Book excerpt: Obesity results from a prolonged imbalance between energy intake and energy expenditure, as depending on basal metabolic rate, heat production, thermogenic effects of the diet & physical activity. Diet-induced obesity in rodents can be achieved by different regimens & approaches. Diets providing a high fat intake have been established as a "gold standard" to generate obese rodent models and have proven to initiate pathologies similar to those in humans. However, this dietary treatment is far from being standardized and its relevance has been criticized. Hence, varieties of high fat diet regimens have been introduced by providing a choice of several palatable food items of variable composition. Further male rats have been considered in developing all types of obesity models and no rationale has been provided as of this discrimination and negligence of using female rats. Another pitfall in the obesity model development is that of the choice of the strain of the rats used for the study. The basis on which strain is being chosen still remains controversial. This present study aimed at comparing & validating different diet induced obesity models utilizing different strains and gender.

Usefulness of using small rodents in animal models to infer aspects of human obesity is discussed in terms of similarities and differences and how obesity is defined. The paradigm of using fat-rich diets in animal models of human obesity is explored according to its adequacy and dietary characteristics related to fatty acid composition. Physiological factors and mechanisms that can play a role in the development of obesity induced by a diet rich in fat are examined, namely the efficiency of nutrient utilization and the possible lack of inhibitory effect of fat on intake. The role of hormones such as leptin, ghrelin and insulin is discussed. Bevavioural mechanisms related to sensory-specific facilitation of eating with fat rich diets, altered feeding rhythmicity and learned eating are considered. The possible reversal of fat-rich diet induced obesity in animal models is explored. This chapter concludes with comments on appropriate design of animal studies and suggestions for future research.

There is a need to explain how new drugs can counter insulin resistance and insulin deficiency to a broad range of professionals, from clinical scientists active in early (and later) phase drug development to specialist physicians and increasingly primary care doctors who must tailor drug regimens to the individual patient. Clinical research methods for measuring insulin action and insulin secretion have become well-established in proof-of-mechanism studies; however, selection of the best techniques is by no means straightforward.
