Book Multienzyme Assemblies and Dynamics in Acetogenesis and Methanogenesis

Download or read book Multienzyme Assemblies and Dynamics in Acetogenesis and Methanogenesis written by Steven E. Cohen (Ph. D.) and published by . This book was released on 2020 with total page 148 pages. Available in PDF, EPUB and Kindle. Book excerpt: The Wood-Ljungdahl pathway of acetogenesis allows for growth utilizing carbon dioxide as the sole carbon source. This process uses a series of metalloenzymes to catalyze the reduction of two molecules of carbon dioxide to acetyl-CoA. Key to this pathway are the bifunctional carbon monoxide dehydrogenase/acetyl-CoA synthase (CODH/ACS) and corrinoid iron-sulfur protein (CFeSP). CODH/ACS is a bifunctional enzyme, with the CODH subunit catalyzing the reduction of carbon dioxide to carbon monoxide. CFeSP is a cobalt-containing corrinoid-dependent methyltransferase. The active site of ACS, termed the A-cluster, is a nickel, iron, and sulfur-containing metallocluster which catalyzes the synthesis of acetyl-CoA from carbon monoxide provided by CODH, a methyl group provided by CFeSP, and CoA. Despite the unprecedented organometallic chemistry performed at the A-cluster much is unknown about this ACS activity. No substrate-bound structures of ACS have ever been reported and the molecular basis for ACS and CFeSP interactions are largely uncharacterized. Here we present the structure of the carbonylated A-cluster in CODH/ACS, highlighting the role of conformational dynamics in catalysis. We further characterize ACS dynamics using negative-stain electron microscopy, demonstrating a much larger extent of conformational flexibility than has been crystallographically observed. The basis for CFeSP:ACS interactions is further interrogated using a series of CFeSP domain constructs, showing the corrinoidbinding Rossmann fold to be sufficient for ACS interactions. Finally, we report preliminary structural characterization of acetyl-CoA decarbonylase/synthase (ACDS), a 2.2 MDa complex of CODH, ACS, and CFeSP found in archaeal methanogens. Together, this advances our understanding of the roles of structure and dynamics in the Wood-Ljungdahl pathway.

Book Organized Multienzyme Systems Catalytic Properties

Download or read book Organized Multienzyme Systems Catalytic Properties written by G. Rickey Welch and published by Elsevier. This book was released on 2012-12-02 with total page 473 pages. Available in PDF, EPUB and Kindle. Book excerpt: Organized Multienzyme Systems: Catalytic Properties describes the kinetic and catalytic properties of organized enzyme systems. This book is composed of nine chapters that specifically cover both immobilized and naturally occurring systems. The first two chapters examine the nature and function of enzyme organization in the mitochondrion, as well as the structural/functional coupling of the components in energy-transducing membrane systems. These topics are followed by discussions on ""dynamic compartmentation"" in soluble multienzyme systems; the allosteric enzyme systems; and allosterism in reversibly adsorptive enzyme systems. Other chapters explore model studies with specific immobilized multienzyme sequences, as regards the analysis of microenvironmental effects, and the mathematical exposition on the kinetic analysis of multienzyme systems in homogeneous solution. The last chapters present some theoretical and experimental studies on the behavior of immobilized systems. These chapters also provide a speculative integrative view of the kind of functional coherence that may be operative in organized states in vivo. This book is of great value to cell biologists, biochemists, and enzyme scientists and researchers.

Book Protein Folding

    Book Details:
  • Author : Charis Ghelis
  • Publisher : Academic Press
  • Release : 2012-12-02
  • ISBN : 0323140920
  • Pages : 580 pages

Download or read book Protein Folding written by Charis Ghelis and published by Academic Press. This book was released on 2012-12-02 with total page 580 pages. Available in PDF, EPUB and Kindle. Book excerpt: Protein Folding aims to collect the most important information in the field of protein folding and probes the main principles that govern formation of the three-dimensional structure of a protein from a nascent polypeptide chain, as well as how the functional properties appear. This text is organized into three sections and consists of 15 chapters. After an introductory chapter where the main problems of protein folding are considered at the cellular level in the context of protein biosynthesis, the discussion turns to the conformation of native globular proteins. Definitions and rules of nomenclature are given, including the structural organization of globular proteins deduced from X-ray crystallographic data. Folding mechanisms are tentatively deduced from the observation of invariants in the architecture of folded proteins. The next chapters focus on the energetics of protein conformation and structure, indicating the principles of thermodynamic stability of the native structure, along with theoretical computation studies of protein folding, structure prediction, and folding simulation. The reader is also introduced to various experimental approaches; the reversibility of the unfolding-folding process; equilibrium and kinetic studies; and detection and characterization of intermediates in protein folding. This text concludes with a chapter dealing with problems specific to oligomeric proteins. This book is intended for research scientists, specialists, biochemists, and students of biochemistry and biology.
