Trans plantation antigens were discovered and characterized by Gorer about 50 years ago, and the biological basis for the unequalled complexity of their variability between individuals within a species, in spite of extreme conservation between species, was the subject of intense research and discussion for many years. During the days of belief in "immune surveillance" against spontaneously developing tumors, it was suggested that histoincompatibility between members of one species would prevent cancer from being a contagious disease and thus a threat to the species. Immunologists involved in human transplantation had to learn and care about the complexity, especially after 1967, when it was found that HLA antigens were the products of the human MHC. Rejection of HLA-identical sib kidney grafts was so rare, even in those days, that cases of rejection were described in scientific papers.

Humans and Old and New World non human primate species, from the lemurs of Madagascar to the African anthro poid apes, represent diverging branches of an evolutionary common trunk. Along with species-specific characters, all primates have retained a number of ancestral traits, relics of their common origin. The comparative study of these species-specific and ancestral traits makes it possible to reconstruct the evolu tionary pathways of humans and nonhuman primates. The discovery of the human blood groups and, later, of the Major Histocom patibility Complex (MHC) had a seminal effect on the field of human genetics, providing the first sound examples of mendel ian polymorphisms. The use of blood group and MHC alleles as genetic markers in biological anthropology gen erated a conceptual revolution and persuaded researchers to begin to think in terms of populations and not only intems of typology. The counterparts of these human red and white cell antigens were found and studied in nunhuman primates, and progress in this field is summarized in this book.

Among the advances has been the initial characterization of the structure of some HLA loci. While this will ultimately improve clinical services, constant reference to serologic data is essential so that the powerful new techniques can be applied in the most effective ways. The human MHC encompasses approximately 4 million base pairs on the short arm of chromosome 6 at cytogenetic location 6p21. 3. The region is divided into three subregions. The telomeric class I region contains the genes that encode the HLA class I molecules HLA-A, -B, and -C. The centromeric class II region contains the genes encoding the HLA class II molecules HLA-DR, -DQ, and -DP. In between is the class III region, originally identified because it contains genes encoding components of the complement pathway. The entire human MHC has recently been sequenced (1) and each subregion is now known to contain many other genes, a number of which have immunological functions. The study of polymorphism within the MHC is well established, because the region contains the highly polymorphic HLA genes. HLA polymorphism has been used extensively in solid organ and bone marrow transplantation to match donors and recipients. As a result, large numbers of HLA alleles have been identified, a process that has been further driven by recent interest in HLA gene diversity in ethnic populations. The extreme genetic variation in HLA genes is believed to have been driven by the evolutionary response to infectious agents, but relatively few studies have analyzed associations between HLA genetic variation and infectious disease, which has been difficult to demonstrate.
