We have approached the problem in the following fashion: first, by dividing the oncology literature into specific subdivisions such as lung can cer, genitourinary cancer, pediatric oncology, etc.; second, by asking emi nent authorities in each of these areas to edit a volume on the specific topic on an annual or biannual basis. Each topic and tumor type is covered in a volume appearing frequently and predictably, discussing current diagnosis, staging, markers, all forms of treatment modalities, basic biology, and more.

Single tumor cells have been identified in normal tissue such as sentinel lymph nodes, as well as in bone marrow, peripheral blood, and other bodily fluids and cells, by molecular technologies. The introduction of polymerase chain reaction-based methods can be regarded as a prototype of this dramatic development towards molecular approaches in new diagnostic procedures. This fact opens up the possibility of clinical use in patients and of influencing treatment strategies. Considerable discrepancies have been described, how ever, in the success rates of these new techniques for the detec tion of minimal residual disease in cancer patients. The study of cancer will be profoundly impacted by the Human Genome Project. I would like to discuss some of these changes. The first draft of the human genome sequence was announced in June 2000, and we have just scratched the surface of the changes it will engender in medicine. A relevant question is what are the long-term effects of the Human Genome Project for medicine? I would argue that there are three, and each of these three point toward the view that systems biology will dominate biology and medicine of the 21st century. First, the Human Genome Project introduced a new type of s- ence—discovery science. Discovery science takes a biological system (e. g. , the genome) and defines all of its elements (e. g. , the sequences of the 24 human ch- mosomes). Thus, it creates a rich infrastructure from which the classical hypo- esis-driven science can be done more effectively. The effective integration of discovery- and hypothesis-driven science is a key for systems approaches to bi- ogy and medicine. Second, the Human Genome Project has provided a “periodic table of life.

Therapeutic proteins or cells may inhibit melanoma cell growth either by augmentation of immune cell function or by inhibition of angiogenesis. Cytokines and melanoma antigens may be used either in vivo as a vaccine to stimulate irnrnune immune cell cell function or ex vivo to stimulate or proliferate cells for infusion. Alternatively, alteration in melanoma cell growth can occur through inhibition of protein signal transduction pathways within melanoma cells or in the endothelial cells constituting the necessary angiogenic support for tumor growth. The great promise of these therapies and their cellular targets constitutes the basis for Melanoma: Biologically Targeted Therapeutics. THE CLINlCAL PROBLEM More than four million people will be diagnosed with melanoma in the first decade of the 21st century. Half of those who will die will be individuals who would otherwise have had a life expectancy of another 25 years or more. These individuals will die of systemic systernic metastases, which are present at the time of primary surgery. Despite use of sunscreens, incidence continues to increase in developed countries worldwide. To reduce mortality, there must continue to be a focus on prevention and earlier detection through public education. Early interventions are always preferable to treatment of disseminated metastatic disease.
