Despite this fact, the organizers of the 3 rd International Conference on Differentiation recognized that there is much emerging evidence which supports the view that both normal cells and many cancer cells share common differentiative processes. Accordingly, the organizers perceived that clinical scientists and developmental biologists would greatly benefit by together considering differentiation. In that way, developmental biologists would be apprised of recent insights in cancer cell biology and the physician scientist would be updated on events in developmental biology and both would gain new understanding of the cell biology of neoplasia. A specific example may reveal the potential value of developmental biologists interacting with cancer physicians. An example chosen at random suggests that probably any paper included in the symposium volume would serve the purpose. Dr. Stephen Subtelny reviewed recent studies by his laboratory concerning germ cell migration and replication in frog embryos. How might those results interest the cancer scientist? Dr. Subtelny showed that primordial germ cells of a fertile graft will reverse their migratory direction and move into a sterile host. Perhaps in this context it would not be inappropriate to state that the germ cells of the graft metastasized into the host. Germ cells from grafts of a different species will populate the previously sterile host gonad.

One paper presents the techniques used in the study on the proliferation kinetics of hemopoietic stem cells, suggesting that the hemopoietic stem cell population is not homogenous but consists of a "primitive pluripotential stem cell." A series of experiments at the Brookhaven National Laboratory investigates the relationship of cell survival, specifically that of stem cells, to the survival of the irradiated test animal. One result of the experiment shows a rapid migration of a number of stem cells from shielded marrow into unshielded marrow at the pressure of a rapid circulating pool. The numbers of stem cells are somewhat dependent on the dose given to the unshielded marrow, and are greater with the greater dose. Another paper also investigates the four methods that are used in the study of cellular kinetics in human tumors. This compendium can prove helpful for biochemists, micro-biologists, cellular researchers, and academicians involved in the study of cellular biology, physiology or oncology.

The principles as well as recent developments in tumor biology were discussed in depth, with emphasis on the regulation of the cell cycle, differentiation, programmed cell death (apoptosis) and genetics of cancer. This book constitutes the proceedings of that meeting. Specifically, the following areas were addressed: (a) enzymes and proteins (cyclins) that control the cell cycle, as well as the role of m as gene in meiosis and transformation; (b) the structural basis for specificity in protein-tyrosine kinase reactions; (c) the differentiation of normal as well as neoplastic cells with respect to molecular mechanism(s) by which chemical agents or growth factors trigger maturation; (d) phenotypic and genetic aspects of apoptosis; (e) the role of growth factors, like IGF-l, FGF, TN, IL-6, etc. , in cell cycle regulation, apoptosis (cell death) and senescence; (f) molecular mechanisms of transcriptional activation of globin genes and stability of mRNAs related to growth proteins and iron metabolism; (g) the cellular and molecular biology of bone marrow hemopoiesis; and (h) neurotrophic factors and the generation of cellular diversity in the central nervous system. It was obvious from the studies presented that neoplastic cell growth, differentiation and apoptosis in many cell types are regulated at several levels.

All adult tissues are made up of lineages of cells consisting of tissue stem cells and their progeny (transit-amplifying cells and terminally differentiated cells). Recent data suggest that cancers arise from rare self-renewing stem cells that are biologically distinct from their more numerous differentiated progeny. Growing evidence suggests that pathways regulating normal stem cell self-renewal and differentiation are also present in cancer cells and cancer stem cells (CSCs). Malignant tumours can be viewed as an abnormal organ in which a small population of tumorigenic CSCs have escaped the normal limits of self-renewal giving rise to abnormally differentiated cancer cells that contribute to tumour progression and growth. This new model has important implications for the study and treatment of cancer.
