They allow novel strategies for improving crop productivity and quality, especially in the agrofood sector. The molecular mechanisms underlying these biotechnological approaches are presented here. Topics included are: pollen development, pollen tube growth, macrosporogenesis and fertilization and the effects of pesticides on sexual plant reproduction. Fertilization in higher plants is a complex process consisting of two events, the fusion of the egg with one sperm cell resulting in the diploid zygote, and the fusion of embryosac nuclei with another sperm cell, leading to a triploid endosperm. This "double fertilization" is preceded by the pollination process and a long lasting interaction between the dipoid pistil and the haploid pollen tube (progamic phase). Fertilization of flowering plants results in the formation of seeds and fruits, our basic food supply.

Critical to their reproductive success has been the evolution of mechanisms to ensure proper timing of flowering. Plants cue flowering as they age, and further coordinate with the sensing of environmental signals such as changing daylengths and seasonal temperatures. The molecular genetics of flowering in the model Arabidopsis thaliana are well characterized, however, relatively little is known in other species. Study of diverse lineages suggests mechanisms regulating flowering time have evolved independently. A conserved feature is the importance of chromatin in regulating flowering time genes. We screened a library of Arabidopsis histone deacetylase mutations and found that HDA9 forms a conserved complex with PWR and HOS15. HDA9-PWR-HOS15 represses flowering through the autonomous pathway. We provide a literature review of the involvement of the Arabidopsis HDAC superfamily in flowering. To overcome redundancy, we created and characterized higher order mutants of the Class I HDAC mutants (hda9/6/19), allowing us to more precisely define their roles in flowering. To explore flowering in other plant lineages, we are studying the temperate grass Brachypodium distachyon. To develop Brachypodium as a model organism, we conducted a study to determine optimal light conditions for indoor growth. We settled on an LED source that results in favorable traits such as compactness and shortened generation time. Finally, we provide an update to discuss how forward and reverse genetics has led to progress in understanding Brachypodium's flowering pathway. Future studies will continue to characterize genes to better understand the molecular basis of vernalization.

A key developmental transition strongly influenced by environmental cues is the change from vegetative to reproductive growth. For example, plants adapted to temperate climates must avoid flowering in the winter when environmental conditions are significantly less favorable for reproductive success than spring and summer. One adaptation seen in a diverse range of flowering plants is the vernalization requirement, the process by which prolonged cold exposure results in the competence to flower. The majority of the molecular characterization of this response has been done in the eudicot Arabidopsis thaliana. Recent studies have shown that vernalization likely evolved independently in different plant lineages thus preventing the transposition of the molecular knowledge acquired in A. thaliana to other families of plants. To build upon our understanding of flowering in temperate grasses, we performed genetic screens in Brachypodium distachyon for mutants that flower early without vernalization. By screening for early flowering mutants, we were able to identify genes that affect flowering either through specific processes or through broad regulatory functions. From these screens we identified BdEZL1, the catalytic subunit of Polycomb repressive complex 2 (PRC2). This complex inhibits flowering through the transcriptional repression of a large number of genes, including several flowering regulators. We also isolated a mutant in a novel gene that appears to function specifically in the regulation of flowering by modulating the vernalization response.
