Book excerpt: A carbonic anhydrase-based metal ion biosensor was optimized for measuring copper in wastewater and seawater using molecular biology methods. CA variants were prepared that enhanced the copper ion specificity and/or decreased the copper affinity were incorporated into the sensor. We have also incorporated two additional, commercially available fluorescent labels (Alexa Fluor 660 and Oregon Green) into the transducer by covalently labeling a CAII mutant containing a single cysteine residue. Finally, we have used made fusions with fluorescent proteins (dsRed-CA) as a new method of transducing metal ion concentration. These methods will be useful in the development of sensors to determine the concentration of metal ions in wastewater and the ocean.

Book excerpt: This report describes exploratory development of the fiber optic metal ion biosensor initially developed under ONR contract NOOOl4-9l-J-l572. The anticipated application is to monitor wastewater outflows to verify the success of treatment procedures for removing Cu(II); of special interest are wastewater outflows from drydocks resulting from hull blasting and washing operations, which contain ppm levels of Cu and Zn prior to treatment. Issues in this phase of development include the response speed and reversibility of the sensor, transducer stability, and the presence of interferents in treated wastewater. We found that the sensor responded in less than one minute to part per trillion levels of free Cu(II), that it was reversible at the expected rate, that treated wastewater could be measured, and that the apoprotein was storable for months.

The fate of such emerging and geogenic contaminants in subsurface systems, especially in the groundwater, depends on several factors. Physicochemical properties of contaminants such as octanol-water partition coefficient, dissociation constant, water solubility, susceptibility to biodegradation under anaerobic conditions, and environmental persistence under diverse geological and pH conditions play a critical role during subsurface mass flow. Thus, advanced wastewater treatment techniques, followed by implementing stricter guidelines, are some of the measures that can safeguard water resources. This book, in general, gives an understanding of the fate and mitigation strategies for emerging and geogenic contaminants in the groundwater. The first and second sections provide a detailed insight into various removal techniques and mitigation approaches. Book excerpt: E. coli, genetically engineered with a mercury(II)-sensitive promoter and the lux genes from Vibrio fischeri, were used as microbial sensors for the detection of mercury. Evaluation of this genetic construction was carried out by determining the effects of various parameters on cell suspensions maintained at constant conditions in a small vessel. The strongest fight intensities and quickest induction times occurred with cells in the mid-exponential growth phase maintained at 280C, concentrated to 1x10 to the 9th power cells/mL, mixed at very fast speeds, and aerated at 2 vvm (volume of air per volume of culture per minute) during light measurement in the small vessel. The sensitivity of these cells to the mercuric ion lied in the range of 0.02-4 micrometer (4-800 ppb) and the total response time was on the order of one hour, depending on the above parameters. The cells exhibited great specificity for mercury. The cells have almost equal specificity for organic and inorganic form of the. mercuric ion and responded more weakly to the mercurous ion. A simple, inexpensive, durable miniature probe was constructed and operated using the optimum parameters found in the small vessel as a guide. Although protection of water resources is the best long-term solution, we will also need innovative novel approaches and technologies to water treatment to ensure an adequate superior quality resource to meet these needs. Solving tomorrow’s water issues will require unique approaches that incorporate emerging new technologies. Great advances have been made in the area of nanotechnology. Due to their unique physical and chemical properties, nanomaterials are extensively used in antibacterial medical products, membrane filters, electronics, catalysts, and biosensors. Nanoparticles can have distinctly different properties from their bulk counterparts, creating the opportunity for new materials with a diversity of applications. Recent developments related to water treatment include the potential use of carbon nanotubes, nanocompositae, nanospheres, nanofibers, and nanowires for the removal of a diversity of chemical pollutants. By exploiting the assets and structure of these new materials, such as increased surface area, high reactivity, and photocatalytic action, it will be possible to create technologies that can be very efficient at removing and degrading environmental pollutants. Understanding and using these unique properties should lead to innovative, cost-effective applications for addressing the complexities of emerging needs for water treatment and protection. Although still in the early stages, research into the application of nanotechnology shows great promise for solving some of these major global water issues. This comprehensive text describes the latest research and application methods in this rapidly advancing field.
