The whole concept was formulated one early Wednesday morning at our weekly research meeting at Children's Hospital in our division of urology. We have been most fortunate to have a close collaboration with Bob Levin, Ed Macarak, and Pam Howard who have helped steer the course of our division's growing interest in basic science. At our weekly meetings our laboratory fellow will summarize their current work. Other ongoing areas of investigation in our labs and elsewhere are discussed. We have always made an effort to try and understand what other groups are doing who are working in the area of bladder smooth muscle research. It occurred to us that the best way to really know what everyone working in this field was doing would be to sponsor a 2-day meeting where we could all gather to discuss our ongoing work. A major limitation of the annual meeting of the American Urologic Association or the urology section of the American Academy of Pediatrics is that the scientfic sessions are limited as these are meant to be primarily clinical meetings (as they should be). For this reason the idea of a meeting devoted solely to research about the urinary bladder had great appeal. In addition to allowing for longer presentations than the standard 5 to 7 minutes, every effort would be made to encourage a dialogue amongst the presenters and the audience.

It is important for such investigators to continue to meet regularly for the purpose of discussing the latest developments in their individual fields, to analyze the significance of current research, to discuss new tactics for unresolved problems, to critically evaluate current theories, and to develop new theories and approaches as needed. The two and a half day meeting was organized into five half day sessions, with each session encompassing one of five topics: (1) Epithelial-Mesenchymal Interactions; (2) Ex tracellular Matrix and Muscle; (3) Nerves and Pharmacology; (4) Infection and Immunol ogy; and (5) Oncology. Each session was introduced by a moderator followed by five to six invited expert speakers with time for extensive interaction from the participants. Two late-afternoon poster sessions allowed further interactions between investigators. This book documents the proceedings of the Bladder Research Congress. It is organ ized into the five half-day sessions of the meeting with moderators overview and an edited transcription of discussions that followed each presentation. While recent years have brought significant progress in regenerative medicine and tissue engineering, the immense challenges encountered when trying to engineer an entire organ have to be acknowledged. Only a step-by-step approach will allow the future production of a living tissue construct ready for implantation and to augment organ function. Book excerpt: The origins of what have come to be known as the "Oxford" Conferences on modelling and the control of breathing can be traced back to a discussion between Dan Cunningham and Richard Hercynski at a conference dinner at the Polish Academy of Sciences in 1971. Each felt that they had benefited from the different perspectives from which the topic of ventilatory control was approached - predominantly physiological in the case of Dr Cunningham and predominantly mathematical in the case of Dr Hercynski. Their judgement at that time was that a conference on the control of breathing which allowed investigators with these different (but related) scientific perspectives to present and discuss their work, might prove fruitful. We would judge that this has amply been borne out, based upon the success of the series of conferences which resulted from that seminal dinner conversation. The first conference, entitled "Modelling of a Biological Control System: The Regulation of Breathing" was held in Oxford, UK, in 1978. Subsequent conferences were: "Modelling and the Control of Breathing" at Lake Arrowhead, California, in 1982; "Con cepts and Formulations in the Control of Breathing" in Solignac, France, in 1985; "Respi ratory Control: A Modeling Perspective" at Grand Lakes, Colorado, in 1988; and "Control of Breathing and Its Modelling Persepctive" at the Fuji Institute in Japan in 1991. The conferences, subsequent to the one in Oxford, have all resulted in well-received published proceedings.

Book excerpt: Much of organic chemistry is based on the ability of suitably structured chemicals to bind together through the formation of covalent bonds. Biochemistry is replete with exam ples of enzymatically catalyzed reactions in which normal body constituents can be linked through covalent bonds during the process of intermediary metabolism. The finding that xenobiotic chemicals that enter the body from the environment, are metabolized to highly reactive species, and then covalently react with cellular macromolecules to induce toxic and carcinogenic effects was an observation that spawned the research featured in the Fifth International Symposium on Biological Reactive Intermediates (BRI V). The group of investigators that became fascinated with this process and its signifi cance in terms of human health began their discussions in Turku, Finland (J 975), and continued them at Guildford, England (1980), College Park, Maryland (1985), Tucson, Arizona (1990), and Munich, Germany (1995). In the ancient Middle East, blood and life were coupled. Blood was the primary substance necessary for life, given to God in sacrifice and forbidden as a food to mortals by Levitical law. Blood was essential for rites of purification and consecration. But the flow of blood during menstruation or parturition rendered a woman unclean. The circumcision of a male child required 33 days of "blood purification" by the mother. ' Circumcision, the visible reminder of the covenant of Abraham lijith Yahweh, was required of newborn Jewish males. It "connote(d) suitability for participation in what God is doing. "2 Hence, free and uncontrolled bleeding of the male child during circumcision, during the ratification of God's covenant, would be noted with awe and concern by those of the Jewish faith. It should not be surprising that the first genetic counseling offered to families with hemophilia is found in the Babylonian Talmud (compilation of Jewish law dated to about the third century AD) and concerns the necessity for circumcision in families with what we would now call hemophilia.

T. ) of which this volume is the scientific proceedings, was held in Istanbul, Turkey on August 22-26, 1994. It was a historical occasion in that it was almost 200 years to the day that one of the founding fathers of oxygen research, Antoine Lavoisier, on May 8, 1794 found his early demise at the hands of the guillotine. This spirit of history set the tone of the conference and in the opening lecture the contribution that this part of the world has given to the understanding of oxygen transport to tissue was highlighted. In particular, the contribution of Galen of Pergamon (129-200) was discussed who for the first time demon strated that blood flowed through the arteries and whose view on the physiology of the circulation dominated the ancient world for well over a millennium. A forgotten chapter in the history of the circulation of the blood is the contribution made by Ibn al Nafis of Damascus (1210-1280) who for the first time described the importance of the pulmonary circulation by stating that all venous blood entering the right ventricle ofthe heart passes to the left ventricle, not through pores in the septum of the heart as had been postulated by Galen, but through the circulation of the lungs.

The fumonisins became the spotlight of mycotoxin research in 1988, when re searchers at the South African Medical Research Council isolated and structurally charac terized the fumonisins. Since 1988, there has been an explosion in the numbers of papers dealing with fumonisin-related topics. The interest in the fumonisins has arisen for several reasons. First, fumonisins are found in measurable concentrations in corn grown throughout the world. Second, these compounds have been implicated as the causative agents in a variety of naturally occurring animal diseases. Finally, there is speCUlation that fumonisins may in part be responsible for the high incidence of esophageal cancer in regions of the world in which corn is the staple grain.

ICOP meetings have been held biennially in the USA, Europe, and Japan in turn. The previous three ICOP meetings (7th to 9th) were held in Shimoda, Japan, in 1988. in WildbadKreuth, Germany, in 1990, and in Williamsburg. Virginia, in 1992. Previous meetings were held in resort areas, this was the first meeting held in a large city. Attendance has grown every year so that nearly 400 participants from 19 different countries attended the Tokyo meeting. At the meeting, novel and updated results on the structure-function. physiology, biology, and pathology of proteases and inhibitors were discussed, together with cellular aspects of proteolysis and protein turnover. Thirty-nine invited papers and eight selected posters were presented orally and 171 poster presentations were discussed. This book documents almost all of the lectures and some selected posters. Since the world of proteolysis and protein turnover is expanding very rapidly. far beyond our expectations, it is impossible to cover all the new aspects of this field. However, this book will give an idea of the current status, trends. and directions of the field, and information necessary to understand what is and will be important in this field. Further. the editors hope that the novel ideas, approaches. methodologies, and important findings described in this book will stimulate further study on proteolysis and protein turnover.

Book excerpt: Development and Implications of Antimicrobial Resistance One of the most ominous trends in the field of antimicrobial chemotherapy over the past decade has been the increasing pace of development of antimicrobial resistance among microbial pathogens. The hypothesis that man can discover a magic bullet to always cure a particular infection has proved false. Physicians are now seeing and treating patients for which there are few therapeutic alternatives, and in some cases, none at all. Until recently there was little concern that physicians might be losing the war in our ability to compete with the evolving resistance patterns of microbial pathogens. Now the general public is very aware of the threat to them if they become infected, thanks to cover story articles in major magazines such as Time, Newsweek, newspapers, and other news sources. Antimicrobial resistance is not a novel problem. Shortly after the widespread introduction of penicillin in the early 1940s, the first strains of penicillin-resistant staphylococci were described. Today it is an uncommon event for a clinical laboratory to isolate an S. aureus that is sensitive to penicillin. Other gram-positive strains of bacteria have become resistant, including the exquisitely sensitive Streptococcus pneumoniae. Sensitivity to vancomycin was once so uniform that it was used in routine clinical laboratories as a surrogate marker for whether an organism should be classified as a gram-positive. That criterion can no longer be relied upon because of emerging resistance among some species. Gram-negative bacteria, viruses, fungi, and parasites all have succeeded in developing resistance.
