deficits, and extent of pathology. Whether one or more disease processes is involved, it seems increasingly unlikely that any single drug will provide efficacious relief across the broad spectrum of symptoms reflected in large subject populations. Rational therapeutic approaches may yield long term amelioration of many symptoms in selected groups of patients; and individual symptoms may be ameliorated in larger populations. Multiple approaches may be combined to optimize therapeutic effects. For example, a variety of approaches remain focused on restoration of cholinergic neurodegeneration and enhancing cholinergic neurotransmission, whereas other approaches attempt to retard the of the brain that occurs during Alzheimer's disease. Chapters cover continuous degeneration topics from growth factor secretion of proteases, which could modify amyloid processing, to neuronal gene delivery using viral vectors. The breadth and thoroughness of the chapters truly embrace a broad spectrurn of models, pathological mechanisms and therapeutic approaches.

Book Nerve Growth Factor Metabolism in the Continuum of Alzheimer s Disease Pathology

Download or read book Nerve Growth Factor Metabolism in the Continuum of Alzheimer s Disease Pathology written by Rowan Pentz and published by . This book was released on 2021 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: "As the Canadian and world populations age, the deaths and years of lost life attributable to age-related neurodegenerative illnesses become ever more significant. There are currently no disease-modifying treatments for Alzheimer’s disease (AD), the most common such disease. Nor can AD be unquestionably detected during the long asymptomatic phase in which AD neuropathology evolves prior to the onset of cognitive symptoms. The presentation and incidence of AD is affected by sex, which remains an understudied aspect of AD pathology.Alzheimer’s disease entails a marked and early loss of basal forebrain cholinergic neurons, which contributes to cognitive decline at the earliest stages of the disease. Insight into the causes of this degeneration has come from the discovery of a metabolic pathway controlling the availability of Nerve Growth Factor (NGF) to the basal forebrain cholinergic neurons that depend on it. Here, we thoroughly and systematically analyze the NGF metabolic pathway throughout the continuum of AD pathology. We demonstrate that deficits in the maturation of proNGF to mature (m)NGF are evident in AD, Mild Cognitive Impairment (MCI), and prior to MCI in frontal cortex tissue from individuals with high brain amyloid but no clinically relevant cognitive impairment (classified as High Amyloid No Cognitive Impairment: HA-NCI). At this stage, these deficits correlate with cognitive status (perhaps subtle preclinical cognitive decline) and the deposition of amyloid/tau. We further demonstrate with in vitro and in vivo experiments that Matrix Metalloproteinase 3 (MMP-3) is a new member of the NGF metabolic pathway, contributing to the degradation of mature NGF, and released from cortical tissue in response to cholinergic stimulation. MMP-3 is also elevated in individuals designated AD, MCI, or HA-NCI and correlates with cognitive status. MMP-3 also shows a sex-specific expression and association with AD pathology; males exhibit more MMP-3 and in males, but not females, MMP-3 levels correlate with cognitive status and to AD neuropathology. We further demonstrate that the degree of the NGF metabolic compromise correlates with the status of the central cholinergic system. We also investigated the ability of NGF pathway protein in blood/cerebrospinal fluid (CSF) to serve as biomarkers of the AD pathological process in Down syndrome (DS), a condition characterized by a progressive and lifelong amyloidosis that culminates in AD by middle age. Distinct profiles of NGF pathway proteins in blood and especially in CSF distinguished DS, DS with AD, and presumed normal karyotypical controls from one another. CSF ProNGF and MMP-9 identified AD from the general DS population with good efficacy, approaching that of existing best practises. Alterations in the levels of NGF pathway proteins consistently correlated with core biomarkers of AD, and both MMP-3 and MMP-9 were differentially expressed by sex.We conclude that the NGF metabolic pathway is dysregulated over the continuum of AD, which contributes to early cognitive and cholinergic deficits. This pathway is therefore a promising potential source of biomarkers and of therapeutic targets in AD"--

Book Novel Approaches to the Treatment of Alzheimer s Disease

Download or read book Novel Approaches to the Treatment of Alzheimer s Disease written by E. Meyer and published by Springer Science & Business Media. This book was released on 2013-03-09 with total page 387 pages. Available in PDF, EPUB and Kindle. Book excerpt: Alzheimer's disease afflicts up to 1 in 5 people over the age of 65 years and causes untold suffering of the patient and their family. The cause of this disease is unknown; indeed, evidence increasingly suggests that there may be multiple Alzheimer-type syndromes with different etiologies, analogous to different types of psychosis. Currently there are no means to prevent the disease, slow its progress or reverse its neurodegenerative consequences. With few exceptions, clinical trials of a variety of compounds have resulted in patient responses that are disappointing with respect to both the proportion of responders and the magnitude of the responses. Novel approaches to the treatment of Alzheimer's disease are clearly warranted. For this reason, we organized the First Suncoast Workshop on the Neurobiology of Aging in St. Petersburg, Florida, which took place from February 26-March 1, 1989. This workshop focused on novel treatments and models for Alzheimer's disease and represented a cooperative venture among academia, government and industry, both in its participants and sponsorship. The Center for the Neurobiology of Aging at the University of Florida, the National Institute on Aging and Taiho Pharmaceutical Corporation in Japan sponsored the workshop in which scientists from the North America, Europe, Japan and other parts of Asia participated.

Book Neurobiology of Alzheimer s Disease

Download or read book Neurobiology of Alzheimer s Disease written by David Dawbarn and published by Oxford University Press. This book was released on 2007-05-03 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Alzheimer's disease is the most common form of dementia in the elderly; 450,000 people in the UK and 4.5 million people in the USA suffer with this disease. This 3rd edition of Neurobiology of Alzheimer's Disease gives a comprehensive and readable introduction to the disease, from molecular pathology to clinical practice. The book is intended for readers new to the field, and it also covers an extensive range of themes for those with in-depth knowledge of Alzheimer's disease. It will therefore act either as an introduction to the whole field of neurodegeneration or it will help experienced researchers to access the latest research in specialist topics. Each chapter is written by eminent scientists leading their fields in neuropathology, clinical practice and molecular neurobiology; appendices detail disease-associated proteins, their sequences, familial mutations and known structures. It will be essential reading for students interested in neurodegeneration and for researchers and clinicians, giving a coherent and cohesive approach to the whole area of research, and allowing access at different levels. For those in the pharmaceutical industry it describes the underlying molecular mechanisms involved in the pathogenesis of Alzheimer's disease and explains how current and potential therapeutics may work.

Book Understanding Alzheimer s Disease

Download or read book Understanding Alzheimer s Disease written by Neal R. Cutler and published by Univ. Press of Mississippi. This book was released on 2010-02-11 with total page 168 pages. Available in PDF, EPUB and Kindle. Book excerpt: Alzheimer's disease has received growing attention in recent years because the affected population is growing quickly as life spans in America creep ever upward. As they near retirement, many Americans start to fear that any minor memory lapse-misplacing the keys or forgetting a name-might spell Alzheimer's. In reality such incidents are a normal part of the aging process. In Alzheimer's disease, such incidents progress to a point at which they interfere with normal daily activities on a regular basis, and memory loss becomes progressively more disabling. This book is written for anyone interested in learning more about Alzheimer's disease-people who have it, their families and caregivers, professionals who serve such people, teachers, and anyone with a general interest in the disease. An important feature of this book is clear explanation of the basic biology of the disease. Here is information about who gets the disease, how it is diagnosed, and what happens to people who have it. A large section is devoted to the subject of caring for affected individuals, including legal and financial planning advice and pointers for finding outside support services. Existing treatments are discussed along with suggestions for handling many symptoms without drugs. Understanding Alzheimer's Disease describes the most current knowledge of how and why it arises and gives information about the many avenues being explored in search of a cure. With its chapters on causes and on the search for treatments, this guide will be of interest to a wider section of the population than other books on the disease.
