Book excerpt: The purpose of this study was to sequence the mitochondrial genome of a North American species of scallop and examine it in a phylogenetic context. The mitochondrial DNA of the bay scallop, 'Argopecten irradians', has been completely sequenced. The entire length was determined to 16221 by long and to code for 12 proteins, 22 tRNAs, and 2 rRNAs. The 'atp8 ' gene was noted to be absent from the genome as in other bivalves. An additional 'trnM' was not located in the genome, a feature noted in other sequenced bivalves. The gene order is unique among molluscs with few shared gene boundaries when compared to other molluscs. The genome was found to be AT-rich (56.93%) and within the range previously noted for other sequenced molluscs. After obtaining the sequence of 'Argopecten irradians' a phylogenetic analysis of existing bivalve ribosomal RNA genes was conducted. Numerous trees were produced, most in agreement with established phylogenies. Bivalve phylogenies generated from protein coding and gene order data deviated from established phylogenies. The possible reasons for this and the implication for phylogenetic studies are discussed.

Book excerpt: Divergence and speciation among highland ichthyofauna of the Mississippi River drainage has been the focus of numerous investigations. This thesis explores the modes of differentiation and speciation among some etheostomid darters of the Central Highlands. Hypothesized modes of divergence were constructed based upon changes in North American drainage patterns inferred from geological evidence. AmtDNA molecular phylogeny was constructed from Etheostoma blennioides and members of the Etheostoma variatum species group. This phylogeny was used to test hypothesized modes of divergence among the two groups. Each mode of divergence predicted a particular phylogenic relationship among taxa. The "real" tree was compared to each hypothetical tree in order infer how the taxa diverged. In addition, a nested analysis was used to infer modes of divergence at an intraspecific level. Within the Etheostoma variatum species group it was found that E. blennius did not form a monophyletic clade which proved to be as divergent from each other as they were from other species within the variatum species group. Conversely, E. kanawhae was paraphyletic with respect to E. osbumi, the two species show minimal differences. Among the sub species of E. blennioides, E.b. newmanii contained two distinct clades, one on each side of the Mississippi River. Divergence with the Etheostoma variatum species group was caused by a combination of factors. The southern highland drainages diverged from the northern drainages prior to the Pleistocene. Speciation in the northern highland drainages was brought about by vicariance due to the advance of glacial ice sheets, and the formation of pro glacial lakes, during the Pleistocene. Populations of E. blennioides in the southern highland drainages diverged from northern populations prior to the Pleistocene. Northern highland populations of E. blennioides diverged east and west of the Mississippi River. Changes in the character of the Mississippi during the Pleistocene made it impassible to highland fauna such as E. blennioides. Ecological differences between these two groups of etheostomid darters caused them to respond to the same environmental factors in qualitatively different ways.
